Book excerpt: One of the primary impediments to the implementation of bioremediation is uncertainty about success in the field. Soils and microbial populations are heterogeneous and it is difficult to extrapolate biodegradation rates from small samples to field scale. While biodegradation rates can be estimated from microcosm studies, in situ methods offer a more meaningful gauge of resident microbial activity. One method used to estimate biodegradation rates in the field is the newly developed Push-Pull technique. While this technique can be conducted on site, it is normally not possible to use target pollutants as the reactive substrates. Consequently, alternative, benign reactive tracers must be used. Ideally, these alternative, reactive tracers interrogate the same enzyme systems that are responsible for the biodegradation of the target pollutant. The objective of this study was to develop a reactive tracer system that could be used to assess toluene-dependent trichloroethylene (TCE) degradation. Our approach has been to determine whether a series of pure strains of toluene-oxidizing bacteria (Burkholderia cepacia G4, Pseudomonas putida F1, Pseudomonas putida mt2, Pseudomonas mendocina KR1), each with different toluene-oxidizing enzymes systems, are capable of cometabolically oxidizing a series of eleven potential alternative substrates. These substrates include simple alkenes, alkanes, and cyclic alkanes. The kinetics (Ks and Vmax) of the biotransformation of these compounds have been determined. While oxidation products were observed for a number of these substrates in connection with one or more of the test organisms, isobutylene was co-oxidized by all test organisms. Oxidation of isobutylene by each organism yielded kinetics constants comparable to the corresponding kinetics of TCE degradation. The enzyme system expressed by Burkholderia cepacia G4 catalyzed the epoxidation of isobutylene while the remaining enzyme systems catalyzed allylic alcohol formation. Isobutylene.

In a previous study, the initial rate of TCE oxidation by Pseudomonas putida Fl declined rapidly. This was shown in vivo, in the present study, to be due to toluene dioxygenase-dependent activation of TCE to produce reactive intermediates. Carbon-14 label from metabolism of radioactive TCE was incorporated into proteins, small molecules, DNA, RNA, and lipid. Alkylation of proteins, including toluene dioxygenase, caused metabolic poisoning and the loss of TCE-degrading ability. P'. putida was able to recover from TCE-mediated cytotoxicity. Toluene dioxygenase enzyme components were purified from recombinant E. coli strains by the use of conventional chromatographic methods. The major products of TCE oxidation by purified toluene dioxygenase have been identified as formic acid and glyoxylic acid. An additional minor product was shown to result from the alkylation of reduced pyridine dinucleotide (NADPH) by a reactive TCE intermediate generated by the toluene dioxygenase catalyzed reaction. Experiments with 14C-TCE showed that enzyme inactivation was due to non-specific alkylation of the proteins by diffusible reactive intermediates. A recombinant E. coli expressing cloned toluene dioxygenase genes oxidized TCE. The rate of TCE oxidation by the E. coli strain was slower than that of P. putida Fl but the rates were sustained for a longer time. The use of recombinant strains or gratuitously induced P. putida Fl could have important implications for bioremediation. Furthermore, a greater understanding of the mechanism of toluene dioxygenase inactivation will be crucial for developing optimum systems for TCE-biotreatment.

Soil and vegetation (Lespedeza cuneata, Paspalum notatum, Pinus taeda, and Solidago sp.) samples from the Miscellaneous Chemicals Basin (MCB) at the Savannah River Site were used in tests to identify critical plant and microbiological variables affecting the fate of trichloroethylene (TCE) in the root zone. Microbiological assays including phospholipid acid analyses, and 14C-acetate incorporation were conducted to elucidate differences in rhizosphere and nonvegetated soil microbial communities from the MCB. The microbial activity, biomass, and degradation of TCE in rhizosphere soils were significantly greater than corresponding nonvegetated soils. Vegetation had a positive effect on microbial degradation of 14C-TCE in whole-plant experiments. Soils from the MCB containing Lespedeza cuneata, Pinus taeda, and Glycine max mineralized greater than 25% of the 14C- TCE added compared with less than 20% in nonvegetated soils. Collectively, these results provide evidence for the positive role of vegetation in enhancing biodegradation.

Microorganisms tolerant to TCE were isolated from an aeration tower at Wurtsmith AFB MI and from industrial sludges at Hill AFB UT. Isolates from the Wurtsmith samples exhibited the highest density of cells with TCE as the only apparent organic carbon source and were selected as the inoculum for degradation studies. However, the capability of selected microorganisms to degrade TCE was not substantiated in both gas chromatography analyses and samples inoculated in culture bottles. Experiments using radiolabeled carbon-14 to study assimilation suggest that TCE is metabolized into a polar form by the microorganism. Subsequent cultures grown in radiolabeled TCE did not duplicate the previous results possible because of contamination, selection of variants or loss of culture. The organism responsible or the apparent TCE degradation may belong in the genus Citrobacter.
