Book excerpt: The developments in mass spectrometry over the past fifteen years have been impressive in their implications in bioanalytical chemistry. The achievements begin with the inventions of Cf-252 Plasma Desorption Mass Spectrometry by Macfarlane and Fourier Transform Mass Spectrometry by Comisarow and Marshall in the mid 1970s. The former showed the feasibility of producing large gas-phase ions from large biomolecules whereas the latter enhanced the capabilities for ion trapping especially in analytical mass spectrometry. A major achievement was the development by Barber of Fast Atom Bombardment (FAB) mass spectrometry, an advance that heralded a new era in biological mass spectrometry. Contemporary and routine instruments such as magnetic sectors and quadrupoles were rapidly adapted to F AB, and nearly the entire universe of small molecules became amenable to study by mass spectrometry. The introduction of FAB also paved the way for improvement of instrument capability. For example, the upper mass limit of magnet sector mass spectrometers was increased by nearly an order of magnitude by the instrument manufacturers. Furthermore, the technique of tandem mass spectrometry (MS/MS) was given new meaning because important structural information for biomolecules could now be produced for ions introduced by FAB into the tandem instrument. The evolution of MS/MS continues today with the development of ion traps, time-of-flight, and sector instruments equipped with array detection.

Fast atom bombardment (F AB) ionization, developed by Barber and coworkers, made it possible for all mass spectrometry laboratories to analyze polar, highly functionalized organic molecules, and in some cases ionic, inorganic, and organometallic compounds. The emphasis of much of this work was on molecular weight determination. Parallel with the development of ionization methods (molecular weight mass spectrometry) for polar biological molecules, the increased mass range of sector and quadrupole mass spectrometers and the development of new instruments for tandem mass spectrometry fostered a new era in structural mass spectrometry. It was during this same period that new instrument technologies, such as Fourier transform ion cyclotron resonance, radio frequency quadrupole ion trap, and new types of time-of-flight mass spectrometers, began to emerge as useful analytical instruments. In addi tion, laser methods useful for both sample ionization and activation became commonplace in almost every analytical mass spectrometry laboratory. In the last 5 years, there has been explosive growth in the area of biological mass spectrometry. Such ionization methods as electrospray and matrix-assisted laser desorption ionization (MALDI) have opened new frontiers for both molecular weight and structural mass spectrometry, with mass spectrometry being used for analysis at the picomole and even femto mole levels. In ideal cases, subfemtomole sample levels can be successfully analyzed. Sample-handling methods are now the limiting factor in analyz ing trace amounts of biological samples.

Not the least of these uses of free-jet expansions, is that of extractive sampling from high temperature, reactive systems. The conversion of the free-jet expanded gases to molecular flow for direct introduction into the ion source of a mass spectrometer offers several advantages, to be illustrated in these pages. Two meetings on this subject were held in 1965 and 1972 in Missouri, sponsored by the Office of Naval Research and Midwest Research Institute. At these meetings rarefied gas dynamicists came together with scientists using free-jet sampling for analytical purposes. After much too long a time, this workshop was convened to bring together modem practitioners of FJMBS (Free-jet, Molecular-beam, mass spectrometry) and long time students of the free-jet process itself, to assess the current state of the art and to forge a community that can foster the development of this novel analytical approach. The intensity of 2.5 x 10 to the 18th power molecules 1/sq.cm./sec. and the spreading angle of 6 deg agree with a theoretical analysis which takes into account attenuation caused by scattered molecules which have been thermally accommodated at the walls of the apparatus. A modified Kantrowitz-Grey system is used to obtain the high-energy beam.

Book excerpt: A molecular beam-mass spectrometer (MBMS) has been developed to sample ambient gas atmosphere in epitaxial growth systems at high temperatures and at atmospheric pressure. At present the MBMS is capable of analyzing ppm quantities of gaseous species that result from chemical transport reactions between the ambient H2 atmosphere and the growth system components (e.g., fused quartz and graphite). The MBMS consists of three differentially pumped stages separated by a nozzle and a skimmer orifice and a detector aperture. The gas is expanded through the nozzle to establish a supersonic flow region (Mach disc) which is sampled by the skimmer to develop a well-collimated molecular beam. The molecular beam is then ionized and detected by a quadrapole mass analyser. The response and interpretation of the data from the MBMS are analyzed and discussed to obtain an overall calibration of the system. The sensitivity for most gaseous species in H2 at atmospheric pressure and at 800 C is better than 1 ppm without chopping the beam. Initial results indicate that there are ppm levels of Ar, CO and H2O in palladium purified H2. In addition, the formation of SiO and H2O by chemical transport reactions between H2 and fused quartz at 700 C and above are in good agreement with existing high temperature thermodynamic data.
