Book excerpt: Oil palm trunk (OPT) is generated from the replantation of oil palm trees at every 25-30 years interval, left as troublesome waste as it becomes the source of infection to young oil palm trees. This OPT contains a high amount of ready-to-use sugar in the form of sap which can be directly fermented to the most fermentation products. The fermentation process for ethanol production from OPT sap was evaluated in order to produce larger amount of bioethanol. The feasibility of yeast S. cerevisiae to produce bioethanol from OPT sap was investigated along with the effect of different strains of S. cerevisiae and different pretreated mediums. It was proven that yeast S.cerevisiae was able to produce bioethanol even though the OPT sap undergoes less pretreatment compared to the previous works done by the other reseachers. The highest bioethanol yield and productivity had been obtained by using S. cerevisiae Kyokai no.7 in heat sterilized sap. The effects of temperature, initial pH, agitation rate, percentage inoculum and time of incubation were explored using 2-level full factorial design in order to find out the main factor that affecting bioethanol fermentation from the OPT sap. It aims to develop alternative resources waste to wealth for bio-ethanol production using available biomass in this country. To produce high production of bioethanol, determine fermentation conditions are important in culture fermentation to predict the optimum temperature and optimum inoculums size. The process selection includes preparation of pure culture, yeast activation, fermentation profile, and validation between two types of yeast, batch fermentation and analysis of the data. The overall process performance is measured by the productivity and quality of bioethanol produced. Glucose was thoroughly nearly consumed after 48 hour for the fermentation profile. Validation experiment between showed that Saccharomyces cerevisiae Kyokai 7 produce high yield of ethanol. The effects of temperature (25- 40 oC),and percentage inoculums (5- 15 %v/v) on ethanol yield were assessed by using 24 full factorial design (FFD) and validated statistically by analysis of variance (ANOVA). The optimum temperature is 25.29 celcius and optimum inoculum size is 10.54 % .Saccharomyces cerevisiae Kyokai 7 produced 41.43% higher ethanol compared to S.cerevisiae. The mean percentage error is 10.32 in screening parameter experiment. The influence of temperature was found to be more pronounced on ethanol yield compare to others parameter.

This study is focus on studying the kinetic parameters of bioethanol production from oil palm trunk sap (OPTS) using Kluyveromyces marxianus. By analyzing specific growth rate, sugar consumption rate and ethanol production rate the kinetic parameters can be determined. The analytical techniques for bioethanol production and substrate consumption were monitored by high-performance liquid chromatography (HPLC) and cell dry weight (CDW) for growth profile. In this study, pH and temperature were varied to study the effect of different pH (3, 4, 5, 6, and 7) and temperature (25oC, 30oC, 35oC, 40oC, and 45oC). The highest ethanol concentration was produced at pH 5 which is 26.75 g/L and at temperature 35oC ethanol concentration production was 45.06 g/L.

Due to limited stock of fossil fuel and rapid increase of energy demand in Malaysia, bioethanol has become a very important and necessary alternative energy source in order to replace the limited fossil fuel stock. Hence, this paper presents the study on bioethanol production by using oil palm trunk (OPT) sap which mainly focusing on substrate concentration effect toward the yield, productivity rate, and kinetic behaviour of cell growth in different substrate concentration This research was carried out by using Saccharomyces cerevisiae Kyokai No.7 in 200mL conical flask, under 30oC anaerobic condition. The research are focusing on the inoculums culture preparation, fermentation process, sample preparation, and analyzing methods. It was found that bioethanol production is increase when substrate concentration increase, from 14g/L in 60% substrate concentration, followed by 18g/L in 80% OPT sap to 22g/L in 100% OPT sap. With the highest sugar content in the medium, S. cerevisiae can produce faster ethanol production within 30 hours of fermentation duration. However, high bioethanol concentration in the process will cause the cell growth rate falls due to the product inhibition. Besides, glucose consumption rate were found out that inversely proportional to the substrate concentration whereas ethanol production rate and cell growth rate were directly proportional to the substrate concentration. Moreover, specific bioethanol production yield was found out that exceed 0.482, 0.516, and 0.465 for 60%, 80%, and 100% of substrate concentration respectively. These results will be useful in the future scale up processes.

This design of experiment is conducted to screen out the significant parameters that affecting the bioethanol production. The parameters that are being screened are the temperature, pH, inoculum percentage and agitation. The microbe involved is Saccharomylesn Cerevisiae Kyokai 7 since it accumulates bioethanol more than any other type of microbes. The Saccharomylesn Cerevisiae Kyokai 7 is cultured in the inoculums preparation and being fermented in the oil palm trunk sap for 48 hours. By using the 2-Level Full Factorial Design and Response Surface Methodology, the optimal parameters levels are determined. The best range value for temperature is in the range of 25oC, pH around 3, agitation speed in the range 250 rpm and inoculums percentage around 10 % for the maximum concentration of bioethanol.

Bioethanol has been gaining much interest recently in terms of research and development. Since there are various factors such as rising oil price, environmental issues and high rate consumption of fossil fuel, the global demand for bioethanol has shown a remarkable increase. Oil palm frond is known to be the most generated oil palm biomass annually. OPF is obtained during pruning for harvesting fresh fruit bunch, and the juice can be produced by pressing the fresh OPF using the conventional sugarcane pressing machine. The OPF juice contains higher glucose content, which is about 70% of the total free sugar. Hence, it has a high potential to be the carbon source for producing bioethanol. The parameters investigated in this research work are temperature, pH and agitation speed. Bioethanol was produced from oil palm frond juice by fermentation with Saccharomyces cerevisiae. The ethanol concentration from the fermentation sample was analysed using HPLC. Using the ethanol concentration obtained from HPLC analysis, the optimum condition for the production of bioethanol was determined using the Response Surface Methodology with the aid of Design Expert software. The results revealed that, a maximum bioethanol concentration of 22.93 g/L can be obtained at the following optimum conditions : temperature of 32oC, pH 6 and agitation speed of 80 rpm. The validation result revealed that at this optimum condition, a bioethanol yield of 30.1% , which is close to the predicted value, can be obtained.

Waste from palm oil production factories has been increasing rapidly and one of the serious problems in the palm oil fruit processing is managing the waste generated by the processes. Recent studies showed that butanol can be used as excellent renewable liquid transportation fuel. In this study, oil palm trunk sap is used as the substrate. The objective of this study is to compare the production of biobutanol from biomass waste (oil palm trunk sap) via fermentation process using different microorganisms. The fermentation type done was submerged fermentation. Fermentation was conducted for five days at 37 0C. Samples were taken for every 6 hours for 3 days, then for every 12 hours. Samples taken were analyzed using gas chromatography-flame ionization detector (GC-FID) and high performance liquid chromatography (HPLC) was used to analyze sugar composition in OPT sap. UV Spectrophotometer is used to analyze biomass. Oil palm trunk (OPT) sap contains sucrose as its dominant sugar followed by glucose, fructose and galactose. Based on the analysis, acetone, butanol and ethanol were produced. Comparisons between the two strains Clostridium acetobutylicum and Clostridium beijerinckii were made, and C. beijerinckii showed higher production of acetone, butanol and ethanol. The highest yield for butanol production by C. acetobutylicum and C. beijerinckii were 13.910 g/L and 15.301 g/L and highest production of ethanol were 11.17 g/L and 12.09 g/L. 4.13 g/L and 4.49 g/L acetone was produced by C. acetobutylicum and C. beijerinckii. Based on this study, it is concluded that OPT sap is preferable substrate to produce butanol by using C. beijerinckii.

In order to improve the current production, the study on locally isolated strains to produce biobutanol by using local plant is proposed. The strain is isolated from soil sample taken at oil palm plantation before proceed with the fermentation process to produce biobutanol. The isolation process is done under strict anaerobic condition since the potential butanol producer strain generally is an anaerobic bacteria. For the fermentation process oil palm trunk sap is used as the carbon sources to produce the biobutanol at 37oC for 5 days. From this research, there are three isolated bacteria which are; isolate 1a, isolate 2b and isolate 3b, which had been identified, can produce biobutanol and other biosolvents. Among the three of the isolated bacteria, isolate 2b was identified to be the best isolated strain in production of butanol and other biosolvents with 5.41 g/L of butanol, 15.04 g/L of acetone and 5.90 g/L of ethanol.

As for this research, a local type of fungi, Pleurotus ostreatus or commercially known as 'Oyster Mushrooms', were used as an agent of delignification and the raw material chosen are oil palm trunk wastes .In order to optimize conditions for production of lignocellulolytic enzymes by the oyster mushroom, a Response Surface Methodology (RSM) based experiment was designed by using Design Expert software. In this research, 30 runs of experiment were done. After obtaining the experimental design table, the experimental procedures were preceded and the values of the parameters were set according to the table at respective value. After that, the lignin content was analyzed by using Klason Lignin Method. With this method, the lignin content can be determined for various values of parameters. Then, the results obtained were analyzed again by using Design Expert software. At the end of this research, the optimum conditions obtained for the lignin degradation of oil palm trunk fiber are with temperature of 25 °C, pH value of 7.0, moisture content of 0.21 ml and fungi to medium ratio of 2.5 with the amount of predicted lignin degraded is 15.37%. Finally, the conformational test was done with the optimum conditions obtained and the error between the actual and predicted value is 18.7%. However, for better results, the center point in RSM should be tested before designing the experiment.

Bioethanol is a bioenergy produced by yeast fermentation at several conditions compared to conventional fossil fuels. Bioethanol is being studied because the unique properties and renewable fuels. The production of bioethanol in industrial scale involved high cost due to the usage of foods as sugar raw materials. Thus, this research is to evaluate the oil palm frond (OPF) juice as raw material for bioethanol fermentation. In this study, the bioethanol produces in shake flask by using Saccharomyces cerevisiae and OPF juice as medium component. Three parameters study are initial pH of medium, agitation speed, and OPF juice concentration. The best condition of parameters determine by the highest ethanol concentration produce. Inoculum prepared in shake flask to cultivate the yeast in medium growth and ferment in 250 ml shake flasks containing medium growth and beef extract. The flasks incubate in 48 hours, 12 hours interval for sample withdraw. The products analyse for sugar concentration and bioethanol concentration. The expected results for the best condition in bioethanol fermentation are in pH 7, 150 rpm agitation speed and 100% volume percentage of OPF juice concentration.
