Book excerpt: This past decade has led to many significant advances in the understanding of the function of excitatory amino acids in synaptic transmission. The cloning of the ionotropic and metabotropic glutamate receptor families of receptor proteins has produced new strategies for the pharmacological modulation of glutamate transmission. The engineering of transgenic animals with modified expression of receptor proteins has created new insights into the function, dysfunction and possible pathology causally related to glutamate receptors. Advances in the pharmacology of glutamate receptors has led to clinical research addressing multiple therapeutic applications of drugs that act on excitatory amino acid systems. A number of NMDA receptor anatagonists have now been studied in humans. AMPA/kainate and metabotropic receptor active compounds have left the preclinical realms of research and have moved towards or are in the clinic.

Book excerpt: Glutamate is the major excitatory neurotransmitter in the brain and dysfunction of glutamate transmission is the likely cause of a variety of diseases including neurodegeneration following cerebral ischemia, Huntington's chorea, amyotrophic lateral sclerosis, epilepsy, spasticity, emesis, chronic pain, and schizophrenia. Excitatory amino acid receptor agonists and antagonists are therefore of major interest as potential drugs for central nervous system disorders. Excitatory Amino Acids is the first book entirely dedicated to the results of human testing of modulators of excitatory amino acid neurotransmitters. - Coverage of the field of excitatory amino acids from synaptic function to preclinical and clinical pharmacology - Description of the development of NMDA (Nmethyl-d-aspartate) and non-NMDA antagonists - Reports of potential drugs in early and late clinical stages of development

Download or read book Molecular and Cellular Biology of Neuroprotection in the CNS written by Christian Alzheimer and published by Springer. This book was released on 2012-11-05 with total page 0 pages. Available in PDF, EPUB and Kindle. Book excerpt: Given the very limited capacity of regeneration in the brain, protecting neurons that are on the brink of death is a major challenge for basic and clinical neuroscience, with implications for a broad spectrum of acute and chronic neurological and psychiatric diseases. This book brings together leading experts from neurobiology, neurophysiology, neuropharmacology, neuroimmunology and clinical neuroscience to highlight the most recent milestones in this rapidly evolving field. The book will serve as a reference for both basic neuroscientists and clinicians interested in an authoritative update on the molecular and cellular biology of neuroprotection and its promises for new therapeutic strategies.
Book Synopsis Nutrition and Traumatic Brain Injury by : Institute of Medicine
Download or read book Nutrition and Traumatic Brain Injury written by Institute of Medicine and published by National Academies Press. This book was released on 2011-07-01 with total page 444 pages. Available in PDF, EPUB and Kindle. Book excerpt: Traumatic brain injury (TBI) accounts for up to one-third of combat-related injuries in Iraq and Afghanistan, according to some estimates. TBI is also a major problem among civilians, especially those who engage in certain sports. At the request of the Department of Defense, the IOM examined the potential role of nutrition in the treatment of and resilience against TBI.
Book Synopsis Excitatory Amino Acids and Epilepsy by : Robert Schwarcz
Download or read book Excitatory Amino Acids and Epilepsy written by Robert Schwarcz and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 702 pages. Available in PDF, EPUB and Kindle. Book excerpt: Human epilepsy is a major public health problem affecting approximately 2 persons per 1000. It is particularly frequent in ohildren where convul sions may lead to brain damage and subsequent seizure activity in adulthood. Temporal lobe epilepsy (synonyms include limbic epilepsy. psychomotor epilepsy and complex partial epilepsy) is the most devastating form of epilepsy in the adult population since: a) it is often extremely resistant to currently available anticonvulsant drugs (i.e •• it is more resistant than tonico-clonic or grand mal seizures) and b) it includes loss of consciousness. thereby limiting performance of many normal functions and leaving the individual susceptible to bodily injury. It is also associated with nerve cell loss. in particular in the hippocampus and other structures of the temporal lobes. In order to promote an appropriate therapy it is essential to understand the etiology of seizures and its relationship to brain damage. Basic research on epilepsy also provides a very useful vehicle to learn about the way the brain functions under normal conditions. For instance. much of our present understanding of the mechanisms of action of GABA and benzo diazepines. control of neuronal activity. etc. has been derived from such stUdies.
Book Synopsis Environmental Neurotoxicology by : National Research Council
Download or read book Environmental Neurotoxicology written by National Research Council and published by National Academies Press. This book was released on 1992-02-01 with total page 167 pages. Available in PDF, EPUB and Kindle. Book excerpt: Scientists agree that exposure to toxic agents in the environment can cause neurological and psychiatric illnesses ranging from headaches and depression to syndromes resembling parkinsonism. It can even result in death at high exposure levels. The emergence of subclinical neurotoxicity-the concept that long-term impairments can escape clinical detection-makes the need for risk assessment even more critical. This volume paves the way toward definitive solutions, presenting the current consensus on risk assessment and environmental toxicants and offering specific recommendations. The book covers: The biologic basis of neurotoxicity. Progress in the application of biologic markers. Reviews of a wide range of in vitro and in vivo testing techniques. The use of surveillance and epidemiology to identify neurotoxic hazards that escape premarket screening. Research needs. This volume will be an important resource for policymakers, health specialists, researchers, and students.
