Book excerpt: The past decade has seen a remarkable increase in the use of electron microscopy as a researm tool in biology and medicine. Thus, most institu tions of higher learning now boast several electron optical laboratories having various levels of sophistication. Training in the routine use of elec tron optical equipment and interpretation of results is no longer restricted to a few prestigious centers. On the other hand, temniques utilized by researm workers in the ultrastructural domain have become extremely diverse and complex. Although a large number of quite excellent volumes of electron microscopic temnique are now dedicated to the basic elements available whim allow the novice to acquire a reasonable introduction to the field, relatively few books have been devoted to a discussion of more ad vanced temnical aspects of the art. It was with this view that the present volume was conceived as a handy reference for workers already having some background in the field, as an information source for those wishing to shift efforts into more promising temniques, or for use as an advanced course or seminar guide. Subject matter has been mosen particularly on the basis of pertinence to present researm activities in biological electron microscopy and emphasis has been given those areas whim seem destined to greatly expand in useful ness in the near future.

Book excerpt: Recent advances in imaging technology reveal, in real time and great detail, critical changes in living cells and organisms. This manual is a compendium of emerging techniques, organized into two parts: specific methods such as fluorescent labeling, and delivery and detection of labeled molecules in cells; and experimental approaches ranging from the detection of single molecules to the study of dynamic processes in organelles, organs, and whole animals. Although presented primarily as a laboratory manual, the book includes introductory and background material and could be used as a textbook in advanced courses. It also includes a DVD containing movies of living cells in action, created by investigators using the imaging techniques discussed in the book. The editors, David Spector and Robert Goldman, whose previous book was Cells: A Laboratory Manual, are highly respected investigators who have taught microscopy courses at Cold Spring Harbor Laboratory, the Marine Biology Laboratory at Woods Hole, and Northwestern University.

Book excerpt: Individual cells behave in surpnsmg ways that cannot be deduced from the averaged results of an organ as assessed by the use of conventional biochemical methods. Thus multicellular plant and animals systems are being investigated by an increasing array of histochemical and cytochemical techniques based on general chemical or specific immunological interactions to identify structural materials and to assess biological activities. In recent years there has been an increasing range of fluorescent probes, along with advanced computerised imaging and analysis techniques, which allows the behaviour of individual living cells to be followed in considerable detail. The parallel use of microinjection, microelectrodes and patch-clamping provides additional information about cells and their responses. Recombinant DNA technology has highlighted the desirability and the power of microinjecting defined materials into specific cells and so manipulating their fundamental biochemistry. New hypotheses are being tested which will form the cornerstone of future developments across the whole spectrum of biotechnology. The First European Workshop on Biotechnology Applications of Microinjection, Microscopic Imaging and Fluorescence was run at the University of East London, U.K, 21st-24th April, 1992 with the objective of bringing together a diverse group of individuals who were using these state-of-the-art applications for biotechnological exploration. A novel feature of the meeting was paiticipation by instrument manufacturers in the programme: there were hands-on workshops (where living cells could be examined), combined with the poster sessions.

Available in PDF, EPUB and Kindle. Book excerpt: In Confocal Microscopy Methods and Protocols, Stephen Paddock and a highly skilled panel of experts lead the researcher using confocal techniques from the bench top, through the imaging process, to the journal page. They concisely describe all the key stages of confocal imaging-from tissue sampling methods, through the staining process, to the manipulation, presentation, and publication of the realized image. Written in a user-friendly, nontechnical style, the methods specifically cover most of the commonly used model organisms: worms, sea urchins, flies, plants, yeast, frogs, and zebrafish. Centered in the many biological applications of the confocal microscope, the book makes possible the successful imaging of both fixed and living specimens using primarily the laser scanning confocal microscope. The powerful hands-on methods collected in Confocal Microscopy Methods and Protocols will help even the novice to produce first-class cover-quality confocal images.

Available in PDF, EPUB and Kindle. Book excerpt: Basic Confocal Microscopy, Second Edition builds on the successful first edition by keeping the same format and reflecting relevant changes and recent developments in this still-burgeoning field. This format is based on the Confocal Microscopy Workshop that has been taught by several of the authors for nearly 20 years and remains a popular workshop for gaining basic skills in confocal microscopy. While much of the information concerning fluorescence and confocal microscopy that made the first edition a success has not changed in the six years since the book was first published, confocal imaging is an evolving field and recent advances in detector technology, operating software, tissue preparation and clearing, image analysis, and more have been updated to reflect this. Several of these advances are now considered routine in many laboratories, and others such as super resolution techniques built on confocal technology are becoming widely available.
