It is a fundamental (and probably “primitive”) cell biological process which is important in single celled organisms such as amoeba; multicellular animals including coelenterates; and in higher animals. In humans and other mammals, specialised immune cells (phagocytes) utilise phagocytosis in their crucial role of engulfing and destroying infecting microbes. Yet, surprisingly, the biophysics and biochemistry underlying the process has only become clear recently with the advent of genetic manipulation and advances in single cell imaging. In this volume, the aim is to bring together recent fundamental advances that give a clear picture of the underlying mechanism involved in phagocytosis. Not only is this an important topic in its own right, but a full understanding of the process will have a potential impact on human medicine, since as antibiotics become less effective in fight infection, researchers are looking at alternative approaches, including enhancing the “natural” immunity brought about by immune phagocytes. The aim is to provide a comprehensive volume on the topic, with separate chapters on identified recent advances, each written by the major contributors in each area. In addition, the volume will attempt to give a wider overview than is often the case in single author reviews, with an emphasis here on the cell biological understanding of phagocytosis using biophysical approaches alongside the biochemical and imaging approaches.

Global prevalence of human brucellosis remains significant. More than half a million new brucellosis cases from 100 countries are reported annually to the World Health Organization (WHO). The majority of these cases are reported in developing countries.In humans, brucellosis (undulant fever, Malta fever) is characterized by an acute bacteremic phase followed by a chronic stage that may extend over many years and may involve many tissues. It is a systemic disease, and many organ systems (nervous system, heart, skeletal system, bone marrow, etc.) may become involved following hematogenous dissemination. Although eradicated in some countries, it remains one of the most economically important zoonosis worldwide as it is responsible for huge economic losses as well as significant human morbidity in endemic areas. Because of the nonspecific clinical manifestations of human brucellosis and the need for prolonged combination therapy with antibiotics that are not routinely prescribed for other infectious diseases, laboratory confirmation of the diagnosis is of paramount importance for adequate patient management. In addition, evidence of brucellosis has serious public health implications because it discloses exposure to a contaminated source (infected animals or their products, unsafe laboratory practices, or a potential biological warfare attack). This book addresses human brucellosis with stress on symptoms including those related to the less recognized disease localizations, risk of exposure, treatment, and prevention. Light is shed on animal brucellosis as it pertains to human exposure. The book also emphasizes on laboratory procedures in culturing and serologic techniques. That first meeting led to new nomenclature, i. e. , the Mononuclear Phagocytes System (MPS), which was published in the Bulletin of the World Health Organization (R. van Furth, et aI. , Bull. WHO, 1972, 46: 845-852). The paradigm of the MPS has the advantage of being much more consistent than the Reticuloendothelial System (RES) (L. Aschoff, Ergebn. Inn. Med. Kinderheilk. 1924, 26:1), because cells with similar characteristics can be classified in the same category. The essence ota new concept is, according to Popper, that it must be susceptible to falsification. This has been attempted several times for the MPS by those who adhere to the RES. However, in biology no one can be certain of truth. Any theory must be based on experimental observations and/or theoretical consider ations and both should be presented in such a way that others can try to refute it, then or later. One may have a theoretical or a pragmatic preference for a theory which at a given time is regarded as possibly truc, but later it may be shown to be false. In other words, what we present in this volume is regarded as true at this moment, but others may consider it as not true and try to disprove it. Controversies often stimulate new research.
