Book excerpt: Classical histology has been augmented by immunohistochemistry (the use of specific antibodies to stain particular molecular species in situ). Immunohistochemistry has allowed the identification of many more cell types than could be visualized by classical histology, particularly in the immune system and among the scattered hormone-secreting cells of the endocrine system. This book discusses all aspects of immunohistochemistry and in situ hybridization technologies and the important role they play in reaching a cancer diagnosis. It provides step-by-step instructions on the methods of additional molecular technologies such as DNA microarrays, and microdissection, along with the benefits and limitations of each method. * The only book available that translates molecular genetics into cancer diagnosis * Methods were developed by internationally-recognized experts and presented in step-by-step manner * Results of each Immunohistochemical and in situ hybridization are presented in the form of color illustrations

This book was released on 2008-02-02 with total page 471 pages. Available in PDF, EPUB and Kindle. Book excerpt: Soon after the first description of monoclonal antibodies in 1976, there was enormous interest in the clinical application of antibodies, especially in the context of cancer. Antibodies appeared to offer the “magic bullet” that would allow the specific destruction of neoplastic cells. H- ever, many years’ effort resulted in very few cases of successful immu- therapy with antibodies. As a result there was a major backlash against antibody therapy, and the field lost a considerable amount of popularity. Fashion, in science as well as in other things, tends to be cyclical. Antibody-based therapy is once again attracting scientists and clinicians. There are several reasons for the renewed optimism; certainly the expe- ence of the last two decades has provided a wealth of information about problems associated with antibody therapy, and possible solutions to these problems. Recombinant antibody engineering has rejuvenated the field, allowing both the modification of antibodies to improve their in vivo pr- erties and the isolation of novel antibody molecules by such techniques as phage display. The results of recent clinical trials have demonstrated unequivocally the benefit of antibody therapy in a number of settings, and, finally, more careful consideration has been taken of the types of disease best treated using this approach.

Book excerpt: As immunohistochemistry enters its fourth decade as a diagnostic tool, this analytical procedure continues to experience poor reproducibility, albeit in specific situations. The problem becomes more important with the mounting pressure to employ the assay in a quantitative manner for the assessment of therapeutic and prognostic markers in a number of neoplasms. Qualitative applications of immunohistochemistry are well established and continue to increase with the range of sensitive antibodies and detection systems available, however, numerous variables that influence the immunoexpression of proteins in formalin-fixed tissue continue to exist in the pre-analytical and analytical phases of the test procedure. This book emphasises these problems in detail and suggests possible solutions.
