Book excerpt: ABSTRACT: A UV Laser Induced Fluorescence (LIF) system, previously developed in our laboratory, was modified and used for a series of applications related to the development and optimization of UV LIF spectroscopic measurements of trace contaminants in drinking water and other water sources. Fluorescence spectra of a number of water samples were studied, including those related to the reverse osmosis water treatment and membrane fouling, domestic and international drinking water, industrial toxins, bacterial spores, as well as several fluorescence standards. Of importance was that the long term detection of the trace level of Dissolved Organic Compounds (DOC) was measured, for the first time to our knowledge, over a one week period and with a time resolution of 2.5 minutes. A comparison of LIF emission using both 266 nm and 355 nm excitation was also made for the first time. Such real-time and continuous measurements are important for future water treatment control. The LIF system was modified to accommodate UV Light Emitting Diodes (LED) as alternative excitation sources, and tested for the detection of trace organic species in water. In addition, a compact system using LED excitation and a spectrometer was xviii developed and underwent initial testing. The original LIF system had two laser sources, 266 nm and 355 nm. The additional sources incorporated in the system were UV LEDs emitting at 265 nm, 300 nm, 335 nm and 355 nm. The LED spectral emission was studied in detail, in terms of spectral variability and power output. It was found that all LEDs had some emission in the visible spectrum, and an optical filter was used to remove it. The signal-to-noise ratio for the LED-based systems was determined and compared with that of the LIF system. The fluorescent signal of the LED-based system was smaller by 1 to 2 orders of magnitude, despite the fact that the LED pulse energy was 2 to 3 orders of magnitude less than the laser's. As such, the fluorescent signal from the LED was greater than expected. Therefore, a UV LED may be a compact and much cheaper optical source for future water measurement instruments.

Book excerpt: Laser-induced fluorescence line narrowing was applied to investigate the local fields and interactions of paramagnetic ions in oxide glasses. Studies included the site dependence of energy levels, radiative and nonradiative transition probabilities, homogeneous line broadening, and ion--ion energy transfer of rare earth ions. These results and the experimental techniques are reviewed briefly; the use of paramagnetic ions other than the rare earths is also considered. Recently, laser-excited fluorescence spectroscopy was used to investigate modifications in the local structure of lithium borate glass caused by compositional changes and phase separation and the site dependence of nonradiative relaxation of paramagnetic ions by multiphonon processes. These results and their implications are discussed. 6 figures.

Book excerpt: Thi s book is a general introduction into in vivo chlorophyll fluorescence and contains the contributions of the first International Ch 1 orophyll Fluorescence Sympos i urn he 1 din the Phys i kzentrum Bad Honnef, F. R. G. from June 6 to 8, 1988. This Symposium was made possible by a generous support from the Wilhelm and Else Heraeus Foundation, Hanau, which is gratefully acknmvledged. The book not only comprises all aspects of the applications of chlorophyll fluorescence in photosynthesis, stress physiol ogy, hydrobiology and remote sensing, but also gives access to measuring techni ques, data acqui si ti on and earl i er 1 iterature references. Thus it is far more than just a common proceedings book, it is a general introduction to all forms of application of the non-destructive in vivo chlorophyll fluorescence including the newest results. In a first chapter the inverse correlation between in vivo chlorophyll fl uorescence and photosynthet i c quantum convers i on and CO? -ass i mi 1 at ion is outlined, the origin and life-time of the chlorophyll fluOrescence at room and liquid nitrogen temperatures are given as well as the induction kinetics (Kautsky effect) and the methodo 1 ogi ca 1 approaches to regi ster different forms of chlorophyll-fluorescence signatures.

Book excerpt: Fluorescence methods are being used increasingly in biochemical, medical, and chemical research. This is because of the inherent sensitivity of this technique. and the favorable time scale of the phenomenon of fluorescence. 8 Fluorescence emission occurs about 10- sec (10 nsec) after light absorp tion. During this period of time a wide range of molecular processes can occur, and these can effect the spectral characteristics of the fluorescent compound. This combination of sensitivity and a favorable time scale allows fluorescence methods to be generally useful for studies of proteins and membranes and their interactions with other macromolecules. This book describes the fundamental aspects of fluorescence. and the biochemical applications of this methodology. Each chapter starts with the -theoreticalbasis of each phenomenon of fluorescence, followed by examples which illustrate the use of the phenomenon in the study of biochemical problems. The book contains numerous figures. It is felt that such graphical presentations contribute to pleasurable reading and increased understand ing. Separate chapters are devoted to fluorescence polarization, lifetimes, quenching, energy transfer, solvent effects, and excited state reactions. To enhance the usefulness of this work as a textbook, problems are included which illustrate the concepts described in each chapter. Furthermore, a separate chapter is devoted to the instrumentation used in fluorescence spectroscopy. This chapter will be especially valuable for those perform ing or contemplating fluorescence measurements. Such measurements are easily compromised by failure to consider a number of simple principles.

This book was released on 2007 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: ABSTRACT: We have investigated the use of deep UV laser induced fluorescence for the sensitive detection and spectroscopic lifetime studies of terbium doped dipicolinic acid (DPA-Tb) and used this to study the optical characteristics of DPA which is a chemical surrounding most bacterial spores. Background absorption spectra, fluorescence spectra, and Excitation Emission Matrix (EEM) spectra were made of the DPA-Tb complex, using both fixed 266 nm wavelength and tunable (220 nm -- 280 nm) UV laser excitations. Of importance, the fluorescence lifetimes of the four main fluorescence peaks (488 nm, 543 nm, 581 nm, and 618 nm) of the DPA-Tb complex have been measured for the first time to our knowledge. The lifetimes of all the fluorescing lines have been measured as a function of DPA-Tb concentration, solvent pH, and solvent composition, including that for the weakest fluorescing line of DPA-Tb at 618 nm. In addition, a new spectroscopic lifetime measurement technique, which we call "Transient Fluorescence Spectroscopy" was developed. In this technique, a weak, quasi-CW, amplitude modulated UV laser (8.5 kHz) was used to measure the lifetimes of the fluorescence lines, and yields insight into energy transfer and excitation lifetimes within the system. This technique is especially useful when a high power laser is not either available or not suitable. In the latter case, this would be when a high power pulsed deep-UV laser could produce bleaching or destruction of the biological specimen. In addition, this technique simulated the excitation and fluorescence emission of the DPA-Tb using a 4-level energy model, and solved the dynamic transient rate equations to predict the temporal behavior of the DPA-Tb emitted fluorescence. Excellent agreement between the experiments and the simulation were found. This technique has the potential to provide a more accurate value for the fluorescence lifetime values. In addition, with the use of asymmetric excitation waveforms, the dynamic transient rate equation analysis may allow for detailed studies of selected transfer mechanisms in a wide range of other spectroscopic applications including rare-earth solid-state lasing materials and biological samples.
