Book excerpt: The eighth workshop in this series on Mechanisms in B-Cell Neoplasia 1990 was held in Wilson Hall at the National Institutes of Health, Bethesda, Maryland on March 28-30. Five major topics formed the basis for the discussions: 1) progress in experimental models of B-cell tumorigenesis, 2) the role of IL-6 in plasma cell tumor formation with particular emphasis on human myeloma, 3) immortaliza tion and regulation of mitosis in B-cells, 4) the mYQ gene in B-cell neoplasia, and 5) the role of EBV and other oncogenes in transforma tion of human B-Iymphocytes. A meeting on the Epidemiology of Myeloma was held at the N. I. H. on the preceding day, and many of those interested in the clinical aspects of myeloma were also participants at the workshop. Experimental Models of B-Cell Tumor Development We have seen in the last eight years the steady growth of model experimental systems, many of which have been designed to be counter parts of the major forms of human B cell tumors, e. g. , follicular lymphomas, Burkitt's lymphomas, acute B-cell leukemia and multiple myeloma. A variety of novel ways of inducing these tumors has been described. Advantage has been taken of the "experiments in nature" to identify critical genes that playa role in tumor pathogenesis. These genes have been identified by being near to viral insertion and chromosomal translocation sites, or by having been incorporated or transduced into a defective transforming retrovirus.

Initially, PI3K inhibitors such as Wortmannin, LY294002 and Rapamycin were used to establish a central role for Pi3K pathway in immune cells. Considerable progress in understanding the role of this pathway in cells of the immune system has been made in recent years, starting with analysis of various PI3K and Pten knockout mice and subsequently mTOR and Foxo knockout mice. Together, these experiments have revealed how PI3Ks control B cell and T cell development, T helper cell differentiation, regulatory T cell development and function, B cell and T cell trafficking, immunoglobulin class switching and much, much more. The PI3Kd inhibitor idelalisib has recently been approved for the treatment of B cell lymphoma. Clinical trials of other PI3K inhibitors in autoimmune and inflammatory diseases are also in progress. This is an opportune time to consider a Research Topic considering when what we have learned about the PI3K signalling module in lymphocyte biology and how this is making an impact on clinical immunology and haematology.

Book excerpt: It is of critical importance to maintain an appropriate balance between proliferation and quiescence or differentiation through out the lifespan of all animals. An important control point in this balance occurs in the G, phase of the cell cycle. On the basis of environmental cues a cell in G, must decide whether to continue through the proliferative cycle and enter S phase (where DNA replication occurs) or to exit from the proliferative cycle into a nonreplicating state. Alterations in the mechanisms that nor mally control this decision can lead to cancer, cell death, or loss of differentiated cellular phenotypes. The identification of the E2F gene family of transcription factors has allowed a more complete understanding of how the cell maintains an appropri ate proliferative state. This volume provides an up-to-date ac count of present reports concerning E2F as well as a framework for future investigations. E2F activity requires heterodimerization of two partners. Either partner can be one of several different transcription factors; E2Fl, E2F2, E2F3, E2F4, or E2F5 can heterodimerize with either DPl or DP2. Cellular promoters whose E2F sites mediate a link between transcription and proliferation drive genes whose products are required for DNA synthesis and genes that encode regulators of cell growth. A detailed analY$is of the role that E2F family members play in transcription from these promoters is presented in the chapter by J. E. SLANSKY and P. J. FARNHAM.

In 1995 the organizers decided that the format of the meeting would be changed and that specific topics relevant to B-cell neoplasia would be discussed. This year's topic is on the c-myc oncogene in B-cell neoplasia which has been discussed in virtually every previous meeting. Some of the presentations announced for the first time dramatic advances in our understanding of c-myc and because this subject has become highly complex it was thought that devoting the whole meeting to this theme would be appropriate. The book, therefore, repre sents a review of many aspects of the myc problem but by no means is truly comprehensive. In a recent Medline search there were 8,505 references to myc, fully illustrating the magnitude of the interest and depth of this field. The organizers of the meeting have each contributed review chapters that summarize different aspects of the meeting. We thank the National Cancer Institute for sponsoring this workshop and the staff of Cygnus, Inc. , for their outstanding organizational assistance. "What is the best way to detect and define pluripotent hematopoietic stem cells?" was the question posed by Dr. Fritz Melchers. After discussing the pros and cons of the available assays for quite some time, it became apparent that this topic required a wider expertise and merited a larger forum. Thus, we decided to extend the discussion and to compile the results in this volume. Much to our delight. many of the pioneers of recent experimental and theoretical developments in stem cell research agreed to contribute their expertise to answer the question. These authors review both past findings and present insights, thus providing an overview of the evolution that has been and is occurring in the field of stem cell research. In the light of recent trailblazing developments in both experimental models and in clinical application it is indeed time to reevaluate our knowledge about stem cells. Trans plantation of hematopoietic stem cells has become more and more prevalent as a curative therapy in a variety of acquired and genetic diseases, including cancer, radiation accident, as an agent for gene therapy, and perhaps even as treatment for autoimmune diseases. Stem cells are now derived not only from bone marrow but also from peripheral blood, cord blood, and fetal liver, greatly increasing their availability for human transplantation and in some cases (fetal tissues) obliterating the need to match donors and hosts.
