Book excerpt: The gas nitric oxide (NO) has burst upon neuroscience only recently, and yet it has permeated into almost every avenue of current research. The unique properties of this novel messenger have revolutionized our way ofthinking about neurotransmission. These special properties have also lead neuroscientists to invoke NO to explain many previously unexplained phenomena in neurobiology. Fortunately, the development of numerous pharmacological agents is now allowing thesehypotheses to be tested. This volume will provide a synopsis of what is now known about NO. How and where NO is produced, how it acts at the molecular level to activate the synthesis of cGMP, and the possible targets of cGMP in the nervous system are reviewed. The roles of theNO/cGMP signal transduction pathway in the central and peripheral nervous systems, in glial cells, and in neuropathology are then explored. Together, these reviews will lead to further work explaining the varied functions of NO.Key Features* Describes how and why NO is produced in the nervous system* Examines all that is known of NOs role as a neurotransmitter* Explores cellular actions and physiological roles in the brain and peripheral nervous systemCovers NOs molecular synthesis to its role in neuropathology

Book excerpt: "Nitric Oxide and Free Radicals in Peripheral Neurotransmission" is a welcome addition to the literature and describes current research into the role of nitric oxide in the peripheral nervous system and its associated organs. Topics covered range from general consideration of nitrergic transmission, in its broadest sense, to elaboration of our current understanding of the role of nitric oxide in transmission in individual peripheral organs, including its role as a backup, or alternate as well as chief transmitter. The influence of nitric oxide and related compounds on the more conventional modes of autonomic transmission are also considered. Aimed at students and researchers in the areas of neuroscience and physiology, "Nitric Oxide and Free Radicals in Peripheral Neurotransmission" also covers the emerging role of abnormal nitric oxide function in disease states and, where appropriate, as potential avenues of therapy.

Book excerpt: Nitric oxide has a tantalizing role in health and disease: while many of its wide-ranging effects are well known, there remains much more to explore and to learn about the interactions of this fascinating molecule in physiological and pathophysiological processes. The volume reviews the myriad effects of nitric oxide as a chemical messenger in the central nervous system, peripheral nervous system, immune system and cardiovascular system. Furthermore, it provides a very practical introduction to the procedures and experimental protocols necessary to work with and study nitric oxide and its synthesizing enzyme, nitric oxide synthase, in the laboratory. In this respect the volume is unique, providing as it does a complete single-volume review of the role of nitric oxide in health and disease, and a very practical introduction to the methods and protocols involved in this intriguing and active area of biomedical research.

Available in PDF, EPUB and Kindle. Book excerpt: This volume of the Handbook of Neuroanatomy is concerned with nitric oxide synthase. In this volume different research areas are presented together, which adds up to the first major review volume on the localization of nitric oxide synthase in the nervous system. The subjects range from developmental aspects in vertebrates to a functional neuroanatomy of the nervous system in vertebrates. This presentation gives an impressive look on the overwhelming presence of NOS in animal organism and the significance of NO - cGMP signaling. By clearly stating the limitations of our present knowledge the book is also a stimulant for further research. This book presents for the first time an overview of NOS and NO-cGMP signaling in the retina and urogenital system. In addition the effects if injuries on the expression of NOS are summarized in a number of models, which has not been done before. The frame of the book is a classical neuroanatomic description of the localization of NOS. Several authors give detailed advice to prevent pitfalls which may occur when different methods to locate NOS are used. In addition, several chapters detail the target structures for NO while describing the localization of NOS at the same time. All these points together make this volume very timely, i.e. overviewing a decade of NO research.

Available in PDF, EPUB and Kindle. Book excerpt: This e-book will review special features of the cerebral circulation and how they contribute to the physiology of the brain. It describes structural and functional properties of the cerebral circulation that are unique to the brain, an organ with high metabolic demands and the need for tight water and ion homeostasis. Autoregulation is pronounced in the brain, with myogenic, metabolic and neurogenic mechanisms contributing to maintain relatively constant blood flow during both increases and decreases in pressure. In addition, unlike peripheral organs where the majority of vascular resistance resides in small arteries and arterioles, large extracranial and intracranial arteries contribute significantly to vascular resistance in the brain. The prominent role of large arteries in cerebrovascular resistance helps maintain blood flow and protect downstream vessels during changes in perfusion pressure. The cerebral endothelium is also unique in that its barrier properties are in some way more like epithelium than endothelium in the periphery. The cerebral endothelium, known as the blood-brain barrier, has specialized tight junctions that do not allow ions to pass freely and has very low hydraulic conductivity and transcellular transport. This special configuration modifies Starling's forces in the brain microcirculation such that ions retained in the vascular lumen oppose water movement due to hydrostatic pressure. Tight water regulation is necessary in the brain because it has limited capacity for expansion within the skull. Increased intracranial pressure due to vasogenic edema can cause severe neurologic complications and death.

Book excerpt: Neurochemical transmission accounts for the majority of information transfer both in the central and peripheral nervous system. The initial findings centred around the experimental work of the two Nobel Prize winners Sir Henry H. Dale and Otto Loewi as well as Wilhelm Feldberg. Their historical findings opened the door to further investigations and extended the list of neurotransmitters to many others such as amino acids, peptides, purines and nitric oxide. In the first part, the publication provides fascinating insights into the life of the three scientists. Their personality and scientific approach are presented through autobiographical sketches and personal memories by authors. Various comments and details of the atmosphere in the laboratory complete the picture of the conditions at the time. The second part is dedicated to the history of the substances, such as neurotransmitters, their antagonists and analogues. The stories of these substances are presented to the reader in a succinct way, including many anecdotes and unusual events on the way to their therapeutic application.
