This book was released on 1995-09-07 with total page 278 pages. Available in PDF, EPUB and Kindle. Book excerpt: Originally published in 1995, this volume looks at the functioning of the vascular endothelium, and the diseases and tissue injury that arise as a result of inflammation and immunological responses. The vascular endothelium is a metabolically highly active layer of cells lining all blood vessel walls. Through its interactions with leucocytes and other mediators, it is central to the development of inflammatory foci and to lymphocyte trafficking around the body. Tissue injury may arise here as a result of abnormal inflammatory or immune responses. The potential for such injury to contribute to autoimmune disease is discussed in this book, particularly in relation to autoimmune vascular disease of the renal, rheumatological and neurological systems, as well as in organ transplantation.

Available in PDF, EPUB and Kindle. Book excerpt: The endothelium, a monolayer of endothelial cells, constitutes the inner cellular lining of the blood vessels (arteries, veins and capillaries) and the lymphatic system, and therefore is in direct contact with the blood/lymph and the circulating cells. The endothelium is a major player in the control of blood fluidity, platelet aggregation and vascular tone, a major actor in the regulation of immunology, inflammation and angiogenesis, and an important metabolizing and an endocrine organ. Endothelial cells controls vascular tone, and thereby blood flow, by synthesizing and releasing relaxing and contracting factors such as nitric oxide, metabolites of arachidonic acid via the cyclooxygenases, lipoxygenases and cytochrome P450 pathways, various peptides (endothelin, urotensin, CNP, adrenomedullin, etc.), adenosine, purines, reactive oxygen species and so on. Additionally, endothelial ectoenzymes are required steps in the generation of vasoactive hormones such as angiotensin II. An endothelial dysfunction linked to an imbalance in the synthesis and/or the release of these various endothelial factors may explain the initiation of cardiovascular pathologies (from hypertension to atherosclerosis) or their development and perpetuation. Table of Contents: Introduction / Multiple Functions of the Endothelial Cells / Calcium Signaling in Vascular Cells and Cell-to-Cell Communications / Endothelium-Dependent Regulation of Vascular Tone / Conclusion / References

Available in PDF, EPUB and Kindle. Book excerpt: This book focusses on a family of growth factors and their receptors, known as vascular endothelial growth factors (VEGF's). VEGF's are critical for endothelial cell function, both during physiological and pathological conditions. Thus, tumors depend on VEGF in order to promote ingrowth of blood vessels. Without VEGF, blood vessels will not form. There is growing awareness that deregulated production or action of VEGF is initiating or driving a large number of diseases. The book is written by the leading scientists in the field for an audience with medical or life science background, and presents an overview of the current knowledge in the area of VEGF and VEDG receptor structure and function in a series of reviews spanning from molecular signal transduction and cellular biology to the clinic.

Available in PDF, EPUB and Kindle. Book excerpt: Cardiovascular immunology is a newly emerging research area, investigating the crosstalk between the cardiovascular and the immune system. This crosstalk is evident through (1) crucial immunological capacities and functions of cardiovascular cell types, including cardiomyocytes, fibroblasts, endothelial cells, pericytes and cardiac resident macrophages, (2) the impact of aberrant immune function on the development of cardiovascular disease such as atherosclerosis, direct and indirect immune-mediated heart disease and vasculitis, and (3) the crucial role of the immune system in cardiac repair and regeneration. The Immunology of Cardiovascular Homeostasis and Pathology covers all these aspects of cardiovascular immunology, starting with homeostatic immunological functions of traditional cardiovascular cell types, and moving then to the role of the immune system in cardiovascular pathology and to recent research into targeting the immune system to boost cardiac healing and regeneration.

Endothelial dysfunction has been recognized as an early step in the development of cardiovascular diseases: respectively, endothelium represents a potential therapeutic niche with multiple targets. The purpose of the book is to point out some recent findings of endothelial physiology and pathophysiology emphasizing various aspects of endothelial dysfunction connected to the body's internal and external environment. While basic features of the endothelium are presented in an introductory chapter, the authors of the following 17 chapters have provided extensive insight into some selected topics of endothelial (dys)function. The book would hopefully be useful for anyone interested in recapitulating endothelial (patho)physiology and expanding knowledge of molecular mechanisms involved in endothelial dysfunction, relevant also for further clinical investigations.

Y.Calne Surgeons are transplanting kidneys in ever increasing numbers-more than 10000 renal allografts have now been reported to the Transplant Registry. With related donors 75% of grafted kidneys continued to function after 2 years, compared with 50% when the donors were unrelated. The therapeutic value is obvious, but the management is largely empirical and results have improved little in the past 5 years. The basic sciences related to tissue transplantation have advanced rapidly. New serological and tissue culture techniques and chemical analysis of antigens and anti bodies have produced complicated data that is almost incomprehensible to the non specialist. Mathematical treatment of genetic probabilities and of immunological kinetics are similarly difficult to follow for those not especially trained. There has always been a gulf between the practical clinician whose patients do not behave like inbred rodents and the biologist who likes carefully controlled experiments with easily observed results. Both realize, however, that predictable and safe control of rejection must involve close collaboration and co-operation between the laboratory and the clinic. Unfortwlately, the different nature of the work and the workers has widened the gap between them. The clinicians tend to improve their techniques and patient care, whilst the biologists seek clearer and more precisely deflned experi ments which lead them to use increasingly artiflcial experimental models.
