A key aspect of genome complexity can be attributed to the process of alternative splicing of precursor mRNA (or pre-mRNA), during which introns are removed and exons are selectively spliced together. This highly regulated process generates different mature mRNA transcripts from a single gene and is widespread throughout the eukaryotic evolution. The majority of the genes expressed in the mammalian central nervous system undergo extensive alternative splicing. Some genes can generate more than a thousand isoforms resulting in diverse proteoforms that can differ in their function, binding preference, catalytic activity, and localization. Perturbation in alternative splicing has been linked to many neurological disorders. Despite its significance, we still lack a comprehensive understanding of its role in neuronal function. Recent studies suggest that alternative splicing is a regulator of tissue identity and development. This is particularly relevant to the brain where developmental regulation and heterogenous cell population play a crucial role in shaping its function. For a long time, technological limitations in detecting and measuring alternatively spliced transcripts hindered the progress in understanding the biology of alternative splicing. Recent advances in long-read and native mRNA sequencing and robust computational tools allow us to capture and quantify in their full-length form. Public transcriptomics datasets can be utilized for the meta-analysis of alternative splicing landscapes of various experimental conditions and disease samples. In addition, protein isoform detection complements mRNA isoform levels. Given its biological significance and recent advances, the objective of this Research Topic is to assemble current knowledge, views, and challenges in the studies of alternative splicing in brain function.

Cancer metastasis is the main cause of death from cancer, and in many cases is difficult to detect or treat. The process by which tumour cells become metastatic is complex and involves many stages, including detachment of cells from the main tumour mass, degradation of the surrounding extra-cellular matrix, invasion into nearby blood vessels, travel and survival through the circulatory system, attachment to a vessel wall, extra-vasation, degradation of the extra-cellular matrix into a distant tissue/organ, and the development of a novel blood supply. In order to accomplish this process, the cells acquire characteristics which are important for each stage. Recently, a class of genes known as metastasis suppressors' has been the subject of intense investigation. For some metastasis suppressor genes, there is strong evidence from both in vitro and in vivo studies to demonstrate key roles in the metastatic process, for others data is much more limited, and their importance uncertain. In this book, chapters are devoted to providing up-to-date summaries of our understanding of individual metastasis suppressor genes. Each is written by a leading authority in the study of that gene. Topics covered include discussions on how each metastasis suppressor was discovered, the mechanisms underlying their loss of expression in tumours and tumour cell lines, their proposed molecular functions, and the consequences to a tumour cell of the loss of this function. The virus occurs world-wide, and most people become infected with EBV sometime during their lives. In the United States, as many as 95 per cent of adults between 35 and 40 years of age have been infected. Infants become susceptible to EBV as soon as maternal antibody protection (present at birth) disappears. Many children become infected with EBV, and these infections usually cause no symptoms or are indistinguishable from the other mild, brief illnesses of childhood. In the United States and in other developed countries, many persons are not infected with EBV in their childhood years. When infection with EBV occurs during adolescence or young adulthood, it causes infectious mononucleosis 35 per cent to 50 per cent of the time. EBV also establishes a lifelong dormant infection in some cells of the body's immune system. One such advance, the development of large-scale approaches to study gene expression, has transformed the way we view the transcriptome. In recent years, these approaches have been applied to studies of alternative RNA splicing, a process where multiple distinct messenger RNAs can be generated from precursor transcripts to produce extensive transcriptomic diversity from a limited repertoire of genes. Global analyses have not only reinforced models initially based on single gene studies, they have also led to numerous insights into general principles governing the regulation and evolution of alternative splicing. In this thesis, I describe how I have combined both large-scale and focused approaches to study alternative splicing regulation during development and in an evolutionary context. Using microarray profiling and comparative genomics approaches, I describe the first large-scale comparative analysis of alternative splicing patterns between humans and chimpanzees. Next, I describe the discovery of a novel neural-specific RS domain splicing factor and the network of alternative exons it regulates to promote nervous system development in vertebrates. Finally, I describe the profiling of alternative splicing patterns during C. elegans development using splicing microarrays and high-throughput sequencing. In this latter study, I also describe two resources that facilitate the analysis of tissue- or cell type-specific splicing events, and enable the function of isoforms to be assessed in vivo . Collectively, these studies have shed light on how differential regulation of alternative splicing has contributed to the evolution of complexity and diversity in biological systems.

I was tempted to write back in my rebuttal to the reviewer that I didn’t realise it had been lost. Uncharacteristic maturity prevailed and I resisted the temptation, but I was struck by the underlying observation that research on the str- ture and function of tropomyosin has been somewhat invisible, particularly in terms of the cytoskeleton isoforms. So, how can it be that one of the two major components of the actin filament has been so thoroughly overlooked? I suspect that the answer is disappointingly pedestrian. Whereas the biochemistry of the 1980s revealed the potential of tropomyosin isoforms to diversify the function of actin filaments, the subsequent disenchantment with isoform biology in general in the 1990s inhibited growth of this field. With the development of more sophisticated experimental - proaches we are now seeing a growing realisation of the importance of tropomyosin in regulating actin filaments beyond its pivotal role in muscle contraction. The opportunity to edit this book came at a time when we had written several reviews on different aspects of tropomyosin function and I had just finished the background reading for a comprehensive review of tropomyosin biology. I realised that the field was simply beyond the capacity of any one person to do the field j- tice.
