Although IGTS8 possesses the ability to specifically remove organic sulfur from coal, a major research need is to develop improved strains of microorganisms that possess higher levels of desulfurization activity and therefore will permit more favorable biodesulfurization process conditions: faster rates, more complete removal, and smaller reactor size. strain improvement is the single most important aspect to the development of a practical coal biodesulfurization process and accordingly is the focus of research in this project. During this quarter the promoter probe vectors that were constructed last quarter were found to be unstable in E. coli. Fragments of R. rhodochrous IGTS8 chromosomal DNA were cloned into pRCAT3 and pRCM1 (previously described in final ICCI report 1993). Many derivatives of pRCM1 and pRCAT3 receiving inserts that regulated the expression of chloramphenicol resistance in Rhodococcus rhodochrous IGTS8 proved to be unstable in E. coli frequently yielding plasmids containing deletions. Stable inserts have been observed ranging from 100 bp to 2.0 kb that regulated expression in Rhodococcus rhodochrous IGTS8. Subtractive hybridization studies continue, several candidates have been isolated and are being confirmed for inducible promoters. Primer extension analysis of the Rhodococcus rhodochrous IGTS8 16S RNA promoter region was initiated this quarter.

Strain improvement is the single most important aspect to the development of a practical coal biodesulfurization process and accordingly is the focus of research in this project. Several possible strong promoters have been isolated and are in the process of being analyzed. When these promoters have been characterized for inducibility, strength, transcriptional start sites and other physical properties, they will be placed in front of the desulfurization genes and expression will be monitored. Improved promoter probe vectors have been constructed, allowing a conclusive screen of all putative Rhodococcus promoters. With the improved methodologies in the handling of Rhodococcus RNA, we have begun to gauge promoter expression using Northern blots. During this quarter we have constructed and successfully used a promoter probe vector using the [beta]-galactosidane gene from E. coli. A chromosomal promoter library was constructed upstream from the [beta]-galactosidase gene. Over 200 colonies were isolated that yielded [beta]-galactosidase activity.

The evaluation of the ability of improved cultures to desulfurize coal will be evaluated during the final quarter of this project.

Book excerpt: This project seeks to determine the identity of representative organosulfur compounds removed by coal desulfurization reactions. Demineralized coals from the Illinois Basin Coal Sample Program are solvent extracted and the extracts fractionated to separate and concentrate the organosulfur compounds for analysis by Gas Chromatography/Mass Spectroscopy. After the sulfur compounds are characterized, the parent extracts will be subjected to reactions previously shown to reduce the organic sulfur content of Illinois coals. The treated extracts will be fractionated and analyzed for organosulfur content by the same method to determine which compounds reacted during the chemical treatment. Finally, the original coal will be subjected to chemical desulfurization, extraction, fractionation and analysis in order to correlate changes in organic sulfur content of the coal with reactions of specific sulfur compounds. These compounds can thus be reliably considered as target molecules for the next generation of desulfurization processes. Work during this quarter included studies to increase the amount of soluble material extracted from coal by several solvents and development of standard chromatographic methods for fractionation and analysis of soluble coal fractions. Pyridine solubility of several coals is enhanced by removal of mineral matter prior to extraction. The effect is modest (12% relative increase) for Illinois No. 6 coal and greater for lower rank coal. Ultrasound and swelling agents cause a large increase in pyridine solubility. Although toluene extracts much less material from Illinois coals than pyridine, removal of mineral matter prior to extraction causes a relatively larger increase in extraction yield (60 - 80% relative increase). Expected aromatic sulfur compounds such as dibenzothiophene have been detected by GC/MS analysis of non-polar fractions of Illinois No. 6 extracts and several oxidized thiophenes were detected in extracts of weathered IBC-105 coal.

At the initiation of this program, the basic elements needed for performing genetic engineering in T. ferrooxidans were either not yet developed. Improved techniques are described which will make it easier to construct and analyze the genetic structure and metabolism of recombinant T. ferrooxidans. The metabolism of the model organic sulfur compound dibenzothiophene (DBT) by certain heterotrophic bacteria was confirmed and characterized. Techniques were developed to analyze the metabolites of DBT, so that individual 4S pathway metabolites could be distinguished. These techniques are expected to be valuable when engineering organic sulfur metabolism in Thiobacillus. Strain isolation techniques were used to develop pure cultures of T. ferrooxidans seven of which were assessed as potential recombinant hosts. The mixotrophic strain T. coprinus was also characterized for potential use as an electroporation host. A family of related Thiobacillus plasmids was discovered in the seven strains of P. ferrooxidans mentioned above. One of these plasmids, pTFI91, was cloned into a pUC-based plasmid vector, allowing it to propagate in E. coli. A key portion of the cloned plasmid was sequenced. This segment, which is conserved in all of the related plasmids characterized, contains the vegetative origin of DNA replication, and fortuitously, a novel insertion sequence, designated IS3091. The sequence of the DNA origin revealed that these Thiobacillus plasmids represent a unique class of replicons not previously described. The potentially useful insertion sequence IS3091 was identified as a new member of a previously undefined family of insertion sequences which include the E. coli element IS30.
