Book excerpt: This work reveals that triangular silver nanoparticles have remarkable optical properties and that the enhanced sensitivity of their nanoenvironment can be used as a new class of optical sensors using localized surface plasmon resonance (LSPR) spectroscopy. The contents of thesis can be divided into two main subject areas: (1) the characterization and optimization of nanoparticles for the basic understanding of their optical properties and (2) the development, testing, and theoretical understanding of model biological assays and nonmodel assays for disease diagnosis. In the first half of this work, an improved understanding of the physical characteristics of noble metal nanoparticles is explored. Specifically, the realization that the sensitivity of these nanoparticles can be optimized for a given adsorbate is revealed. First, it is demonstrated that the short range (viz., 0--3 nm) distance dependence of the electromagnetic fields that surround these nanoparticles can be systematically tuned by changing their composition, size, and structure. Second, multilayer shells are assembled onto surface-confined noble metal nanoparticles to study the long range distance dependence. It is shown that this dependence (viz., 3--35 nm) is non-linear and has a tunable sensing range. Finally, methods to enhance the nanosensor's response based on interactions between the nanoparticle's plasmon resonance and the adsorbate's molecular resonance is revealed. In the remainder sections, we explore the use of these nanoparticles as biological sensors. First, the model system of biotin/streptavidin is confirmed to be highly specific with low nonspecific influences. Next, the demonstration of an immunoassay (biotin/anti-biotin) is performed. Finally, the LSPR nanosensor is shown to be a promising platform for the possible mechanistic understanding of Alzheimer's disease.

This field has shown a dramatic growth thanks to the creation of sensitive thin films onto diverse optical fiber configurations. In this sense, a wide range of optical fiber devices have been successfully fabricated for monitoring biological, chemical, medical or physical parameters. In addition, the use of nanoparticles into the sensitive thin films has resulted in an enhancement in the response time, robustness or sensitivity in the optical devices, which is associated to the inherent properties of nanoparticles (high surface area ratio or porosity). Among all of them, the metallic nanoparticles are of great interest for sensing applications due to the presence of strong absorption bands in the visible and near-infrared regions, due to their localized surface plasmon resonances (LSPR). These optical resonances are due to the coupling of certain modes of the incident light to the collective oscillation of the conduction electrons of the metallic nanoparticles. The LSPR extinction bands are very useful for sensing applications as far as they can be affected by refractive index variations of the surrounding medium of the nanoparticles, and therefore, it is possible to create optical sensors with outstanding properties such as high sensitivity and optical self-reference. In this chapter, the attractive optical properties of metal nanostructures and their implementation into different optical fiber configuration for sensing or biosensing applications will be studied.

Book excerpt: In recent times, metallic nanoparticle plasmonics coupled with applications towards biosensing has gathered momentum to the point where commercial R&D are investing large resources in developing the so-called localized surface plasmon resonance (LSPR) biosensors. Conceptually, the main motivation for the research presented within this thesis is achievement of fully-operational LSPR biosensor interfaced with the state-of-the-art microfluidics, allowing for very precise control of sample manipulation and stable read-out. LSPR sensors are specifficaly engineered by electron beam lithography nanofabrication technique, where nanoparticle interactions are optimized to exhibit increased sensitivity and higher signal-to-noise ratio. However, the overall performance of LSPR lab-on-a-chip device depends critically on the biorecognition layer preparation in combination with surface passivation. As an introduction, the principles of plasmonic biosensing are identified encompassing both Surface Plasmon Resonance (SPR) and Localized SPR. Being successfully implemented into commercial product, the governing physics of SPR is compared to LSPR in chapter 1, together with advantages and disadvantages of both. Chapter 2 describes methods necessary for LSPR biosensor development, beginning with nano-fabrication methods, the modelling tool (COMSOL Multiphisics), while the basics of micro-fabrication in microfluidics conclude this chapter, where passive and active microfluidics networks are discerned. Particularly attractive optical properties are exhibited by closely-coupled nanoparticles (dimers), with the dielectric gap of below tens of nm, which were theoretically predicted to be very suitable as LSPR biosensing substrates. Chapter 3 is subjected to optical characterization (dependence on the size of the dielectric gap) of nanofabricated dimer arrays. The acquired data demonstrate the advantages of the nanofabrication methods presented in chapter 2 and the technique for fast and reliable determination of nanoparticle characteristic parameters. The initial biosensing-like experiments presented in chapter 4 (no integration with microfluidics) proved for the first time, the theoretical predictions of higher sensitivity, yielding additionally the specific response as function of analyte size and dielectric gap between nanoparticles. The overall response of different dimer arrays (various gaps) provides information about adopted conformation of analyte protein once immobilized. Broad resonances of dimers feature higher noise when employing them for the real-time LSPR biosensing. As a way to circumvent such problem, the feasibility of employing far-field interaction within the nanoparticle array to spectrally narrow resonance is investigated in chapter 5 by optimizing the array periodicity and introducing thin waveguiding layers. Finally, the concluding chapter 6 is dedicated to a full assembly of a Lab-on-a-chip (LOC) LSPR biosensor, starting with interfacing plasmonic substrates with compatible active microfluidic networks, allowing the precise sample delivery and multiplexing. The prototype device consisting of 8 individual sensors is presented with typical modes of operation. The bulk refractive index determination of various samples demonstrates the working principle of such device. Finally, various strategies of biorecognition layer formation are discussed within the on-going research.
