Book excerpt: This report on Future Trends in Inflammation IV records another of the multidisciplinary meetings organized by the European Biological Research Association. The organizers wish to record their thanks to all the participants and delegates who made this such an enjoyable meeting. Our thanks are due to a number of members of the pharmaceutical industry who so kindly donated funds. These funds were used to provide grants for young research workers to attend this meeting. Many people have kindly given their time and help to make this congress possible. It is impossible to list them by name, but we are profoundly grateful. We were dubious at the outset as to whether to hold this meeting in a period when many other meetings are being organized on inflammation. The enthusiastic attendance proves that we were right in going ahead. The next international meeting will be held in France in 1983. For further details please contact: Professor J. P. The finding that cytokines are elevated in various affective and psychotic disorders brings to the forefront the necessity of identifying the precise research domain criteria (RDoCs) that inflammation is responsible for. This task is certainly the most advanced in major depressive disorders. The reason is that a dearth of clinical and preclinical studies has demonstrated that inflammation can cause symptoms of depression and conversely, cytokine antagonists can attenuate symptoms of depression in medical and psychiatric patients with chronic low grade inflammation. Important knowledge has been gained on the symptom dimensions that inflammation is driving and the mechanisms of action of cytokines in the brain, providing new targets for drug research and development. The aim of the book “Inflammation-Associated Depression” is to present this field of research and its implications in a didactic and comprehensive manner to basic and clinical scientists, psychiatrists, physicians, and students at the graduate level.

Book excerpt: This book provides readers with the most up-to-date information on cutting-edge research concerning chronic inflammation. We now know that when inflammation becomes chronic, it acts as a strong disease-promoting factor in a variety of disorders including arteriosclerosis, obesity, cancer, and Alzheimer disease. Chronic inflammation is hence called as the “silent killer”; it upsets the body’s homeostatic mechanism insidiously. In spite of these developments, we know very little about the mechanism underlying chronic inflammation. Particularly, we do not know precisely what induces chronic inflammation or what promotes its prolongation in a spatiotemporal framework. Neither do we have clear knowledge about how chronic inflammation destroys various tissues or how it predisposes individuals to many different diseases. To make the situation worse, we have no effective treatment against chronic inflammation. Since 2010, two major research programs (CREST and PRESTO) aimed at clarifying the mechanisms underlying chronic inflammation were launched in Japan, and investigators of different research areas with a brilliant track record were selected by their research proposals. Subsequently they have made their best efforts to answer the conundrum concerning chronic inflammation. This book is a compendium of such research efforts. In each chapter, the CREST- or PRESTO-funded researchers summarize their original work concerning mechanisms of induction, progression, or resolution underlying chronic inflammation. The most emphasized characteristic is the molecular aspect of chronic inflammation. The book thus presents the most recent progress made in the molecular understanding of chronic inflammation.

Book excerpt: A new perspective on the link between inflammation and cancer Inflammation is the human body’s normal biological response to threats in the modern world, as well as a defense against the harmful influence of pathogens, the environment, and poor nutrition. But what happens when the inflammatory response is triggered repeatedly and sustained for long periods of time? Cancer and Inflammation Mechanisms: Chemical, Biological, and Clinical Aspects discusses the mechanisms by which chronic inflammation can lead to cancer, the various causative agents, and possible prevention methods. A compilation of the latest information coming out of the various fields of cancer research, this book provides a detailed look at inflammation-related carcinogenesis from the perspective of researchers at the forefront of the field. It takes an interdisciplinary approach to the topic, and provides comprehensive information about the major factors at work in inflammation, cancer, and the intersection of the two conditions. Topics include: A general overview of inflammation-related cancer The biochemistry of inflammation and its effects on DNA Molecular biology and the role of microRNA in carcinogenesis Specific causative agents including oncogenic viruses, asbestos, and nanomaterial Anti-inflammatories, nutraceuticals, and other preventative measures A deeper understanding of the mechanisms behind inflammation-related carcinogenesis can lead to better patient outcomes by improving diagnostics and prevention, as well as altering the approach to treatment. Cancer and Inflammation Mechanisms: Chemical, Biological, and Clinical Aspects provides the knowledge base researchers need to push the field forward.

Available in PDF, EPUB and Kindle. Book excerpt: The acute inflammatory response is the body's first system of alarm signals that are directed toward containment and elimination of microbial invaders. Uncontrolled inflammation has emerged as a pathophysiologic basis for many widely occurring diseases in the general population that were not initially known to be linked to the inflammatory response, including cardiovascular disease, asthma, arthritis, and cancer. To better manage treatment, diagnosis, and prevention of these wide-ranging diseases, multidisciplinary research efforts are underway in both academic and industry settings. This book provides an introduction to the cell types, chemical mediators, and general mechanisms of the host's first response to invasion. World-class experts from institutions around the world have written chapters for this introductory text. The text is presented as an introductory springboard for graduate students, medical scientists, and researchers from other disciplines wishing to gain an appreciation and working knowledge of current cellular and molecular mechanisms fundamental to inflammation.

Book excerpt: In November 1998 many of the key leaders of new drug discovery for inflammatory diseases gathered at Hershey, Pennsylvania for the 9th International Conference of the Inflammation Research Association. The Conference was held over a five day period and provided a stimulating environment for the open exchange of important advances in basic inflammation research as well as new drug discovery and development. This book encompasses some of the highlights of several presentations made at the Conference. It contains some of the latest and important developments in the field of inflammation research. Topics include the status of eotaxin and chemokines in asthma and allergy, signal transduction and regulation of diverse mediators such as the JNK group of MAP kinases, TNF and IL-1 signaling of NF-kB as well as regulators of AP-1, macrophage metalloproteinases, lymphotoxin and further insights into the role of MCP-1 in disease. Also discussed are drug targets in rheumatoid and osteoarthritis, fibrotic diseases,...

Available in PDF, EPUB and Kindle. Book excerpt: The microcirculation is highly responsive to, and a vital participant in, the inflammatory response. All segments of the microvasculature (arterioles, capillaries, and venules) exhibit characteristic phenotypic changes during inflammation that appear to be directed toward enhancing the delivery of inflammatory cells to the injured/infected tissue, isolating the region from healthy tissue and the systemic circulation, and setting the stage for tissue repair and regeneration. The best characterized responses of the microcirculation to inflammation include impaired vasomotor function, reduced capillary perfusion, adhesion of leukocytes and platelets, activation of the coagulation cascade, and enhanced thrombosis, increased vascular permeability, and an increase in the rate of proliferation of blood and lymphatic vessels. A variety of cells that normally circulate in blood (leukocytes, platelets) or reside within the vessel wall (endothelial cells, pericytes) or in the perivascular space (mast cells, macrophages) are activated in response to inflammation. The activation products and chemical mediators released from these cells act through different well-characterized signaling pathways to induce the phenotypic changes in microvessel function that accompany inflammation. Drugs that target a specific microvascular response to inflammation, such as leukocyte-endothelial cell adhesion or angiogenesis, have shown promise in both the preclinical and clinical studies of inflammatory disease. Future research efforts in this area will likely identify new avenues for therapeutic intervention in inflammation.
