Domenico Bertoloni Meli here explores Malpighi’s work and places it in the context of seventeenth-century intellectual life. Malpighi’s interests were wide and varied. As a professor at the University of Bologna, he confirmed William Harvey’s theory of the circulation of blood; published groundbreaking studies of human organs; made important discoveries about the anatomy of silkworms; and examined the properties of plants. He sought to apply his findings to medical practice. By analyzing Malpighi’s work, the author provides novel perspectives not only on the history of anatomy but also on the histories of science, philosophy, and medicine. Through the lens of Malpighi and his work, Bertoloni Meli investigates a range of important themes, from sense perception to the meaning of Galenism in the seventeenth century. Bertoloni Meli contends that to study science and medicine in the seventeenth century one needs to understand how scholars and ideas crossed disciplinary boundaries. He examines Malpighi’s work within this context, describing how anatomical knowledge was achieved and transmitted and how those processes interacted with the experimental and mechanical philosophies, natural history, and medical practice. Malpighi was central in all of these developments, and his work helped redefine the intellectual horizon of the time. Bertoloni Meli’s critical study of this key figure and the works of his contemporaries—including Borelli, Swammerdam, Redi, and Ruysch—opens a wonderful window onto the scientific and medical worlds of the seventeenth century.

These diseases mainly include Alzheimer’s disease (AD), Parkinson’s disease (PD), Huntington’s disease (HD), and amyotrophic lateral sclerosis (ALS). They are significantly associated with age and degeneration, becoming a devastating global health issue as a result of the growth of mean life expectancy worldwide. Few or no effective treatments are available for age-related neurodegenerative diseases, causing large socioeconomic and personal costs. The last 15 years have seen a boom in the use and integration of “omic” approaches (such as genomic, transcriptomic, epigenomic and metabolomic techniques) to study neurodegenerative disease in an unprecedented way. While, to date, no therapy based on these approaches is available to cure neurodegenerative diseases. Therefore, it is necessary to search for new, safe, and effective drug candidates and treatment strategies. In this Research Topic, our prime focus would be on the use and integration of “omic” approaches to study biomarkers, pathogenesis, disease progression and new therapeutic target of neurodegenerative diseases, so as to search for new, safe, and effective drug candidates.

Evidence-based medicine seeks to achieve improved health outcomes by making evidence explicit and by developing explicit methods for evaluating it. To date, evidence-based medicine has largely focused on evidence of association produced by clinical studies. As such, it has tended to overlook evidence of pathophysiological mechanisms and evidence of the mechanisms of action of interventions. Book excerpt: This book, Mechanisms in the Pathogenesis of Enteric Diseases 2, is an out come of the Second International Rushmore Conference on Mechanisms in the Pathogenesis of Enteric Diseases, held September 3D-October 3, 1998 in Rapid City, South Dakota, USA. Its chapters represent many of the reviews and papers presented at the conference. The meeting was organized by members of the North-Central Regional Research Committee "NC-62", a consortium of researchers of bovine and swine enteric diseases from land-grant institutions supported by the United States Department of Agriculture. The Rushmore Conferences were conceived as a forum for an interdiscipli nary discussion of mechanisms of infectious enteric diseases. It was intended that such a discussion would stimulate cross-pollination of ideas, and nurture synergistic collabora tions among scientists who might otherwise not interact. Enteric diseases are caused by widely divergent pathogens and parasites in broadly different settings, and affect multi ple organ systems. Some enteric diseases affect a single species, while others may affect multiple species, perhaps including human beings. Some enteric diseases were present in antiquity, while others have recently emerged. Knowledge regarding a particular disease or pathogen has frequently proven useful in understanding another disease or pathogen, because common themes in pathogenesis exist. As this knowledge base grows,strategies in the prevention and control of various enteric diseases often converge. Cross-discipli nary discussions and collaborations facilitate growth of this knowledge base, as well as development of tools for disease interdiction.

Book excerpt: It has been more than 25 years since the identification of the FMR1 gene and the demonstration of the causative role of CGG-repeat expansion in the disease pathology of fragile X syndrome (FXS), but the underlying mechanisms involved in the expansion mutation and the resulting gene silencing still remain elusive. Our understanding of the pathways impacted by the loss of FMRP function has grown tremendously, and has opened new avenues for targeted treatments for FXS. However, the failure of recent clinical trials that were based on successful preclinical studies using the Fmr1 knockout mouse model has forced the scientific community to revisit clinical trial design and identify objective outcome measures. There has also been a renewed interest in restoring FMR1 gene expression as a possible treatment approach for FXS. This special issue of Brain Sciences highlights the progress that has been made towards understanding the disease mechanisms and how this has informed the development of treatment strategies that are being explored for FXS.
