The selection first offers information on gas chromatography, electrochromatography, and electroanalytical methods in trace analysis. Discussions focus on analytical applications, apparatus and techniques, titration methods, anodic stripping of deposited metals, and polarography. The book then examines the high-frequency method of chemical analysis, field emission microscopy, and theory and principles of sampling for chemical analysis. The publication takes a look at flame photometry and microwave spectroscopy. Topics include sample treatment required for flame photometric determinations; factors affecting precision and accuracy in flame photometry; theoretical background of microwave spectroscopy, and problems connected with quantitative analysis. The manuscript then elaborates on analytical applications of nuclear magnetic resonance; fluorescent x-ray spectrometric analysis; and neutron spectroscopy and neutron interactions in chemical analysis. The selection is a dependable reference for readers interested in the application of physical methods in chemical analysis.

Book excerpt: Physical Techniques in Biological Research, Second Edition, Volume I, Part A: Optical Techniques focuses on the methods and theory applied to relatively pure preparations of biological substances that are derived from cells or other tissue elements. This book covers a wide variety of nonoptical techniques. Organized into five chapters, this edition begins with an overview of the fundamental principles of operation of optical instruments that can be explained in terms of the simple operations of lenses or light rays. This text then examines the limits imposed on direct observation, either through an optical instrument or with the unaided eye, by the nature of light itself. Other chapters consider the potential of certain other microprobe methods. This book discusses as well the process of molecular spectroscopy. The final chapter deals with optical rotation and its dependence on wavelength. This book is a valuable resource for biologists, scientists, physicists, physical chemists, and research workers.

Book excerpt: Microparticle Characterization via Confocal Microscopy Physical Methods in Chemistry and Nano Science Confocal microscopy was invented by Marvin Minsky in 1957, and subsequently patented in 1961. Minsky was trying to study neural networks to understand how brains learn, and needed a way to image these connections in their natural state (in three dimensions). He invented the confocal microscope in 1955, but its utility was not fully realized until technology could catch up. In 1973 Egger published the first recognizable cells, and the first commercial microscopes were produced in 1987. Chapter Outline: A brief history of confocal microscopy The basics of fluorescence How confocal microscopy is different from optical microscopy Parts of a confocal microscope Two-photon microscopy Microparticle characterization A summary of confocal microscopy's strengths and weaknesses The Open Courses Library introduces you to the best Open Source Courses.

Book excerpt: Many people look upon a microscope as a mere instrument(l); to them microscopy is instrumentation. Other people consider a microscope to be simply an aid to the eye; to them microscopy is primarily an expan sion of macroscopy. In actuality, microscopy is both objective and sub jective; it is seeing through an instrument by means of the eye, and more importantly, the brain. The function of the brain is to interpret the eye's image in terms of the object's structure. Thought and experience are required to distinguish structure from artifact. It is said that Galileo (1564-1642) had his associates first look through his telescope microscope at very familiar objects to convince them that the image was a true representation of the object. Then he would have them proceed to hitherto unknown worlds too far or too small to be seen with the un aided eye. Since Galileo's time, light microscopes have been improved so much that performance is now very close to theoretical limits. Electron microscopes have been developed in the last four decades to exhibit thousands of times the resolving power of the light microscope. Through the news media everyone is made aware of the marvelous microscopical accomplishments in imagery. However, little or no hint is given as to what parts of the image are derived from the specimen itself and what parts are from the instrumentation, to say nothing of the changes made during preparation of the specimen.

Book excerpt: Following three printings of the First Edition (1978), the publisher has asked for a Second Edition to bring the contents up to date. In doing so the authors aim to show how the newer microscopies are related to the older types with respect to theoretical resolving power (what you pay for) and resolution (what you get). The book is an introduction to students, technicians, technologists, and scientists in biology, medicine, science, and engineering. It should be useful in academic and industrial research, consulting, and forensics; how ever, the book is not intended to be encyclopedic. The authors are greatly indebted to the College of Textiles of North Carolina State University at Raleigh for support from the administration there for typing, word processing, stationery, mailing, drafting diagrams, and general assistance. We personally thank Joann Fish for word process ing, Teresa M. Langley and Grace Parnell for typing services, Mark Bowen for drawing graphs and diagrams, Chuck Gardner for photographic ser vices, Deepak Bhattavahalli for his work with the proofs, and all the other people who have given us their assistance. The authors wish to acknowledge the many valuable suggestions given by Eugene G. Rochow and the significant editorial contributions made by Elizabeth Cook Rochow.

Book excerpt: This is the second of three volumes of Methods in Molecular Biology that deal with Physical Methods of Analysis. The first of these, Spectroscopic Methods and Analyses dealt with NMR spec troscopy, mass spectrometry, and metalloprotein techniques, and the third will cover X-ray crystallographic methods. As with the first volume. Microscopy, Optical Spectroscopy, and Macroscopic Techniques is intended to provide a basic understand ing for the biochemist or biologist who needs to collaborate with spe cialists in applying the techniques of modern physical chemistry to biological macromolecules. The methods treated in this book fall into four groups. Part One covers microscopy, which aims to visualize individual molecules or complexes of several molecules. Electron microscopy is the more familiar of these, while scanning tunneling microscopy is a new and rapidly developing tool. Methods for determining the shapes and sizes of molecules in solution are described in Part Two, which includes chapters on X-ray and neutron scattering, light scattering, and ult- centrifugation. Calorimetry, described in Part Three, provides the means to monitor processes involving thermodynamic changes, whether these are intramolecular, such as conformational transition, or the interactions between solutes or between a solute and its sol vent. Part Four is concerned with optical and infrared spectroscopy and describes applications ranging from the measurement of protein concentration by UV absorbance to the analysis of secondary struc ture using circular dichroism and Fourier-transform infrared spec troscopy.
