Book excerpt: "Many birth defects arise as a result of improper patterning of neural crest cells (NCCs), the most rostral of which migrate ventrally from the neural folds into the pharyngeal arches, transient structures on the ventral embryo. This thesis examines the instructive signals and transcription factor networks that establish dorsoventral identity of NCCs and thus eventual formation of the craniofacial skeleton. One such signal is endothelin-1 (Edn1), which is produced by the surrounding arch ectoderm. Produced as an immature form, Edn1 undergoes two processing events, the second mediated by endothelin converting enzyme (Ece1). Mature Edn1 then binds to the endothelin-A receptor (Ednra) on NCCs. Loss of Ece1 leads to a lack of mature Edn1 and thus subsequent defects in dorsoventral patterning. However little is known about the factors that regulate Ece1. We show here that Nkx2.5 is required for proper craniofacial development and acts by regulating ece1 expression ..."--Abstract.

These derivatives range from the mesenchymal bone and cartilage comprising the facial skeleton, to neuronal derivatives of the peripheral sensory and autonomic nervous systems, to melanocytes throughout the body, and to smooth muscle of the great arteries of the heart. For these cells to correctly progress from an unspecifi ed, nonmigratory population to a wide array of dynamic, differentiated cell types-some of which retain stem cell characteristics presumably to replenish these derivatives-requires a complex network of molecular switches to control the gene programs giving these cells their defi ning structural, enzymatic, migratory, and signaling capacities. This review will bring together current knowledge of neural crest-specifi c transcription factors governing these progressions throughout the course of development. A more thorough understanding of the mechanisms of transcriptional control in differentiation will aid in strategies designed to push undifferentiated cells toward a particular lineage, and unraveling these processes will help toward reprogramming cells from a differentiated to a more naive state. Such studies are a large, and it seems ever-expanding, part of most life-science groups. It is at newcomers to this field that this book is primarily aimed. A number of different plants and animals serve as common model org- isms for developmental studies. In Molecular Methods in Developmental Bi- ogy: Xenopus and Zebrafish, a range of the molecular methods applicable to two of these organisms are described, these are the South African clawed frog, Xenopus laevis, and the zebrafish, Brachydanio rerio. The embryos of both of these species develop rapidly and externally, making them particularly suited to investigations of early vertebrate development. However, both Xenopus and zebrafish have their own advantages and disadvantages. Xenopus have large, robust embryos that can be manipulated surgically with ease, but their pseudotetraploidy and long generation time make them unsuitable candidates for genetics. This disadvantage may soon be overcome by using the diploid Xenopus tropicalis, and early experiments are already underway. The transp- ent embryos of zebrafish render them well-suited for in situ hybridization and immunohistochemistry, and good for observing mutations in genetic screens.
