Book excerpt: A cultura da cana-de-açúcar possui enorme importância no cenário agrícola brasileiro por séculos, tomando o Brasil o maior produtor mundial com esta cultura. A capacidade de aumento na quantidade de cana-de-açúcar produzida no Brasil enfrenta limitações físicas (existência de novas áreas de cultivo) e tecnológicas (aumento da capacidade produtiva por programas de melhoramento, melhoria dos ambientes de cultivo). Estudos de inoculação identificaram uma mistura contendo cinco espécies de bactérias diazotróficas endofiticas como capaz de aumentar significativamente a matéria seca das plantas e incorporar até 33% do nitrogênio contido na planta via FBN, em comparação com valores obtidos de plantas não inoculadas. Neste trabalho, foram estudados parâmetros microbiológicos e agronômicos da associação entre plantas de cana-de-açúcar e bactérias diazotróficas endofiticas. Demonstramos a influência da umidade do solo na sobrevivência de algumas espécies endofiticas na ausência da planta hospedeira, sendo possível detectar células de bactérias endofiticas após 40 dias de inoculação no solo com elevada umidade. Demonstramos a permanência das espécies inoculadas ao longo de dois ciclos produtivos da cana-de-açúcar, em níveis populacionais que variaram de acordo com a época de coleta, desde uns poucos isolados até populações acima 105 células por grama de tecido vegetal. Os efeitos da inoculação de duas misturas de bactérias diazotróficas endofiticas foram avaliados em duas variedades de cana-de-açúcar micropropagadas, sob diferentes condições de manejo, ocorrendo tanto o aumento como a diminuição da produtividade em resposta aos tratamentos de inoculação, conforme as interações entre os parâmetros avaliados. A variedade de cana-de-açúcar SP70-1143 obteve contribuições significativas na produtividade de colmos e na produtividade de sacarose, na ausência de adubação nitrogenada, para as classes Planossolo e LVA (baixa e média fertilidade natura4 respectivamente). Na classe de solo TRE (elevada fertilidade natural), não houveram diferenças significativas entre os tratamentos de inoculação e as plantas controle para a variedade SP70-1143, enquanto para a variedade SP81-3250 notamos uma diminuição não significativa na produtividade de colmos, para as três classes de solo. As maiores contribuições da FBN foram observadas no Planossolo, chegando a 31,3% no primeiro corte da variedade SP70-1143, e 42,7% no segundo corte da variedade SP81-3250. Além disso, a contribuição significativa da FBN esteve associada a aumentos significativos na produtividade apenas para a variedade SP81-3250. Na avaliação da transmissibilidade dos efeitos da inoculação, foram encontradas diferenças significativas na produtividade das parcelas, havendo correspondência entre as parcelas oriundas dos tratamentos de inoculação e as parcelas que receberam fertilizante nitrogenado, além do reisolamento de todas as espécies utilizadas nas inoculações. The sugarcane culture has great importance in Brazilian agribusiness, turning Brazil the largest world producer of this crop. Several species of plant growth promoter microrganisms have being described in association with sugarcane, what turns possible searching for species to use as inoculant. Inoculation studies identified a mixture containing five species of endophytic diazotrophic bacteria able to increase significantly the dry matter content, and to incorporate up to 33% of the total nitrogen contained in the plant through BNF. In this work, we studied microbiological and agronomic parameters of the association between sugarcane plants and endophytic diazotrophic bacteria. We demonstrated the influence of soil moisture in survival of some of the endophytic bacteria, in absence of host plant, and that was possible to detect culturable cells in selective semi-solid media after 40 days of inoculation in the presence of high moisture. We also evaluated permanence ofthe inoculated species along two productive sugarcane cycles, in population levels that varied in time from some few isolates, to populations above 105 cells per gram of plant wheight. In another experiment, we tested two different mixtures of endophytic diazotrophic bacteria, in two micropropagated sugarcane varieties, under different levels of N-fertilizer, in a field trial for two consecutive crops, in three soil classes (Ultisol, Alfisol and Oxisol). We found the increasing as well as the decreasing of productivity, depending on interactions among tested parameters. The sugar-cane variety SP70-1143 obtained significant contributions in stem and sucrose productivity from the biological f1Xation, in absence of nitrogen fertilization, when cultivated in soil classes Oxisol and Ultisol (the lowest and average natural fertility, respectively). In the Alfisol soil class (high natural fertility), there were no significant differences between the inoculation treatments and control p1ants of the SP70-1143 variety; while the SP81-3250 variety presented not significative decreases in productivity of stems in all the soil classes. It was demonstrated that the productivity and FBN were under influence of the plant genotipe, soil class, and nitrogen fertilization level. The 1argest FBN contributions were observed in the lowest fertility soil, reaching up to 31.3% in the first cut for SP70-1143 variety, and 42.7% in the second cut ofSP81- 3250 variety. It was demonstrated that the significant contribution of FBN was not always associated to significant increases in plant productivity. In the evaluation of transmissibility of the inoculation effects, we found significant differences among productivities, having correspondence with productivity of p1ants originated from inoculated treatments, and plants that received nitrogen fertilization, besides reisolation of all the species used in in vitro inoculations.

An over-arching theme of all chapters is the interaction of the plants and trees with microbes and this theme allows other aspects of soil microbiology, such as interactions with arbuscular mycorrhizal fungi and the impact of soil-stress factors on biological nitrogen fixation, to be addressed. Furthermore, a link to basic science occurs through the inclusion of chapters describing the biogeochemically important nitrogen cycle and its key relationships among nitrogen fixation, nitrification, and denitrification. The volume then provides an up-to-date view of the production of microbial inocula, especially those for legume crops.
