The toxins produced by cyanobacteria, cyanotoxins, are harmful to both humans and animals. Rising temperatures due to global climate change, increased nutrient loading, and other anthropogenic impacts on waterbodies are expected to increase the prevalence of cyanobacteria. It is vital that we protect our drinking water supplies and natural water resources. Modeling the production and movement of these toxins is an important step in limiting exposure to them and evaluating management strategies to mitigate their impact. Cyanotoxins are diverse and the conditions under which they are formed are variable and depend on species, strain, and environmental factors. The research provided here offers an overview of some of the environmental factors and cyanobacteria species that are associated with toxin production, and the research also presents preliminary models for the transport and fate of cyanotoxins. Cyanotoxins can be either intracellular or extracellular and a model for each was developed. The models were first tested using published data from laboratory experiments, and then the models were incorporated into the two-dimensional (longitudinal and vertical) hydrodynamic and water quality model CE-QUAL-W2. The toxin models were tested using a model of Henry Hagg Lake (Oregon). Additional research was done to improve the water quality predictions of the CE-QUAL-W2 model of Henry Hagg Lake that had previously been developed. This involved updating the model simulation period through the end of 2020 and calibrating the model to better match field data through the new simulation period. The preliminary models were able to capture similar dynamics as the published data from the laboratory experiments, but the toxin data available at Henry Hagg Lake was minimal so it was difficult to compare the model results to the field data using the CE-QUAL-W2 model. Four scenarios were conducted to test the functionality of the toxin models in CE-QUAL-W2. The predicted results from each test scenario matched expected outcomes based on the parameters used in each scenario. Further applications of the toxin models to other waterbodies with more consistent toxin data will help verify the accuracy of the preliminary models. In addition, further research of the environmental factors that affect toxin production is necessary to incorporate variable rates of toxin dynamics. While the simulations of the Henry Hagg Lake CE-QUAL-W2 model closely match the field data for many water quality parameters, additional calibration of the model is required to refine the results. The preliminary models should provide a framework to develop more specific models through continued research of cyanotoxins.

Book excerpt: Previous studies on harmful algae blooms (HABs) and cyanotoxin contamination in Lake Erie mainly focused on surface water bodies, and the knowledge of groundwater contamination caused by cyanotoxin migrated from HABs polluted lakes remains notably limited. Following a recent proof-of-concept, MODFLOW-based modeling study (Abesh et al., 2022), this study aimed to advance our understanding of such a contamination issue using a newer generation hydrologic model, i.e., HydroGeoSphere (HGS). Specifically, a 3-D HGS model was developed to simulate pumping-induced reverse groundwater flow from Lake Erie to coastal aquifers. The model domain, encompassing South Bass Island, its aquifer, and the surrounding lake, was discretized into 15 vertical layers, consisting of 7,543 2-D triangular elements each. The model was applied to explore how different hydrogeologic settings (including fracture configuration, lake levels, hydraulic conductivity, dispersivity, and reactions coefficients) would impact the transport processes. Simulation results show that fractures along longitudinal flow directions and in the forms of a connected matrix, high lake levels, high dispersion, low hydraulic conductivity, and low retardation factors would facilitate such movements. We estimated the arrival time of microcystin and identified pumping strategies for management and decision-making on drinking wells. Based on simulation results, wells closer to the lake with higher pumping rates would reach EPA's health advisory (HA) levels faster. For example, a water well 25 m away from the coast could exceed HA levels for infants-children (0.3 ug/l) and adults (1.6 ug/l) in about 47 and 115 days, respectively, if pumping at 200 m3/d. At the same rate but 50 m away from the lake, it would take 50 days to reach the infants-children level, but hard to reach the adult level. Pumping iv wells over 75 m away from the lake either took very long days or did not reach HA levels and hence seemed safe from microcystin contamination. However, unfavorable hydrogeologic conditions (e.g., fractured geology, increasing lake levels due to climate change, and higher microcystin concentrations caused by severe HABs events in the lake) could cause uncertainties in microcystin transport and threaten nearby wells. Therefore, we strongly recommend that drinking wells along the Lake Erie coast should be regularly monitored and tested for the presence of microcystin.

Book excerpt: Anatoxin-a (ANTX) is a neurotoxin produced by several freshwater cyanobacteria and has been implicated in the death of livestock and domestic animals from consumption of tainted surface waters. ANTX is unstable under normal conditions and is somewhat problematic to extract and study. Accelerated solvent extraction (ASE) combined with liquid chromatography-mass spectrometry (LC/MS) was used to develop an efficient extraction and analytical method for both ANTX and the more commonly encountered hepatotoxic microcystins produced by cyanobacteria. The effects of nitrogen supply on the cellular production and release of ANTX was investigated in Aphanizomenon issatschenkoi (Ussaczew) Proschkina-Lavrenko (Nostocales). In contrast to the predictions of the carbonnutrient balance hypothesis, the maximum production was observed under moderate N stress. In addition, steady state fugacity-based models were employed to investigate ANTX's distribution and fate in freshwater ecosytems. ANTX was not found to be very persistent in aquatic ecosystems and did not appear to bioaccumulate in fish, at least not from the dissolved phase.

Eutrophic freshwater lakes that receive regular inputs of nutrient-rich runoff from agricultural land are an ideal environment for cyanobacteria blooms, which are associated with cyanotoxin production, but cyanotoxins could also be present in, or originate from, the soils, groundwater and the vadose zone of agricultural watersheds. Humans will be exposed to these toxins when they consume cyanobacteria-contaminated groundwater and crops grown in contaminated soils. However, we do not understand fully how agricultural nutrient loading may trigger cyanotoxin production and the occurrence of cyanotoxins in ecosystem compartments besides eutrophic waterbodies. One of the major challenges is to develop quantitative methods that can detect the biologically significant cyanotoxins at low concentrations, and another challenge is to sensitize the agricultural community to the risk of cyanotoxins to agricultural plants, which has consequences for the health of animals and humans that consume those plants. Therefore, my thesis aimed to address these knowledge gaps. First, I completed a critical review, proposing that nutrient loading from agricultural runoff may trigger beta-N-methylamino-L-alanine production in cyanobacterial blooms, based on evidence from Lake Winnipeg, Canada. I provide evidence that allochthonous agricultural nutrients may trigger beta-N-methylamino-L-alanine production by influencing the composition of potentially toxic cyanobacteria species and the nitrogen availability. Then, I confirmed that cyanotoxins are present in ecosystem compartments beside eutrophic waterbodies, based on the detection of microcystins in agricultural soils and subsurface water (drainage water, well water and municipal drinking water) in agricultural watersheds in Quebec, Canada. However, the semi-quantitative method in this study was not conclusive and led me to develop a method to extract and quantify the cyanotoxins in soil. My new method can efficiently detect 15 cyanotoxins in soils. Finally, I evaluated the phytotoxicity of microcystins in soil on agricultural plants and determined the human health risk from consuming microcystins-contaminated plants. Based on my meta-analysis, microcystins are potentially most phytotoxic to potato but leafy vegetables such as dill, parsley and cabbage could bioconcentrate ~3 times more microcystins than other agricultural plants. Consuming leafy vegetables containing microcystins could be risky to adults and children because the estimated daily intake values (> 0.2 μg kg-1d-1) exceed the WHO guidelines (0.04 μg kg-1d-1). In conclusion, my research showed that cyanotoxins are detectable in terrestrial and subterranean environments where they are seldom studied, and reveals a public health risk associated with microcystins in the edible components of agricultural plants. My findings support the One Health approach to manage the risk of public exposure to toxic substances, and indicate that cyanotoxins warrant investigation in a greater number of environments than are monitored at present"--
