Book excerpt: Nanostructured nanocomposites based on poly(3-hydroxybutyrate) (PHB), poly(hydroxybutyrate-co-hydroxyvalerate) (PHBHV) as a matrix polymer and hydroxyapatite (HA) as a bioceramic was studied for its possible use as a bone implant material. Nanostructures such as compact nanocomposites, 3-dimensional porous structures and electrospun mats were fabricated based on applications requiring load bearing and non load bearing ability. Isothermal crystallization of neat PHB showed large spherulites at higher temperatures. These spherulites developed intra and interspherulitic cracks due to internal stress. Addition of nano HA reduced the spherulitic size because of nucleating efficiency. In compact nanostructures, HA was uniformly dispersed in polymer matrix that resulted in uniform dispersion even upto 30 wt% filler loading. The brittle PHB matrix was transformed into quasi ductile matrix because of uniform dispersion and optimized interparticle distance. Selection of HA nanoneedles and nanospheres as a filler led to increase in both strength and toughness as compare to that of neat PHB. Nanoneedles altered the micromechanical deformation processes thereby enhancing toughness. Also cell addition was performed on the substrates using primary osteoblasts. Cell density on nanocomposites substrates was far better than neat PHB. Immuno histology on the substrates showed the enhanced increase in Collagen Type-I expression. 3-dimensional porous structures were produced using templates consisting of sugar and salt grains of varying grain size. In freeze-drying and particulate leaching processes smaller grain sizes resulted in pores with 46 to 70 mm, whereas grain sizes of more than 500 mm resulted in pores with 250 to 350 mm in diameter. The application of polymer solutions with a concentration of 5% (w/v) resulted in larger pore sizes compared to solutions of 10% (w/v). Also electrospun nanofiber nanocomposites were developed by two different techniques. A random nanofiber of 250-400 nm were spun from composite solution were the both the nanoparticles were uniformly dispersed inside the fiber. It was also possible to orient the nanoparticles in the spinning direction. Additionally, parallel plate collector was used to get highly aligned nanofibers with the same dimensions. In conclusion, this work is focused on the development of complete toolkit for diverse bone tissue engineering applications and concepts used here can also be applied to various biodegradable polymers and nanocomposites.
