Book excerpt: This book deals with the cellular biology, biochemistry and physiology of photoreceptors and their interactions with the second-order neurons, bipolar and horizontal cells. The focus is upon the contributions made by these neurons to vision. Thus the basic neurobiology of the outer retina is related to the visual process, and visual defects that could arise from abnormalities in this part of the retina are highlighted in the first 16 chapters. Since all vertebrate retinas have the same basic structure and physiological plan, examples are given from a variety of species, with an emphasis upon mammals, extending to human vision. The last four chapters approach the problem from the other end. This part of the book covers a range of clinical conditions involving visual abnormalities that are due to cellular defects in the outer retina. Although the contents of this book do not represent the proceedings of a conference, the concept arose at an international symposium on 'Recent Advances in Retinal Research' which was held at the International Marine Centre in Oristano, Sardinia. We hope that the book will give a coherent, up to date review of the neurobiology and clinical aspects of the outer retina and encourage further integration of these areas. Retinal neurobiology has been an intense field of investigation for several decades. More recently, it has seen significant advances with the application of modern techniques of cell and molecular biology.

The sequence is 'open' when read in an appropriate frame, which is to say that there is a long run of amino acid codons before the appearance of a terminator codon. How can we find out if this 'unidentified reading frame' (URF) really codes for a genuine protein, and how can we identify it if it exists? There are two general strategies, both of which can also be applied to the characterization of any 'open reading frame' (ORF), whether or not it has been 'identified'. The first and simplest approach involves computer searching and analysis; the second employs antibodies raised against synthetic peptides patterned on the sequence of the expected gene product. Both methods have been used with great success by many investigators. Each has, nonetheless, its pitfalls and frustrations. Book excerpt: Astrocytes were the original neuroglia that Ramón y Cajal visualized in 1913 using a gold sublimate stain. This stain targeted intermediate filaments that we now know consist mainly of glial fibrillary acidic protein, a protein used today as an astrocytic marker. Cajal described the morphological diversity of these cells with some ast- cytes surrounding neurons, while the others are intimately associated with vasculature. We start the book by discussing the heterogeneity of astrocytes using contemporary tools and by calling into question the assumption by classical neuroscience that neurons and glia are derived from distinct pools of progenitor cells. Astrocytes have long been neglected as active participants in intercellular communication and information processing in the central nervous system, in part due to their lack of electrical excitability. The follow up chapters review the “nuts and bolts” of ast- cytic physiology; astrocytes possess a diverse assortment of ion channels, neu- transmitter receptors, and transport mechanisms that enable the astrocytes to respond to many of the same signals that act on neurons. Since astrocytes can detect chemical transmitters that are released from neurons and can release their own extracellular signals there is an increasing awareness that they play physiological roles in regulating neuronal activity and synaptic transmission. In addition to these physiological roles, it is becoming increasingly recognized that astrocytes play critical roles during pathophysiological states of the nervous system; these states include gliomas, Alexander disease, and epilepsy to mention a few.
