Book excerpt: Several previous Advanced Study Institutes have concentrated on the techniques of light scattering, while the biological appli cations were not fully explored. Many of the techniques are now standardised and are being applied to a wide range of biologically significant problems both in vivo and in vitro. While laser light scattering methods are superior to conventional methods, there was a general reluctance among biologists to adopt them because of the complexity of the physical techniques and the accompanying mathe matical analysis. Consequently valuable opportunities for advancing the understanding of the biological problems were being missed. Advances in the design and commercial availability of standard light scattering instruments, and the availability of standard computer programs, made the more widespread use of these techniques a practical reality for the biologist. While biologists are unable to cope with the complexities of the physical techniques, physicists are generally unaware of the nature and scale of the biological problems. The meeting at Maratea was an attempt to bring these two groups together and provide an impetus for the application of laser light scattering techniques to biology. This volume differs from the three previous proceedings on laser light scattering in the NATO ASI series (B3, B23, B73), in that it has been published in the Life Sciences series rather than the Physics series, reflecting the shift in emphasis from the development of a new technique to its application in biology.

Light scattering by small particles such as, for instance, droplets in the terrestrial clouds is a well understood area of physical optics. On the other hand, exact theoretical calculations of light scattering p- terns for most of nonspherical and irregularly shaped particles can be performed only for the restricted values of the size parameter, which is proportional to the ratio of the characteristic size of the particle to the wavelength?. For the large nonspherical particles, approximations are used (e. g. , ray optics). The exact th- retical techniques such as the T-matrix method cannot be used for extremely large particles, such as those in ice clouds, because then the size parameter in the v- iblex=2?a/???,wherea is the characteristic size (radius for spheres), and the associated numerical codes become unstable and produce wrong answers. Yet another problem is due to the fact that particles in many turbid media (e. g. , dust clouds) cannot be characterized by a single shape. Often, refractive indices also vary. Because of problems with theoretical calculations, experimental (i. e. , la- ratory) investigations are important for the characterization and understanding of the optical properties of such types of particles. A unique apparatus using a laser as a light source for ultramicroscopy and angular scattering experiments is described. The results obtained utilizing this apparatus for investigations of imperfections in ruby crystals are presented and discussed. These investigations were done as a function of crystal growth direction, annealing, boule morphology, polarization, chromium concentration, gamma-irradiation, and temperature. The results of this work demonstrate the usefulness of this new equipment as a convenient, nondestructive technique for characterizing imperfections in crystals of relatively large size. They also indicate the existence of both randomly distributed scatterers and oriented scattering regions in ruby crystals and show how these scattering centers are altered by changes in the various parameters studied in this investigation. Book excerpt: Static light scattering (SLS) and dynamic light scattering (DLS) techniques are used to measure characteristics of particles in dispersion. SLS measures the dependence of the average scattered intensity I, on the scattering angle and is sensitive to spatial variations in the dielectric constant E. DLS measures the time dependence of the scattered light. One fundamental application of light scattering techniques is the accurate measurement of the size distribution of particles in dispersion. Detailed experimental investigation of SLS and DLS has been attempted in this work, with experimental work based on dilute water dispersions of two different spherical particles, polystyrene latexes and poly(N-isopropylacrylamide) (PNIPAM) microgels. Size information is obtained from SLS measurements in the form of a particle size distribution G (R,) where R, is the static radius. Size information is obtained from DLS measurements in the form of a decay rate distribution G (I?) which depends on the decay rate I?. Although both SLS and DLS can be used to obtain size information from the scattered light, the information obtained using SLS and DLS is different. For the three polystyrene latex sphere samples studied in this thesis, the mean static radii obtained are consistent with that of the mean radii provided by the supplier. For all three samples, the apparent hydrodynamic radius is larger than the mean static radius by about 12%. For PNIPAM microgel spheres, the apparent hydrodynamic radius is also larger than the mean static radius. The size of the PNIPAM microgel particles is extremely temperature sensitive; the radius decreases by a factor of three as the temperature is raised from 15OC to 50?? Both the hydrodynamic and static radii show this behavior. The effect of the chemical crosslinker (N, N'-methylenebisacrylamide) content on the temperature sensitivity of the PNIPAM microgels and the temperature dependence of the ratio of the apparent hydrodynamic radius to the mean static radius are also discussed.
