Book excerpt: Achiral wide bite angle ligands have been shown to be highly active and to induce excellent chemo- and regioselectivities in many homogeneously catalyzed reactions. However, only a few examples of chiral wide bite angle ligands are known so far. A diphenyl ether backbone was selected to allow maximum synthetic versatility and potential for a modular approach to design and synthesize such chiral diphosphorus ligands. Three synthetic strategies have been explored in this thesis: i) introduction of chiral substituents in the ligand backbone, ii) the use of P-stereogenic donor atoms and iii) the synthesis of chiral mixed-donor ligands bearing chiral auxiliary groups on the phosphorus atoms. Functionalization of the 3,3'-positions of 2,2'-bis(diphenylphosphino)diphenyl ether by carboxylic acid or ether auxiliaries was achieved via straightforward four-step routes to generate a library of ligands that were tested in various catalytic reactions. In the Pd-catalyzed asymmetric allylic alkylation of l,3-diphenyl-2-propenyl acetate and cyclohexyl-2-enyl acetate with dimethyl malonate the enantioselectivity was found to depend on the size of the chiral auxiliary introduced within the diphenyl ether backbone and its proximity to the phosphorus donor groups and hence to the active metal centre. Two types of mixed donor bidentate diphosphorus ligands based on the diphenylether backbone have been established, i.e. phosphine-phosphite and phosphine-phosphonite derivatives. A small ligand library bearing different chiral auxiliaries was accomplished via straightforward syntheses that enable derivatization of the respective phosphite and phosphonite moieties in the final step. In the Rh-catalysed hydrogenation of several benchmark substrates high conversion and moderate to high enantioselectivities (up to 97% for dimethyl itaconate) were obtained. The enantioselectivity was influenced by the size of the ortho-substituent on the chiral auxiliary group of the phosphite or phosphonite fragment. Two modular synthetic approaches for the preparation of novel wide bite angle diphosphine ligands containing stereogenic P-atoms have been developed. Both protocols involved diphenylether as backbone and the chiral ephedrine based precursor (2R[subscript(P)],4S[subscript(C)],5R[subscript(C)])-oxazaphospholidine borane as initial auxiliary to induce chirality at phosphorus. Various novel diphosphines were isolated as highly enantioenriched compounds with dr-ratios up to 95:5.

With this knowledge in hand it may be possible to rationally design next generation ligands and catalysts to affect improved substrate transformations, with higher selectivities and faster reaction times. The success of Burk's DuPhos ligands in asymmetric hydrogenation demonstrated that chiral phosphacycles can be a potent source of chiral induction, whilst in a similar vein the work of van Leeuwen and Kamer established the wide bite angle xanthene based ligands as excellent catalysts in a range of reactions including hydroformylation. In a preliminary study with Osborne they showed that combining these wide bite angle ligand backbones with Burk's phospholane moieties led to a new powerful ligand in asymmetric allylic substitution. To examine the potential of combining these two ligand features further we designed and synthesised nine new C2-symmetric bidentate wide bite angle bisphosphacyclic ligands, featuring phosphetane, phospholane or diazaphospholane rings, aiming at a wide diversity of steric and electronic properties. The application of these ligands as chiral auxiliaries in transition metal catalysed reactions, including; hydrogenation, hydrocyanation, hydroformylation and allylic alkylation has been investigated. Good to excellent enantioselectivities were observed in all reactions, with maximum ee's of 92.5% observed in hydrogenation, using N-(3,4-dihydro-1-napthalenyl)-acetamide as substrate, and of 96.2% in the alkylation of 1,3-diphenyl-2-propenyl acetate.

Book excerpt: A series of N-salicyl-beta-aminoalcohol ligands had been synthesized by three component Mannich type reaction followed by ring opening of oxazolidine derivatives with hydroxylamine hydrochloride. The reactions provided a series of chiral N-salicyl-beta-aminoalcohol ligands in high yields (84-92%) without any racemization. These synthesized compounds were evaluated as ligands for catalytic asymmetric Strecker reactions. N-Benzhydrylaldimines derived from aromatic and aliphatic aldehydes reacted withTMSCN in the presence of 10 mol% of Ti-ligand complex to give the alpha-aminonitriles in excellent yields and in up to >98% ee. In addition, the catalyst loading was successfully reduced to 2.5 mol% which is very effective and extremely simple for large scale synthesis. The presence of 2-propanol is essential to ensure good conversion and reaction rate. The absolute configuration of all products derived from the (S)-ligand was confirmed to be S. Racemization of alpha-aminophenylacetonitriles is catalyzed by weak acids such as silica (SiO[subscript 2]) or even methanol. However, the racemization can be suppressed by addition of either a base such as triethylamine (NEt[subscript 3]) or strong acid such as hydrochloric acid (HCI). Importantly, optically active alpha-aminoacetonitriles can be easily converted to arylglycines by complete hydrolysis with minimal racemization. (>80% and >90% ee). A transition state model to explain the enantioselectivity of the reaction is proposed. The present chiral catalysts showed their catalytic ability in not only asymmetric strecker reaction but also asymmetric Pudovic reaction as well as asymmetric Michael addition.
