Book excerpt: Studies have been made of the toxicology and chemistry of nitrogen trifluoride, tetrafluorohydrazine, chlorine trifluoride, bromine pentafluoride, and oxygen difluoride. Lethality of each agent by inhalation has been determined, as well as estimates of lethality after intraperitoneal administration. Experimental evidence suggests that the lethal effect of interhalogens is by corrosive local destruction of pulmonary surfaces, resulting in failure of gas exchange. Oxygen difluoride is thought to pass intact into the pulmonary cells where it reacts with biochemical reducing systems to ultimately cause cell death and structural failure. The nitrogen fluorides both cause intrinsically lethal levels of methemoglobin formation, but other pharmacologic activity by these compounds or their derivatives may also contribute to their toxic activity.

AMOX produced strong irritation resulting in pulmonary edema and hemorrhage after inhalation; and local denatured changes and accumulation of clear exudate after intraperitoneal injection. TAMA was less toxic on the weight basis. Both AMOX and TAMA were quickly absorbed after IP injection. Fluoride increased quickly in the blood and concentrated in various tissues especially in the thyroid, bone and teeth. Most of the administered fluoride was excreted in the urine and a considerable amount was also recovered in the feces.

Workers and residents in communities surrounding industrial facilities where these substances are manufactured, used, or stored and in communities along the nation's railways and highways are potentially at risk of being exposed to airborne EHSs during accidental releases or intentional releases by terrorists. Pursuant to the Superfund Amendments and Reauthorization Act of 1986, the U.S. Environmental Protection Agency (EPA) has identified approximately 400 EHSs on the basis of acute lethality data in rodents. Acute Exposure Guideline Levels for Selected Airborne Chemicals, Volume 18 identifies, reviews, and interprets relevant toxicologic and other scientific data for selected AEGL documents for bromine chloride, carbonyl fluoride, selected halogen fluorides, and oxygen difluoride in order to develop acute exposure guideline levels (AEGLs) for these high-priority, acutely toxic chemicals. AEGLs represent threshold exposure limits (exposure levels below which adverse health effects are not likely to occur) for the general public and are applicable to emergency exposures ranging from 10 minutes (min) to 8 h. Three levels - AEGL-1, AEGL-2, and AEGL-3 - are developed for each of five exposure periods (10 min, 30 min, 1 h, 4 h, and 8 h) and are distinguished by varying degrees of severity of toxic effects. This report will inform planning, response, and prevention in the community, the workplace, transportation, the military, and the remediation of Superfund sites.

However, with the demonstration that water-borne fluoride was a causative agent of both mottled enamel and increased resistance to dental caries, the ground work was laid in the 1930's and early 1940's for a greatly increased interest in the biological effects of fluorides in human beings. During this time and earlier for that matter, work also had been going steadily ahead in the less spectacular area of effects produced in poultry and livestock when fluorine-containing rock phosphate was incorporated in the ration, and when pasture land was contami nated with fluorides released during the large-scale conversion of rock phosphate to fertilizer and phosphoric acid.
