Book excerpt: In the United States, the critical task of preserving our plant genetic resources is the responsibility of the National Plant Germplasm System (NPGS). NPGS undergoes a thorough analysis in this book, which offers wide-ranging recommendations for equipping the agency to better meet U.S. needsâ€"and lead international conservation efforts. The book outlines the importance and status of plant genetic conservation and evaluates NPGS's multifaceted operations. Two options for revamping NPGS within the U.S. Department of Agriculture are included.

Available in PDF, EPUB and Kindle. Book excerpt: Ex situ preservation of germplasm for higher plant species has been accom plished using either seeds or clones, but storage of these under typical condi tions does not provide the extreme longevities that are needed to minimize risk of loss. Costs of maintenance and regeneration of stocks are also high. Systems that provide virtually indefinite storage should supplement existing methods and it is within this context that cryopreservation is presented. The use of low temperature preservation was initially more a concern of medicine and animal breeding, and was expanded to plants in the 1970s. Sur vival after cryogenic exposure has now been demonstrated for diverse plant groups including algae, bryophytes, fungi and higher plants. If survival is com monplace, then the eventual application is a cryopreservation system, whereby cells, tissues and organs are held indefinitely for use, often in the unforeseen future. The increasing interest and capabilities for application could not have occurred at a more opportune time since expanding human populations have placed unprecedented pressures on plant diversity. This book emphasizes cry opreservation of higher plants and was initially driven by the concern for loss of diversity in crops and the recognized need that this diversity would be essential for continued improvement of the many plants used by society for food, health and shelter. The interest in cryopreservation has been expanded by conservationists and their concerns for retaining, as much as possible, the diversity of natural populations. The need for cryopreservation, thus, is well established.

Book excerpt: This publication provides an approach by which conservationists can determine whether or not long-term seed storage is feasible for a particular species, i.e. whether or not that species shows orthodox seed storage behaviour, and provides advice on the implementation of the protocol, examples of ways in which the results from seed storage studies could be misinterpreted due to confounding factors, as well as several alternative approaches for estimating seed storage behaviour prior to carryng out actual investigations with the seeds. In particular, the latter section introduces the concept of a multicriteria approach for estimating seed storage behaviour.

Book excerpt: Cryopreservation has proven to be an important tool for the storage and conservation of plant genetic resources. This book is a unique resource for plant scientists, providing more than 100 ready-to-use cryopreservation protocols for plant types from algae and bryophytes to a range of flowering plants. It includes techniques for diverse plant parts such as dormant buds, pollen, and apical meristems and for cell types such as suspension and callus cultures.

Book excerpt: Topics for the Beltsville Symposia are selected to highlight specific areas of research and science policy that are of concern to scientists in the Beltsville Area as well as to the general scientific community. Each sympo sium in the series is structured to provide a realistic appraisal of current findings, research progress, and relevant policy issues within the constraints established by the organizing committee. Thus, the presentations and dis cussions that have marked these symposia have had a strong appeal to the broad community of scientists. Knowledge of the diversity of living organisms is still quite limited. Since the time of Linnaeus, about 1.7 million species have been described. The actual number has been estimated between 5-50 million. Many species, land races, and strains are vanishing. Clearly, the world's scientific institutions are inadequately equipped to attain sufficient knowledge of a significant fraction of the diverse living forms. Also, efforts in the collection and preservation of germplasm of plants and animals urgently need to be strengthened. These mattes are critically important to future generations. This symposium addresses vital concerns of biotic diversity and germ plasm preservation from diverse perspectives. Many of the parts provide concrete recommendations for action, and they call attention to areas of research that must be pursued with intensity.

Book excerpt: The long-term storage and maintenance of viable plant cells and organs is an area of active concern across the range of pure and applied plant sciences. In academic, government and commercial laboratories, the extended storage of propagules of one sort or another, with maximum protection of the genome from mutation and altered expression, is often a very necessary activity that can draw heavily on resources and effort. However, preservation per se is typically not an activity in its own right, but a facilitating technology that is part of a larger programme of work. Consequently, there are many laboratories that do not have the benefit of a specialist in storage technology, and have to delegate the responsibility to individuals, or teams, who are faced with a daunting learning curve. To maximise the chances of success, in the shortest possible time and with minimum losses, these researchers need sources of reference that are au thoritative and soundly based in practical experience.

Available in PDF, EPUB and Kindle. Book excerpt: A number of interdisciplinary fields related to Plant Cell Biotechnology are discussed. The two main directions are: Plant cell culture in agricultural applications for the improvement of crops and industrial applications in the production of secondary metabolites. A number of areas such as physiological and biochemical aspects of autotrophic cells, gene characterization in higher plants, transformation of plant cells, genetic stability in plant cell cultures, somatic hybridization and somatic embryogenesis are treated. Recent knowledge on somaclonal and gametoclonal variation as well as on the obtainment of protoplasts and their use for the isolation and culture of heterocaryons as tools for plant breeding are considered. Furthermore, the knowledge on biomass production in fermentor conditions and the role of immobilization for increased production and scale-up of plant cells are discussed.
