Book excerpt: This study is part of an inter-laboratory collaborating project led by the Environmental Protection Agency (EPA). The EPA provided water samples collected from a river, effluent of wastewater treatment plant (WWTP) and water treatment plant (WTP) in northeastern United States that uses chlorine disinfection. Non-targeted analysis was utilized to screen for a wide range of chemicals. The samples were prepared by solid-phase extraction (SPE) using Oasis HLB and analyzed by comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC×GC/TOF-MS) in nontargeted analytical manner. By using non-targeted analysis, chemicals not targeted can be screened, while a targeted analysis analyzes known chemicals in a sample leaving unknown chemicals undetected. In this study, true detection of a chemical was determined by meeting the following criteria: it must be found only in the sample, must not be found in any field blank sample, and must have a mass spectral similarity score of 500 (50%) or above using the NIST mass spectral library. Data analysis was done by using the instrument software, ChromaTOF, and its add-in feature, “Statistical Compare”. After the automatic data analysis, manual review followed. In total, 382 unique compounds were identified from all six sample locations collected. After manual review, 135 compounds showed high similarity with the NIST mass spectra, and 247 were deemed as low identified compounds. In the wastewater effluent, a majority of the compounds were ingredients of pharmaceutical and personal care products (PPCPs) and flavoring/food additives. In the treated drinking water, 6 disinfection byproducts (DBPs) were detected and not detected in the feed water to the drinking water facility, but produced after treatment. Possible sources of the trace chemicals were discussed. Of all the chemicals tentatively identified, only two from the treated drinking water sample, tribromoacetic acid and dibromochloromethane are regulated in drinking water. This study confirms previous findings that PPCPs and DBPs continue to be pollutants of source waters. Also, the results indicate that non-target analysis was useful to evaluate the efficiency of wastewater and drinking water processes. Further study requires confirmation of the tentatively identified compound, quantification of the compounds for risk assessment and source investigation. The CD-ROM, an appendix to this thesis, is available for viewing at the Media Center in the SDSU Library.

Book excerpt: To assess the effectiveness of the advanced oxidation process (AOP), treated wastewater samples after reverse osmosis (ROP) and ultraviolet light/hydrogen peroxide (UVP) were analyzed by a non-targeted qualitative approach for the presence of organic compounds. Five sampling events were analyzed that occurred at Orange County Water District (OCWD)’s Advanced Water Purification Facility (AWPF) in Fountain Valley, CA. Tap water samples were also collected from a kitchen faucet at OCWD for two of the sampling events. Triplicates for each sample type were collected. The water samples were prepared by solid-phase extraction (SPE) and analyzed by comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC×GC/TOF-MS). While targeted analysis approaches are used to detect known chemicals, non-targeted approaches provide a broad overview of all those that are detected, and consider unknown chemicals. In this study, true detection of a chemical was determined by meeting two criteria: it must be found in all triplicates and must not be found in any of the field blanks. Tentatively identified chemicals were determined by meeting the first two criteria and also meeting a mass spectral similarity score of 700 or above. On average, 71% of the number of chemicals detected in ROP were reduced after UVP among all sampling events. Fourteen compounds were detected and tentatively identified in UVP and also found in ROP. Majority of the 14 compounds were reduced after UVP by an average of 52.2% of their peak areas. The compounds that were most frequently found in UVP were two halogenated acetonitriles, dichloroacetonitrile and bromochloroacetonitrile, in which other studies have commonly found after AOP. The number of chemicals detected in tap water was significantly higher than the number in UVP. In the tap water sampling events, 4-chlorobenzotrifluoride (CBTF) was detected in high abundance and was further confirmed with an authentic standard. Possible sources of the trace chemicals were discussed. None of the compounds identified in this study is under regulation. This study concludes that AOP was effective to remove trace chemicals in ROP. Further study requires confirmation of the tentatively identified compounds and quantitation of the compounds for risk assessment and source investigation. In the Netherlands, drinking water is primarily prepared from surface water and groundwater. To protect the environment and drinking water sources, it is important to adequately monitor the water quality and identify contaminants of emerging concern (CECs). Current European legislation on water quality assessment dictates the use of targeted chemical analysis to analyze a list of priority substances in water bodies. By only measuring a limited list of compounds, however, environmental risks and adverse effects on water quality may be underestimated. To improve this assessment, effect-based bioassays are applied more and more. Bioassays measure the combined potency of all (known and unknown) bioactive compounds in a sample that affect the same endpoint. Effect-directed analysis (EDA) combines chemical analyses and bioassays into one platform to identify CECs, to prioritize them for future monitoring. Previously, such a platform was developed in the High-Throughput EDA project (2013 – 2017, funded by NWO-STW). In this project, the Division of Bioanalytical Chemistry and the VU Department of Environment & Health of Vrije Universiteit Amsterdam collaborated with Het Waterlaboratorium. The developed platform was implemented at Het Waterlaboratorium for monitoring applications outside an academic context. In the HT-EDA project, bioassays that monitor mutagenic and endocrine disrupting activity were applied for the identification of compounds with EDA. Furthermore, the platform allowed for high-resolution fractionation, simplifying the composition of mixtures in fractions in well plate format. The identification success rate was limited, however, which could be (partly) attributed to a lack of fast identification strategies.

Book excerpt: Disinfection by-products (DBPs) are potentially toxic compounds formed when drinking water is treated with disinfectants, such as chlorine or chloramine. A large proportion of the exposure to DBPs is still unknown and the health risks observed through epidemiological studies cannot be explained by DBPs known today. In this thesis, a part of the unknown DBP fraction is investigated, covering a wide range of non-volatile, chlorine/bromine-containing DBPs. The goals were to investigate how the compositions of these DBPs differ between water treatment plants, how their occurrence changes in the distribution system until reaching consumers and how new treatment techniques can reduce their formation and toxicity. To analyze unknown DBPs, a non-targeted approach adopting ultra-high-resolution mass spectrometry, Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS), was used, where the mass of molecules is measured with such accuracy that the elemental composition of individual DBPs can be calculated. A panel of bioassays was used to assess the combined toxic effects from these DBP mixtures. The results show that the formation of these DBPs to a large extent was specific to each water treatment plant and that local conditions influenced DBP formation, based on e.g., the abundance of organic material with certain chemical structures, bromide and disinfection procedure and agent (chlorine or chloramine). The DBPs were detected in both chlorinated and chloraminated water and in all tap water samples, demonstrating that they are part of human exposure. The number of DBP formulae decreased and the DBP composition changed between drinking water treatment and consumer taps, suggesting that DBP exposure to consumers is not necessarily resembling measurements at the treatment plants. Evaluation of new treatment techniques showed that suspended ion exchange and ozonation have potential to decrease the formation and toxic effects of DBPs and that the removal of organic matter can influence qualitative aspects of DBP formation, such as the proportions of chlorine-containing (less toxic) versus bromine-containing (more toxic) DBPs. Through increased knowledge about the role and relevance of non-volatile DBPs, this work can contribute to future monitoring and actions to reduce the health risks associated with DBPs in chlorinated or chloraminated drinking water. The National Research Council was asked by the U.S. Environmental Protection Agency to review the state of the science and create a far-reaching vision for the future of toxicity testing. The book finds that developing, improving, and validating new laboratory tools based on recent scientific advances could significantly improve our ability to understand the hazards and risks posed by chemicals. This new knowledge would lead to much more informed environmental regulations and dramatically reduce the need for animal testing because the new tests would be based on human cells and cell components. Substantial scientific efforts and resources will be required to leverage these new technologies to realize the vision, but the result will be a more efficient, informative and less costly system for assessing the hazards posed by industrial chemicals and pesticides.

The emerging environmental contaminants can be defined as a wide range of chemicals that have been determined in the environment which may present serious health risks for humans. The occurrence of these contaminants indicate that both household and industrial chemicals have been introduced to water resources, a wide variety of chemicals, such as disinfection byproducts, pharmaceutical and personal care products and so on, have been detected at cetiain levels in either water bodies or treatment plants in worldwide. Although developments in new regulations and detection methods have taken place in the past decades that impact water analysis, there is currently no validated EPA or consensus organization methods for many of the listed emerging environmental contaminants. This body of work developed LC/MS/MS or ICP-MS based techniques for water analysis of several classes of emerging environmental contaminants, including herbicides degradation byproducts; cyanotoxins; N-nitrosamines and heavy metal leaching from plastic bottles. In addition, the developed methods were used to conduct high throughput screening of these emerging contaminants in water samples of various types, and to investigate the removal efficiency of these contaminants by using various oxidants and physical treatment with emphasis on analysis and treatment"--Abstract, leaf iv.
