The Symposium was named after Alcuin, an Anglo-Latin poet, educator and cleric from York, who was invited to join the court of Charlemagne at Aachen. Alcuin’s first 50 years were spent in Yorkshire, were he was first a pupil, later headmaster of the cathedral school of York, the most renowned of its day. Charlemagne was gathering at Aachen the leading Irish, English and Italian scholars of the age. Alcuin was appointed head of the Palatine school, where Charlemagne himself, his family, his friends, and his friends’s sons were taught. Alcuin introduced the traditions of Anglo-Saxon humanism into Western Europe and was the foremost scholar of the revival of learning known as the Carolingian Renaissance. He also promoted the use of the beautiful Carolingian minuscule script, the ancestor of modern Roman typefaces.

The present endeavor is more modest in scope. Volume 32 appeared in two separate parts, each having its own subeditors, and together the two parts covered nearly all areas of insulin pharmacology. Such comprehensiveness seemed impractical in a new volume. The amount of in formation related to insulin that is now available simply would not fit in a reasonable amount of space. Furthermore, for better or worse, scientists have be come so specialized that a volume providing such broad coverage seemed likely in its totality to be of interest or value to very few individuals. We therefore decided to limit the present volume to the following areas: insulin chemistry and structure, insulin biosynthesis and secretion, insulin receptor, and insulin action at the cellular level. We felt these areas formed a coherent unit. We also felt, perhaps as much because of our own interests and perspectives as any objective reality, that these were the areas in which recent progress has been most dramatic, and yet, paradoxically and tantalizingly, these were the areas in which most has yet to be learned. Even with this limited scope, there are some major gaps in coverage. Regrettably, two important areas, the beta cell ATP-sensitive potassium channel and the glucose transporter, were among these. Nevertheless, the authors who con tributed have done an excellent job, and we would like to thank them for their diligence.

Book excerpt: The observation of invariant or highly conserved sites within a protein's primary sequence is a strong indication that throughout the course of the protein's evolutionary history these particular sites have played a fundamental role in maintaining some aspect of the protein's proper function. An understanding of what these evolutionary restraints are may provide insights into protein design and engineering. Insulin is a small protein tractable for such structure/function studies of highly conserved sites. The organization of this thesis is as follows. Chapter 1 is an overview of insulin and the insulin receptor. A discussion of parameters relevant to clinically used insulins is also presented within this chapter. Chapters 2 and 3 provide structure/function studies of a pair of highly conserved insulin residues (PheB24 and TyrB26) and thus serve as the main focus of this thesis. This was accomplished by systematically substituting and testing a range of amino acids at these positions. In nearly every case, mutations which conferred high receptor binding affinity and native-like activity came with biophysical costs. These chapters also further defined what side chain parameters are allowable for substitution into the PheB24 and TyrB26 receptor binding pockets which were recently elucidated by an X-ray crystal structure of a domain-minimized insulin receptor construct (μIR) bound to insulin. Novel insights into insulin structure and function were also gleaned in these studies by the incorporation of the non-standard amino acids cyclohexanylalanine and 3-iodotyrosine. Chapter 4 describes how one such substitution, 3-iodo-TyrB26, can be used to optimize a rapid-acting insulin analog's resistance to fibrillation without the concomitant cost of a reduction in its onset of action; a problem seen in analogs utilizing canonical amino acids. Such properties may in the future be of some therapeutic value. A summary which explores how the principles learned in this thesis can be utilized within the context of protein design as well as future directions are presented in Chapter 5.

Book excerpt: The elucidation of the mechanism of cellular signal transduction is one of the key challenges in biology. The first step in this pathway has been particularly refractory to study. In the case of insulin action, it is the binding of insulin to its cell surface receptor, a large integral transmembrane glycoprotein. The binding of insulin results in the activation of the cytoplasmic domain of the receptor as a tyrosine kinase. While much is known about the interaction of the activated receptor with its substrates leading to downstream signal transduction, how the binding of insulin to the extracellular domain of the receptor can lead to receptor activation remains unknown. The specific goal of this study is to localize the insulin-binding site of the insulin receptor (IR), and, if possible, to elucidate in molecular details the insulin-IR interaction and the structure-function relationship of the insulin receptor. To achieve this goal with biochemical approach, I used the baculovirus expression system to produce in large quantity the extracellular domain of the human insulin receptor, and used various specific photoaffinity probes to identify the insulin-contacting sites. Peptide identification methods of high sensitivity were developed to detect the labeled IR fragment containing the putative insulin-binding site by HPLC peptide mapping analysis. A small fragment of 1-3 kDa that might contain the putative insulin-binding or contacting site on the receptor à subunit was detected. However, partly due to the lack of an insulin photoprobe with a high efficiency of cross-linking, and a high efficiency peptide mapping method, I failed to obtain a sufficient quantity of the fragment for amino acid sequencing. In an alternative approach, using gold-labeled insulin as a ligand, and sets of electron micrographs obtained by low-dose low temperature dark field scanning transmission electron microscopy (STEM) of the insulin-IR complex, I successfully reconstructed the 3D quaternary structure of the whole 480-kDa human insulin receptor (HIR), complexed with insulin, at a resolution of 20 Å. Contiguous high densities within the 3D structure indicated a two-fold symmetry for this dimeric receptor and the presence of subdomain structures which begin with the extracellular subdomain involved in insulin binding and terminate with the intracellular tyrosine kinase subdomains. These subdomains were confirmed by fitting known high-resolution crystal domain substructures. In addition, the 3D insulin-HIR complex provides evidence for the involvement of only a single insulin molecule in receptor activation. It also explains many of the known characteristics of insulin binding to its receptor and many biochemical findings in the studies of the insulin receptor.
