Book excerpt: We use Molecular Dynamics (MD) simulations to investigate how solid substrates interact with a dipalmitoylphosphatidylcholine (DPPC) lipid bilayer and how substrates affect the lipid membrane structurally and dynamically. Supported lipid bilayers are extensively used in experiments as a simplified model for cell membranes. Few simulations of supported bilayers have been performed. We use the MARTINI coarse-grained model to study the supported bilayer computationally. When a lipid bilayer is deposited on the solid substrate, the undulation of the proximal leaflet is suppressed to a higher extent than that of the distal leaflet. Properties of the bilayer including density profiles and lateral mobility exhibit heterogeneity. The proximal leaflet is ordered under the influence of a smooth surface, while the distal leaflet remains the similar to the leaflets in a free-standing bilayer. The lateral pressure profile of the supported bilayer is also different from that of a free-standing bilayer. Membrane proteins undergo undesired conformational changes due to the change in the lateral pressure profile. We study in particular the high local pressure in the proximal leaflet and estimate the interfacial energy associated with it. The interfacial energy falls in the range of activation energy barriers of the large mechanosensative channel (MscL) and may contribute to the opening of MscL without external pressure. Experiencing the strong inwards tension, the proximal leaflet prefers a smaller area per molecule than the distal leaflet and free-standing bilayers. We employ the umbrella sampling technique to calculate the potential of mean force (PMF) of lipid translocation. The free energy difference given by the PMF of various supported bilayer systems indicate that the proximal leaflet needs more lipids than the distal leaflet. The equilibrium density imbalance between two leaflets in the supported bilayer is roughly 4-7%. In addition, the equilibrium area per molecule of the proximal leaflet is estimated to be around 58 Å2, about 4 Å2 smaller than that of the free-standing bilayer. The lipid flip-flop rate in a supported bilayer with the same number of lipids in both leaflets is higher than in the free-standing bilayer, because the energy barrier of flip-flop is lowered by a few kBT.

Due to their complexity, supported lipid bilayers have been used as alternates to living cell membranes for study of membrane dynamics, membrane structure, and lipid-protein interactions, among other topics. The tools of modern genetic engineering and bioorganic chemistry allow us to couple many types of biomolecule to supported membranes. This results in wide variety of interfaces that can be used to control, organize and study the properties and function of membranes and membrane-associated proteins. The overarching goal has been to develop robust and easily fabricated biomimetic membrane platforms to facilitate the study of transmembrane proteins for sensing applications.Despite the large volume of research, development of solid supported membrane bilayers that recreate the proper environment has proven to be difficult. Some fundamental drawbacks have been protein substrate interactions leading to premature denaturing of the protein and minimal-to-no mobility within the membrane. To ameliorate these issues, polymer supported lipid bilayers are widely used. One of the main attractions for using polymeric cushions is to mimic thermodynamic and structural properties of free bilayer more specifically the cytoskeleton and extra cellular matrix, which is an important feature of the native cellular environment.Chapter 1 describes the fabrication and characterization of polymer supported bilayers and reviews the lateral mobility of the lipids and proteins on various types of polymer supports. Also presented are potential practical applications including biosensors and future directions of development of polymer supported lipid bilayers. Chapter 2 provides the mobility of a polymer supported lipid bilayer with a polymer cushion that is pH responsive and structurally tunable in order to provide an environmentally sensitive interface. In details, the simple preparation of poly(acrylic acid) (PAA) cushioned membranes is described. The PAA cushion provides a tunable, water rich environment for supported lipid membrane applications. Conveniently the thickness of the PAA cushion can be controlled by manipulating the pH of the surrounding aqueous solution and the concentration of the spin-coating solution. As demonstrated in this work, the structure and continuity of cushioned membranes remains intact and this approach offers a robust and facile means to fabricate biological membranes for biophysical studies and sensing applications.

Book excerpt: Over the past century, the scientific community has spent a great deal of effort trying to understand the cellular membrane bilayer on an intimate level. Langmuir deposition and vesicle fusion has provided a means to deposit membrane bilayers atop solid inorganic supports. These simple membranes remain fluid in nature due to a 10-30 Å water cushion that rests between the membrane and the solid substrate. Although these mimetic templates are ideal for simple lipid membrane, placement of membrane proteins into these systems results in loss of membrane fluidity, and the proteins often become denatured. It has been suggested that portions of the embedded protein extend beyond the vertical length of the membrane and water cushion, and negatively interact with the inorganic support. Due to these substrate-protein interactions, polymeric cushions and tethers have been used to adjust the vertical distance between the membrane and solid support. Poly(ethylene glycol) (PEG) amphiphiles were one of the first polymers used to remedy the substrate-protein interactions because of its hydrophilic and bioinert qualities. It was found that membranes tethered with PEG amphiphiles actually have up to three different motilities within a single system. There has been much speculation as to why these systems contain different motilities, but no real studies have been conducted to assess the issue. One focus of this dissertation was to study PEG-amphiphile/lipid interactions at the air-water interface. During the process of characterizing these binary mixtures, an interesting meta-stable domain changing phenomena was found to occur at low surface pressures within a narrow concentration regime in these mixtures. These serendipitous results will be described. Due to the different mobile fractions within a single PEG cushioned/tethered membrane system, other polymers have been explored. In this work the development of pH responsive, hydrophilic, and cross-linked poly (acrylic acid) (PAA) based cushioned membrane bilayers are also depicted. The cellular membranes own cushion inspired the design of these cushions; the cytoskeleton, which is a netted network that is able to change shape, based on environmental changes. In addition Gramicidin A transmembrane protein was embedded into PAA cushioned membrane bilayers.

Biomimetic membranes would make a significant contribution to alleviate water stress, environmental threats, and energy consumption.
