Available in PDF, EPUB and Kindle. Book excerpt: Although much is known with respect to blood cell formation and function, many new concepts in the areas of the regulation of hematopoietic stem cell commitment and the subsequent survival, proliferation, and differentiation of progenitors have been elucidated in the last five years. Our understanding of the microenvironment where stem cells reside and commit to distinct blood types (the niche) has grown significantly in recent years. Furthermore, blood cells have been used as the key model system to study microRNA function and the role of microRNAs in the transformation of normal cells into cancer cells. The current volume Molecular Basis of Hematopoiesis, edited by Amittha Wickrema & Barbara Kee, provides the most recent developments in the area in addition to a chapter on the utilization of basic science knowledge for the treatment of blood diseases. Each chapter in this book has been written and edited by faculty in major academic and research institutions around the world, who are pushing the frontiers of research in this important area.

Book excerpt: This book provides a concise update on current understanding of the biology of acute and chronic leukemias and other bone marrow neoplasms, including myelodysplastic and myeloproliferative disorders, and explores new and emerging treatments. There is a particular focus on the molecular abnormalities that are drivers of leukemia and on their detection by modern molecular techniques. Knowledge of the ways in which genomic and metabolic abnormalities in the hematologic neoplasms affect prognosis and treatment decision making is reviewed. Detailed attention is devoted to targeted therapies, including novel drugs, and to potential targets for future drug development. In addition, readers find in-depth discussion of cellular and antibody-based immunotherapies as well as the role of hematopoietic stem cell transplantation in the treatment of leukemias and bone marrow malignancies. The book is of special interest for hematologists, oncologists, and cancer researchers; it is also of value for hematology trainees and medical students.

Available in PDF, EPUB and Kindle. Book excerpt: In the summer of 1988, my developmental biology professor announced to the class that hematopoietic stem cells (HSCs) had finally been purified. Somehow, I never forgot the professor’s words. When I started working in Dr. Irv Weissman’s labo- tory at Stanford as a postdoctoral fellow, I realized that the findings mentioned by the professor were from Weissman’s laboratory and had been published in a 1988 edition of the journal Science. It has been over 20 years since the publication of that seminal paper, and since then tremendous advances in understanding the biology and maturation of HSCs, namely the process of hematopoiesis, which includes lymphocyte development, have been made. These discoveries were made possible in part by advancements in technology. For example, recent availability of user friendly fluorescence activated cell sorting (FACS) machines and monoclonal an- bodies with a variety of fluorescent labels has allowed more scientists to sort and analyze rare populations in the bone marrow, such as HSCs. All classes of hematopoietic cells are derived from HSCs. Stem cell biology draws enormous attention not only from scientists, but also from ordinary people because of the tremendous potential for development of new therapeutic application to diseases that currently lack any type of effective therapy. Thus, this type of “regenerative medicine” is a relatively new and attractive field in both basic science and clinical medicine.

Book excerpt: Hematopoietic stem cells give rise to all the differentiated elements of the blood, and much progress has been made in recent years in understanding the intricate network of cytokines that regulate the survival, self-renewal and differentiation of these cells. This special issue of Acta Haematologica contains selected contributions from the 10th Symposium on 'Molecular Biology of Hematopoiesis', held in Hamburg in 1997. Leading specialists in the field discuss the roles of cytokines and report preclinical and clinical experience with growth factors and chemotherapy in the treatment of leukemia and lymphoma. Revealing the mechanisms responsible for normal and abnormal cell proliferation and differentiation, and presenting innovative approaches for blood and marrow transplantation, this publication is recommended reading for all oncologists and hematologists.

Available in PDF, EPUB and Kindle. Book excerpt: Philadelphia chromosome-negative myeloproliferative neoplasms (MPNs) - including polycythemia vera (PV), essential thrombocythemia (ET) and primary myelofibrosis (PM) - are clonal stem cell disorders characterized by acquired activating mutations which result in an excessive production of red blood cells, platelets, and neutrophils. Recent advances in the understanding of the molecular biology and genomics of MPNs have led to the development of novel targeted treatment approaches; however, the high rate of life-threatening vascular events and the high risk of disease progression with myelofibrotic and leukemic transformation continue to present challenges in the management of MPNs. Thereby, there remains a need for novel treatment strategies. Recently, research has revealed a role for clonal hematopoiesis, driver mutations and inflammation in thrombosis risk, which may provide a basis for novel thrombosis-prevention strategies.

Available in PDF, EPUB and Kindle. Book excerpt: Hematopoiesis, or the process of blood formation, has been extensively studied at both basic and clinical levels. Human diseases such as thalassemia, immunodeficiency, and leukemia represent defects in this process. Approaches to treat these disorders have required a basic understanding of the biology of blood cells. For instance, hemapoietic stem cell replacement or bone marrow transplantation has been used to ameliorate disease. This volume focuses on hematopoiesis at a cellular and molecular level, and establishes the basis for clinical manipulation of hematopoietic cells for therapeutic benefit. In Part I, the cellular characteristics of progenitors and stem cells are explored. Emphasis is placed on purification of stem cells and both in vitro and in vivo assays. The regulation of normal and leukemis stem cells is illustrated. An excellent discussion of potential use of these cells for gene therapy concludes this section. Hemapoiesis is easily studied during embryogenesis. Part II develops the concept of the waves of hemapoiesis during development. Comparative hematology is making a major comeback as a field in the 1990's. One hope is that general principles of hematopoiesis will be established by studying many models and systems. Part III delves into critical factors that regulate hematopoiesis, including both intracellular and extracellular signals. Part IV and V describe lineage programs for myeloid and lymphoid lineages. These chapters are meant to be illustrative of the different cell fates, but are not exhaustive. Part VI examines the genetics of hematopoisis, particularly in animal models. The hematopoietic system is in constant contact with stromal cells and endothelial cells during development and in the adult. Evidence suggests that endothelial cells and blood cells may arise from a common progenitor, the hemangioblast. Part VII and VIII discuss the stromal and endothelial cells with the emphasis on their interaction with hematopoietic cells.
