Book excerpt: NMR is a technique that can spoil you. It is true that it is not as sensitive as some of the spectroscopic methods, but the quality of the information and the incredible ease of sample preparation make it one of the tools that have opened new vistas in biological research. This volume is intended to show how those vistas can range from the molecule to the microbe to man himself. Further, it is what prompts the rather broad title.

Available in PDF, EPUB and Kindle. Book excerpt: Phosphorous-31 NMR: Principles and Applications focuses on the evolution of phosphorus-31 nuclear magnetic resonance (NMR) as a widely applied spectroscopic probe of the structure and dynamics of phosphorus-containing compounds. The selection first offers information on the principles and empirical observations of phosphorus-31 chemical shifts and the principles and applications of phosphorus-31 spin-spin coupling constants. Discussions focus on introduction and basic principles, directly bonded phosphorus coupling constants, and two-bond and three-bond coupling constants. The text then ponders on phosphorus-31 NMR of enzyme complexes and paramagnetic probes of enzyme complexes with phosphorus-containing compounds. The manuscript elaborates on the use of chiral thiophosphates and the stereochemistry of enzymatic phosphoryl transfer and DNA and RNA conformations. Topics include helix-coil transitions, sequence dependence of double-stranded DNA conformations, dynamic behavior of RNA and DNA, biological and genetic significance, and stereochemical problems studied with chiral thiophosphates. The conformation and dynamics of nucleic acids and phosphoproteins, relaxation behavior of nucleic acids, and theory and applications of solid-state phosphorus-31 NMR to nucleic acids are also discussed. The selection is a valuable reference for readers interested in the principles and applications of phosphorus-31 nuclear magnetic resonance.

Available in PDF, EPUB and Kindle. Book excerpt: Nuclear Magnetic Resonance spectroscopy is a dynamic way for scientists of all kinds to investigate the physical, chemical, and biological properties of matter. Its many applications make it a versatile tool previously subject to monolithic treatment in reference-style texts. Based on a course taught for over ten years at Brandeis University, this is the first textbook on NMR spectroscopy for a one-semester course or self-instruction. In keeping with the authors' efforts to make it a useful textbook, they have included problems at the end of each chapter. The book not only covers the latest developments in the field, such as GOESY (Gradient Enhanced Overhauser Spectroscopy) and multidimensional NMR, but includes practical examples using real spectra and associated problem sets. Assuming the reader has a background of chemistry, physics and calculus, this textbook will be ideal for graduate students in chemistry and biochemistry, as well as biology, physics, and biophysics. NMR for Physical and Biological Scientists will also be useful to medical schools, research facilities, and the many chemical, pharmaceutical, and biotech firms that offer in-house instruction on NMR spectroscopy.

Available in PDF, EPUB and Kindle. Book excerpt: Nuclear Magnetic Resonance is a powerful tool, especially for the identification of 1 13 hitherto unknown organic compounds. H- and C-NMR spectroscopy is known and applied by virtually every synthetically working Organic Chemist. Con- quently, the factors governing the differences in chemical shift values, based on chemical environment, bonding, temperature, solvent, pH, etc. , are well understood, and specialty methods developed for almost every conceivable structural challenge. Proton and carbon NMR spectroscopy is part of most bachelors degree courses, with advanced methods integrated into masters degree and other graduate courses. In view of this universal knowledge about proton and carbon NMR spectr- copy within the chemical community, it is remarkable that heteronuclear NMR is still looked upon as something of a curiosity. Admittedly, most organic compounds contain only nitrogen, oxygen, and sulfur atoms, as well as the obligatory hydrogen and carbon atoms, elements that have an unfavourable isotope distribution when it comes to NMR spectroscopy. Each of these three elements has a dominant isotope: 14 16 32 16 32 N (99. 63% natural abundance), O (99. 76%), and S (95. 02%), with O, S, and 34 14 S (4. 21%) NMR silent. N has a nuclear moment I = 1 and a sizeable quadrupolar moment that makes the NMR signals usually very broad and dif cult to analyse.

Book excerpt: Over the years since NMR was first applied to solve problems in structural biology, it has undergonedramaticdevelopmentsinbothNMRinstrumenthardwareandmethodology. While it is established that NMR is one of the most powerful tools for understanding biological p- cesses at the atomic level, it has become increasingly difficult for authors and instructors to make valid decisions concerning the content and level for a graduate course of NMR in str- turalbiology. BecausemanyofthedetailsinpracticalNMRarenotdocumentedsystematically, students entering the field have to learn the experiments and methods through communication with other experienced students or experts. Often such a learning process is incomplete and unsystematic. This book is meant to be not only a textbook, but also a handbook for those who routinely use NMR to study various biological systems. Thus, the book is organized with experimentalists in mind, whether they are instructors or students. For those who have a little or no background in NMR structural biology, it is hoped that this book will provide sufficient perspective and insight. Those who are already experienced in NMR research may find new information or different methods that are useful to their research. Because understanding fundamental principles and concepts of NMR spectroscopy is essential for the application of NMR methods to research projects, the book begins with an introduction to basic NMR principles. While detailed mathematics and quantum mechanics dealing with NMR theory have been addressed in several well-known NMR books, Chapter 1 illustrates some of the fundamental principles and concepts of NMR spectroscopy in a more descriptiveandstraightforwardmanner.

This book discusses the development of analytical tools for the study of the chemistry of phosphorus, which promoted research in nucleic acid chemistry. Organized into 22 chapters, this book begins with an overview of the importance of the bacterial cell wall in maintaining the integrity of the cell in various environments. This text then examines the chemical problems concerning hypermodification and deprotection steps. Other chapters consider the reactive phosphorylating intermediates used in the oligonucleotide chemistry. This book discusses as well the possible role of phosphodiester triazolides and tetrazolides in the phosphotriester formation with arenesulfonyltriazolides and tetrazolides. The final chapter deals with the isolation of proteins involved in the synthesis and recognition of mRNA caps. This book is a valuable resource for phosphorus chemists, biologists, scientists, research workers, teachers, and students.

The application of magnetic resonance methods to the study of actual biological systems as contrasted to cell-free samples, although not entirely novel, as demonstrated by Civan and Shporer in Volume I, has taken on new dimensions with the use of phosphorus-31 and carbon-13 NMR in studying cells, tissues, and organelles. The applications of 31 P NMR to such systems is reviewed in this volume, while carbon-13 will be covered in a later one. The use of nitroxide spin labels has grown to the point where it now may be considered a common biological technique. The synthesis and applications of a new class of nitroxides is described in this volume. ESR spectroscopy of paramagnetic ions is a powerful approach to studying molecular and structural details, as the chapter by Boas, Pilbrow, and Smith on the ESR of copper in Volume 1 has shown. In this volume the ESR of molybdenum and iron is treated in a comparable fashion. In the first volume some aspects of 1 H NMR spectroscopy of certain classes of In this volume the high-resolu biological macromolecules were discussed.· tion multinuclear NMR spectra of peptides, including the physiologically significant peptide hormones, are reviewed.
