Book excerpt: Noncrystalline thermooxidatively stable aromatic p olyamides were synthesized through the triphenyl phosphite-promoted polycondensations of 3,3-(sulfonylbis(1,4-phenylenoxy)) dibenzoic acid and 3,3(carbonylbis(1,4-phenylenoxy)) dibenzoic acid with 3,3-(1,3-phenylendioxy)dianiline. Alternatively, the polymers could be prepared through the low temperature polydensations in N-methylpyrollidone of the corresponding diacid chlorides with the aromatic diamine. An additional polyamide was synthesized through the self-condensation of 3-(4-(3-aminophenoxybenzoyl)phenoxy) benzoic acid. This previously unreported monomer had been prepared by a two-step reaction sequence from 4,4-difluorobenzophenone. Inherent viscosities in the range of 0.27-0.63 dl/g (N, N-dimethylacetamide, 30C, 0.2 g/d1) were obtained. Polymer structures were established by elemental analyses and infrared spectroscopy. Thermal characterization was carried out by differential scanning colorimetry, thermal gravimetric analysis, and isothermal aging. Book excerpt: New polyamides containing reactive diacetylene groups in their repeat structures were synthesized from a diacetylene diacid chloride and two different diamines using interfacial polymerization techniques. Thermal analysis and carbon 13 NMR spectroscopy were used to estimate the extent of conversion of the diacetylene groups to polydiacetylene network chains upon exposure to electron beam radiation in the solid state. The diacetylene groups contained in a polyamide based on an aliphatic diamine were found to be more reactable than those incorporated in a polyamide based on an aromatic diamine. The cross-polymerized polyamide-diacetylenes were found to exhibit reversible thermochromic behavior during temperature cycling which occurs with complete retention of hydrogen bonds and without any hysteresis effects. The thermochromic reversibility of these materials was attributed to the strongly hydrogen bonded chain segments pendent to the newly formed polydiacetylene chains. (AW).

Book excerpt: Thianthrene-2,7- and -2,8-dicarboxylic acids plus a synthetic intermediate, 4,4'-thiobis(3-chlorobenzoic acid), have been synthesized in good yields via nucleophilic aromatic substitution of Na2S on N, N-dimethyl-3,4- dichlorobenzamide. New aromatic polyamides having inherent viscosities of 1.29 to 2.39 dL/g were prepared by the direct polycondensation reaction of the dicarboxylic acids with 4,4'-oxydianiline and 1,4-phenylenediamine in N-methyl- 2-pyrrolidinone using triphenyl phosphite and pyridine. The fused-ring thianthrene-based polyamides were more soluble than analogous poly(thioether amide)s. Polymer films were cast from either DMF or DMAc/LiCl solutions and analyzed by FTIR and NMR. All prepared aramids displayed good thermal stability by DSC and TGA.

Book excerpt: This thesis is divided into four sections, all related to polyanhydride synthesis and characterization: first, the synthesis and characterization of polyethylene glycol containing aromatic polyanhydrides, second, the microwave polymerization of polyanhydrides, third the parallel synthesis and high-throughput dissolution screening, and finally preliminary investigations into the parallel investigation of blend phase behavior of a ternary polyanhydride blend. Oligomers of ethylene glycols were used to selectively modify the hydrophobicity of an aromatic polyanhydride. Aromatic diacids were synthesized from chlorinated triethylene and pentaethylene glycols and hydroxy benzoic acid. The diacids were acetylated with acetic anhydride to produce homopolymers of polyanhydrides for the stabilization of proteins and drugs. The glycol containing polyanhydrides were characterized by NMR, GPC, DSC and dissolution testing. The release characteristics of the new polyanhydrides were evaluated with a hydrophilic (Acid Orange) and hydrophobic dye (Rhodamine B Base) and the release mechanisms were investigated. Microwave radiation was used to explore the high throughput synthesis of polyanhydrides for materials library characterization. Aliphatic diacids were acetylated in situ for two minutes in a closed reaction vessel before removal of excess acetic anhydride and subsequent polymerization, all in the microwave. Reaction conditions such as acetic anhydride equivalents, duration of microwave exposure and comparision to conventional polycondensation were explored. Microwave polymerization times of 6 to 8 minutes yielded optimum number average molecular weights in contrast to conventional polycondensation, which took between 1.5 to 2 hours under high vacuum. A strategy was developed to synthesize copolymers of polyanhydrides using photolithographically generated microwells. The copolymers were characterized by NMR and spectroscopic FTIR as well as by parallel dissolution of a model dye from 25 copolymers using image analysis. Moreover, preliminary work was conducted on optical methods to investigate the phase behavior of a ternary blend of polyanhydrides in a parallel fashion. In conclusion, this thesis explored the synthesis and characterization of various polyanhydride-based homopolymers and copolymers by conventional as high-throughput techniques for controlled drug delivery applications.
