The control of gene expression is a central-most topic in molecular biology as it deals with the utilization and regulation of gene informa tion. As we see huge efforts mounting all over the developed world to understand the structure and organization of several complex eukaryotic genomes in the form of Gene Projects and Genome Centers, we have to remember that without understanding the basic mechanisms that gov ern the use of genetic information, much of this effort will not be very productive. Fortunately, however, research during the past seven years on the mechanisms that control gene expression in eukaryotes has been extremely successful in generating a wealth of information on the basic strategies of transcriptional control. (Although regulation of gene ex pression is exerted at many different levels, much of the emphasis in this series will be on transcriptional control. A future volume, however, will deal with other levels of regulation). The progress in understanding the control of eukaryotic transcription can only be appreciated by realizing that seven years ago we did not know the primary structure of a single sequence specific transcriptional activator, and those whose primary structures were available (e. g. , homeo domain proteins) were not yet recognized to function in this capacity.

Book excerpt: Since the 1st Yamaguchi Symposium on Liver Diseases in 1989, this series of symposia has provided opportunities for exchanges of information on the topic between leading Japanese hepatologists and internationally renowned scientists. Somewhat unusually for meetings held in Japan, the official language of the symposium is English. The pro ceedings of these symposia are published under the title Frontiers in Hepatology and distributed worldwide. The 12th symposium was held on December 9 and 10,2000, at the ANA Hotel, Ube, Japan. The theme selected by the Organizing Committee was "Growth, Proliferation, and Apoptosis in Hepatocytes;' each of which is important in the understanding of the pathophysiology of intractable liver disease. Nine Japanese hepatologists were invited to give presentations, as was leading u.S. researcher Professor D.A. Brenner, recently elected editor-in-chief of the journal Gastroenterology. The reports given at the two-day meeting were valuable in furthering our under standing of the complicated signaling system involved in hepatocyte differentiation, proliferation, and apoptosis. Progress in this field is rapid, and another symposium on the same theme will be held in the near future. We believe that these proceedings are useful in summarizing current information on this important topic. The Organizing Committee would like to express special thanks to all participants and to the Viral Hepatitis Research Foundation of Japan for its continuing financial support.

New concepts in the field of transplantation, for instance the transplantation of living donor organs or the splitting of organs, are urgently required, to safeguard the treatment of patients with severe liver disease. The development and clinical application of cell therapy for patients with liver disease could soon present a significant enhancement of the therapeutic options. The aim of such cell therapy is to repair or improve the biological function of the chronically and acutely damaged liver. Even though systematic trials are not available, individual case reports and small series already show promising clinical results. Present concepts of cell therapy for liver diseases based on the use of primary hepatocytes have recently been considerably extended through new data on the biology of stem cells. The adult haematopoetic stem cell as a pool for hepatocyte grafts - what would be the perspectives for the clinical application? Abstract: Abstract of thesis entitled Functional characterization of the liver-enriched transcription factor CREB-H submitted by King-Tung Chin, Tony for the Degree of Doctor of Philosophy at the University of Hong Kong April 2005 The establishment and maintenance of a differentiated phenotype in the liver require a specific gene expression program coordinated by a group of liver-specific transcription factors. Dysregulation of liver-specific transcription is a hallmark of hepatocellular carcinoma. We have previously characterized transcription factor LZIP to be a candidate tumor suppressor targeted by an oncoprotein encoded by hepatitis C virus. Similarity of LZIP to Drosophila fat body-specific transcription factor called box B-binding factor 2 suggests the existence of additional LZIP-like liver-enriched proteins in mammals. In search of proteins closely related to LZIP, we have identified a novel liver-enriched transcription factor CREB-H. This thesis reports on the functional characterization of CREB-H. First I performed bioinformatic analysis of CREB-H-related sequences. LZIP and CREB-H represent a new subfamily of bZIP proteins distinct to other well- characterized members of the family such as CREB, C/EBPα and ATF6. One notable feature of this new subfamily of bZIP factors is the existence of a unique putative transmembrane domain in the middle of protein right next to the highly conserved bZIP region. viiiNext I characterized the properties of CREB-H protein in cultured hepatoma cells using a combination of biochemical and cell biological methods. Reporter transcriptional assays indicated that CREB-H has one activation domain and two repression domains. Electrophoretic mobility shift assays and analysis of transcriptional activities demonstrated that CREB-H activates transcription by binding to cAMP responsive element, box B, and ATF6-site/UPRE-binding element. Confocal microscopy revealed that CREB-H localizes ambiently to the endoplasmic reticulum. In addition, I provided the first evidence for proteolytic processing of CREB-H in cultured mammalian cells. Importantly, I observed that proteolytic cleavage that removed the transmembrane domain led to nuclear translocation and activation of CREB-H. To investigate the roles of CREB-H in liver-specific transcription, I asked whether CREB-H regulates the promoter of hepatic gluconeogenic enzyme phosphoenolpyruvate carboxykinase. Analysis of transcriptional activities using reporter assays and chromatin immunoprecipitation assays verified the activation of phosphoenolpyruvate carboxykinase promoter by CREB-H. Interestingly, I found that this activation by CREB-H can be further stimulated by cAMP and protein kinase A. Another salient finding in this thesis is the underexpression of CREB-H in liver cancer. Northern blotting, Western blotting and semi-quantitative reverse transcription-polymerase chain reaction analyses indicated that CREB-H is exclusively abundant in adult liver but aberrantly underexpressed in hepatoma tissues and cells. Consistent with this expression pattern, the enforced expression of CREB-H suppressed the proliferation of cultured hepatoma cells. ixIn summary, my findings suggest that the liver-enriched bZIP transcription factor CREB-H is a growth suppressor that plays an important role in hepatic physiology and pathology.

Although most c- tributions in this volume focus on mammalian circadian clocks, the historical int- duction and comparative clocks section illustrate the importance of various other organisms in deciphering the mechanisms and principles of circadian biology. Circadian rhythms have been studied for centuries, but only recently, a mole- lar understanding of this process has emerged. This has taken research on circadian clocks from mystic phenomenology to a mechanistic level; chains of molecular events can describe phenomena with remarkable accuracy. Nevertheless, current models of the functioning of circadian clocks are still rudimentary. This is not due to the faultiness of discovered mechanisms, but due to the lack of undiscovered processes involved in contributing to circadian rhythmicity. We know for example, that the general circadian mechanism is not regulated equally in all tissues of m- mals. Hence, a lot still needs to be discovered to get a full understanding of cir- dian rhythms at the systems level. In this respect, technology has advanced at high speed in the last years and provided us with data illustrating the sheer complexity of regulation of physiological processes in organisms. To handle this information, computer aided integration of the results is of utmost importance in order to d- cover novel concepts that ultimately need to be tested experimentally.
