Book excerpt: This thesis tests the hypothesis that innate immunity may be enhanced immediately following a stressful event. The experiments characterize the acute effects of the fight or flight response on some immunological and endocrine parameters in rainbow trout (Oncorhynchus mykiss). Plasma cortisol and catecholamines were elevated within seconds of the initiation of an acute handling stressor consisting of 30 seconds in the air and five, 10 or 20 minutes in a shallow bucket of water. Plasma lysozyme activity increased after stress, however, the increases were not statistically significant unless variation was reduced by serial bleeding of the same individual trout before and after stress. A more "resting" fish was achieved by use of the anesthetic 2-phenoxy-ethanol which was surreptitiously introduced into the tanks before the initial bleed. Individual fish were then revived in freshwater and stressed as before. Enhancement of lysozyme activity was evident although levels of plasma stress hormones in fish that were anesthetized, revived and stressed were less than when fish were similarly stressed without anesthetic. Levels of cortisol and catecholamines increased within seconds of capture and aerial exposure, returned to near pre-stress levels after the fish had been placed in a shallow bucket of water for 30 seconds, then increased again. Evaluation of the influence of acute stress on survival following challenge with the pathogen Vibrio anguillarum yielded equivocal data. Results presented here suggest that enhancement of innate defenses as part of the fight or flight response merits further evaluation.

In offshore oil and gas operations, produced water (the water produced with oil or gas from a well) accounts for the largest waste stream (in terms of volume discharged). Its discharge is continuous during oil and gas production and typically increases in volume over the lifetime of an offshore production platform. Produced water discharge as waste into the ocean has become an environmental concern because of its potential contaminant content. Environmental risk assessments of ocean discharge of produced water have yielded different results. For example, several laboratory and field studies have shown that significant acute toxic effects cannot be detected beyond the "point of discharge" due to rapid dilution in the receiving waters. However, there is some preliminary evidence of chronic sub-lethal impacts in biota associated with the discharge of produced water from oil and gas fields within the North Sea. As the composition and concentration of potential produced water contaminants may vary from one geologic formation to another, this conference also highlights the results of recent studies in Atlantic Canada.

In this Research Topic we will focus on relevant aspects related to fish response to both biotic and abiotic stressors as a means of improving their welfare, integrating both physiological and behavioral responses. In fish the adaptive responses to rearing may differ depending on several factors, including the characteristics of the stimuli and the intrinsic properties of the species or the selected line. If complete adaptation is missing, the application of single or multiple stimuli may result in a stress response. Stress responses have been linked to systemic adjustments which negatively impacts the immune system of fish, as well as their growth and reproductive performance. Therefore, one of the main interests of this Topic is to understand the allostatic responses of aquatic organisms to several stimuli/ factors that may be present during rearing. An additional focus of this Topic will be on conditions that may improve fish welfare by decreasing the stress response, such as implementing adequate swimming conditions. The focus of the Topic is to contribute to our present knowledge to better understand the physiological response of aquatic organisms, which is central to improving aquaculture practices. Emphasis will be placed on potentially existing interactions between physiological and endocrine pathways to build an overall stress response. Another aspect that will be considered in this Topic is how intraspecific variability may affect the stress response.

Although these have been important tools for exploring the complexities of the immune responses of fish species, the next generation of knowledge requires the use of new tools and methods capable of unveiling the diversity of immune cells and molecules in jawed fish, the network of interactions and responses, and the mechanisms leading to immune protection against pathogens. For more than 10 years, many laboratories have been engaged in developing antibodies against key molecules of jawed fish immune systems in several different species because the lack of sequence conservation of most immune genes makes it impossible to use the large panel of antibodies against mammalian molecules for recognition of fish molecules. The numerous genes encoding cytokines and receptors have now been used to make recombinant proteins available, allowing studies of the complex network of cytokine and receptor functions central to fish immune responses. From many laboratories, new and interesting knowledge about immune cells, their functions and interactions can be studied with the availability of new tools and methods have begun to provide a clear understanding fish immunity at the protein and cellular levels. The evidence is divided into three categories: - Anatomical, cellular and molecular bases for the exchange of information between immune, endocrine and neural cells, - reciprocal effects between immune and neuroendocrine mechanisms, and - immune-neuroendocrine regulatory circuits. Immunologically triggered neuroendocrine responses can be either beneficial or deleterious for the host.
