Book excerpt: The purpose of this dissertation is to develop analytical methods that aid in the detection of forensic analytes. Forensic analytes require methods with increased sensitivity and low limit of detection capabilities. Improvements in separation techniques, surface enhanced Raman spectroscopic techniques, and wire-less gas sensing can each assist in the detection of trace evidence. When surface enhanced Raman is coupled with thin-layer chromatography a mixture of compounds can be separated and transferred to a metal substrate to be detected using Raman spectroscopy. Surface enhanced Raman scattering enhances the Raman signal intensity by placing a metal substrate in close proximity to an analyte. The new method gives a chemically specific intensified signal along with a chromatographic separation. A traditional separation is performed on a TLC plate, allowed to dry, wetted with a solvent, placed in contact with a metal substrate, and detected using Raman. More efficient chromatographic platforms can be implemented with this method. New efficient chromatographic platforms are also beneficial to the detection of forensic analytes. Recently, photolithographically nanofabricated open system pillar arrays have proved to be more efficient separation platforms when compared to traditional TLC. These platforms are a form of ultra-thin layer chromatography. This dissertation describes the effects of manipulation on the inter-pillar gap distances with respect to band dispersion. The studies herein manipulate the pillar arrays in order to optimize the separation platform. The third method developed involved gas sensing of volatile organic compounds. An amorphous ferromagnetic micro-wire was coated with a polymer, where the polymer swelled in response to the gas introduced. When the gas caused the polymer to swell a differential stress response was applied on the micro-wire. The fabricated sensor was tested on simple organic gases but has capabilities to detect low concentrations of low vapor pressure forensic analytes. All three projects were significant advancements in analytical method development. The analytes used were either fluorescent dyes or volatile organic compounds to test feasibility of each method. More efficient chromatographic platforms were fabricated, surface enhanced Raman was coupled to TLC, and a micro-wire gas sensor was calibrated for the studies performed in this dissertation.

Studies have been done on the usefulness of diffuse reflectance (DR) with Fourier transform infrared (FT-IR) spectrometry, using a direct deposit sampling technique for the analysis of ballpoint pen inks. This technique allows screening of a composite ink as a whole, including its dye components, resins, and other additives. A total of 184 ink samples were analyzed by FT-IR, and searchable spectral libraries were created of both whole and extracted inks. The results indicate that the additional information provided by the DR analysis, combined with the complementary information from the TLC analysis, provides enhanced value to the forensic examination of inks.

Book excerpt: The gel ink pen is the fastest growing pen class available on the modern market. Consequently, its prevalence in forensic casework is expected to increase. This poses a challenge to forensic scientists, since the chemistry of gel inks differ to other commonly encountered inks, thus discrimination by traditional methods like Thin Layer Chromatography (TLC) is limited and a new analytical methodology is required for distinguishing different formulations effectively. A study to evaluate discriminating potential of modern spectroscopic techniques alongside traditional methods of ink examination for within and between brand variations of two commonly encountered colours was undertaken. Within brand variation was detected between multiple samples of a limited number of inks using filtered light, FTIR-ATR and specifically Raman Spectroscopy. For 31 blue gel inks, a discriminating power of at least 0.93 was achieved using a combined analytical sequence of TLC, filtered light (IR absorption and IR fluorescence), Vis-MSP (transmittance), FTIR-ATR and Raman Spectroscopy (514.5 nm, 685 nm, 785 nm and 830 nm combined). A discriminating power of 0.98 was achieved for 25 red gel inks using a combination of TLC, filtered light (IR fluorescence only), FTIR-ATR and Raman Spectroscopy (785 nm only). Hybrid gel inks were readily identified using solubility testing, TLC and Raman Spectroscopy combined, with 92% of red inks tested found to contain both dyes and pigments. A library of Raman spectra from 200 known pigments was compiled and CI Pigment Blue 15:1 and 15:3, and Pigment Red 112 and 254 identified as likely colour components of several blue and red gel inks respectively. An objective multivariate statistical methodology incorporating peak selection, data binning and normalisation was developed and successfully applied to both IR and Raman Spectroscopic data. Hierarchical Cluster Analysis (HCA) and Self-Organising Feature Maps (SOFM) were more effective than Principal Component Analysis (PCA) for successful clustering.
