Book excerpt: During the beginning of the postpartum period, 80 to 100% of lactating dairy cows have bacterial contamination within their uteri. Presence of uterine bacteria may contribute to increased number of days open and services per conception within these cows. Expulsion of uterine contamination is crucial for uterine involution and reproductive health. Susceptibility to bacterial infection within the uterus can be attributed to a decrease in neutrophil function during the first two weeks of the postpartum period. Prostaglandin F2[alpha] (PGF2[alpha]) has been found to induce uterine contractility, increase neutrophil propagation and affect the uterine environment. The objective of this study was to determine the effects of a single injection of prostaglandin F2[alpha] (Lutalyse, Pfizer Animal Health, New York, NY) on uterine health and reproductive performance in dairy cows. Holstein cows from a commercial dairy were randomly assigned to receive an intramuscular injection of either Lutalyse or saline on Day 7 or 14 postpartum (Day 0= day of parturition). Services per conception and days open were evaluated to determine the effects on reproductive performance due to treatment. Cytobrush and uterine swabs were performed on a subset of Day 14 postpartum cows at 0 and 24 h after injection to evaluate changes in uterine neutrophil and bacterial populations. The pro-inflammatory cytokines known to induce mobilization of neutrophils, interleukin-1[beta], interleukin-6, and tumor necrosis factor-[alpha] were also measured at 0, 3, 6 and 12 h after injection of Lutalyse or saline on Day 14 postpartum to determine if PGF2[alpha] induced an increase in cytokines similar to initial pathogen detection. No significant differences were seen in days open, services per conception, interleukin-1[beta] or tumor necrosis factor-[alpha] concentrations. Interleukin-6 concentration was greater in saline cows at 12 h compared to Lutalyse treated cows at 3, 6 and 12 h after injection. Additionally, an increase in uterine neutrophils (P=0.05) and a decrease in bacteria (P=0.05) in response to Lutalyse were observed. Although PGF2[alpha] has been shown to have beneficial effects on the postpartum uterus, a single injection protocol may simply not be adequate to induce significant improvement in reproductive performance.

Lactating cows (n = 3,217) from 2 dairy herds were screened for retained placenta (RP; >24 hours after parturition), metritis (MET) within 20 days in milk (DIM), and clinical endometritis (CE) at 26 ± 3 DIM. Milk yield and components from the DHIA test-days up to 305 DIM and reproductive performance at first service were collected. Weekly, a list of cows was obtained using on-farm computer records screened for RP, MET, and CE. Parity (lactations 1, 2 and =3) of cows was accounted for milk yield, milk components, and reproduction. The statistical analyses were performed using MIXED (milk yield and components), GLIMMIX (risk factors associated with uterine diseases and reproduction), and CORR (correlation of risk factors with uterine diseases) procedures of SAS. Cows diagnosed with MET or CE had a negative effect on milk yield, increased milk fat percentage and SCC, and reduced reproductive performance at first service. Book excerpt: Dairy cattle have strong behavioral need to rest and deprivation of their lying time (LT) has been associated with lameness and changes in feeding behavior and endocrinological milieu. However, the effects of LT during the transition period on metabolic stress, inflammation, health and performance have not yet been fully investigated. Therefore, the following tree specific aims were conducted. A total of 1052 Holstein cattle (401 nulliparous heifers and 651 parous cows) from 3 commercial dairy farms were enrolled for this prospective cohort study. The first aim (Chapter 3) was to evaluate the association of pre- and postpartum lying time (LT) on calcium and energy balance (NEFA and BHB), inflammation (haptoglobin and hemogram), and health status of dairy cows. Prepartum LT had significant quadratic association on serum NEFA concentration, but was not significantly associated with serum Ca concentration within 48 h after calving. Increased LT within 14 DIM was associated with increased risk of diagnosis with ketosis within 14 DIM, inflammation, and incidence of postpartum diseases. Findings from this study suggest that LT along with Ca and energy balance are critical for transition cow health. The second aim (Chapter 4) was to assess the association of post-partum LT on milk yield, components (milk fat and protein percentage), and somatic cell count (SCC). Milk yield at first DHIA test was not associated with LT within the first 14 DIM, but was negatively correlated with the daily coefficient of variation (CV) of LT within 14 DIM. However, health status was associated with milk yield and components, and health status was associated with LT within 14 DIM. The third aim (Chapter 5) was to evaluate the association of lying time (LT) within the first 14 days in milk (DIM) and health status on culling within 60 DIM, cyclicity, and reproductive performance. LT had a positive linear association with culling, for every hour increment of LT within 14 DIM the culling risk within 60 DIM increased by 1 percentage point, and negative quadratic association with cyclicity at 42 DIM. Multiparous cows with a LT of 9-13 h/d had greater probability of pregnancy up to 300 DIM compared to cows with a LT >13 h/d. These findings suggest that there is an optimum daily LT range for early postpartum cows housed in freestall barns, different than reported for mid lactation cows, with the potential for improved reproductive performance.
