Book excerpt: Recent years have seen an explosion of research on imaging biomarkers for Alzheimer's disease and related neurodegenerative disorders, fueled by the realization that the enormous cost of conducting clinical trials for disease-modifying treatments stands as a major obstacle on the path to finding cures. Imaging biomarkers promise to reduce this cost by allowing more accurate detection of prodromal disease (enabling trials to recruit individuals that are most likely to benefit from treatment) and allowing more accurate monitoring of the effects of disease and treatment on the brain (enabling earlier detection of outcomes in trials). Recent research on imaging biomarkers for neurodegenerative diseases has been aided by the availability of large imaging datasets like the Alzheimer's Disease Neuroimaging Initiative (ADNI) and its European and Asian counterparts.Much of the progress in improving imaging biomarkers for neurodegeneratice disease has been driven by the development of novel computational imaging analysis methodology, particularly in the areas of segmentation, normalization, image-based classification, and high-dimensional statistics. Yet the venues where such work is typically presented (AAIC/ICAD, ISMRM, HBM) are more focused on applications, with only occasional short symposia related to computational methodology. Thus, we believed that it was highly appropriate and timely to hold a workshop at MICCAI that would specifically focus on the methodological aspects of imaging biomarker research in AD and related disorders.The workshop, held on October 5, 2012 in Nice, France, included an traditional original research paper track with 21 papers spanning the range of topics including image segmentation, longutidinal imaging analysis, functional and diffusion imaging biomarkers, and validation. It also included a challenge component focused on measurement of longitudinal brain atrophy in AD, with 9 papers.
