Available in PDF, EPUB and Kindle. Book excerpt: The growth and function of many tissues are influenced by hormones. Therefore it is quite understandable that hormones play a role in the development and treatment of malignant tumors. Numerous publications address this topic; however, the results of many studies are controversial and have not been unequivocally accepted. For nearly 50 years the carcinogenic effect of steroid hormones has been under debate, and their therapeutic value a matter of discussion for equally as long. The present volume concentrates on substantiated data first obtained from the study of tumors developing from hormone regulated or hormone-producing tissue, e.g., the thyroid, adrenal glands, prostate, and the female genital tract. Through a joint approach from the field of molecular biology, biochemistry, and histopathology, advances in the management of these tumors have been elaborated. Another exciting example is the endo nuclear diagnosis of adrenal tumors. Antihormones, i.e. antian drogens or GnRH analogues have proved to be important indeed since they exhibit a destructive effect on prostate carcinomas and breast cancer. Further improvements can be expected in the localization of hormones in tumor tissue by specific antibodies. A special chapter is dedicated to the diffuse endocrine cell system (DECS), the clinical significance of which has mainly become obvious in the gastroenteropancreatic tract.

Available in PDF, EPUB and Kindle. Book excerpt: Since our previous symposium in 1995, the pace of research in hormones and cancer has accelerated. Progress in our understanding of hormonal carcinogenic processes has been a direct result of the advances made in cell biology, endocrinology, and carcinogenesis at the molecular level. The newer fields of molecular genetics and cytogenetics already have and are expected to continue to playa major role in furthering our understanding of the cellular and molecular events in hormonal carcinogenesis. It has become increasingly clear that the risk of naturally occurring sex hormones in carcinogenic processes, both in human and in animal models, requires only minute quantities of hormones, at both the serum and tissue levels. Moreover, hormone target tissues for neoplastic transformation, perhaps with the exception of the liver, generally have relatively modest ability to metabolize sex hormones, such as the breast and prostate. Table 1 summarizes the serum, and in most cases, the tissue levels of sex hormones, both endogenously and exogenously ingested, which are associated with increased risk for endocrine-associated cancers such as breast, endometrium, and prostate, as well as the hormone levels of four experimental models that have been shown to elicit high tumor incidences. In contrast to the human, in which the hormone levels are cyclic, however, the latter require continuous hormone exposure at these relatively low levels.

Available in PDF, EPUB and Kindle. Book excerpt: The clinical benefits of hormone replacement therapy in women have to be carefully balanced against the possible risks, and a particular theoretical concern relates to risks associated with various forms of female oncology. Because of conflicting reports, gynecologists and oncologists especially need a single, authoritative resource of up-to-date information. Hormone Replacement Therapy and Cancer, published in association with the International Menopause Society, provides the very consensus statement that clinicians need in this difficult and complex area. Many of the world's leading specialists have contributed important chapters that provide state-of-the-art knowledge about the effects of hormones on women and possible cancer risks. The introductory section deals with carcinogenesis, and the other main sections cover HRT and breast cancer, endometrial cancer, colon cancer, melanoma and epithelial ovarian cancer. The concluding chapters discuss the benefits and risks of sp ecific therapies. An authoritative clinical reference with extensive bibliographic references and index, Hormone Replacement Therapy and Cancer covers all aspects of HRT and cancer based on the research available up to June 2001.

Available in PDF, EPUB and Kindle. Book excerpt: The link between the pineal gland and cancer is a rapidly emerging research field due to promising experimental and clinical trials with melatonin. The pineal gland acts as a transducer of environmental light to regulate rhythmic processes, including reproductive function in seasonally breeding animals and the entrainment of circadian rhythms, such as the sleep-wake cycle, in man. This book elucidates the physiological significance of the pineal gland and surveys phenomena and mechanisms of pineal - tumor interaction at the neuroendocrine, neuroimmune, neural, and molecular levels. Yet unidentified low-molecular-weight pineal substances with tumor-inhibiting capacity, a possible involvement of melatonin in electromagnetic field effects on cancer, and the oncotherapeutic potential of melatonin are also addressed. The encouraging results should incite further research to elucidate the exact nature of the link between the pineal gland and cancer for the benefit of patients.

Available in PDF, EPUB and Kindle. Book excerpt: It has been over a decade since the First International Symposium on Hormonal Carcinogenesis convened in 199 1. Since then, the field has rapidly expanded with considerable progress in both breast and prostate cancers; while ovarian and endometrial cancer have been hampered, in part, due to the absence of suitable hormone-mediated animal models. While knock-out, transgenic, and cell-culture systems have been extremely useful in identifying specific genelprotein alterations and the ensuing pathways affected, the precise molecular mechanisms whereby sex hormones elicit their oncogenic effects still remain elusive. Moreover, despite the considerable progress made in breast cancer research, the exact role of progestins in the presence or absence of estrogen in breast growth, differentiation, and malignant transformation is lacking. Elucidating the incipient molecular alterations in earlylpre-invasive lesions elicited by these hormones is a growing important focus of this field. The main purpose of these Symposia has been to address vital questions that impact our understanding of the causation, dependency, progression, resistance, and prevention of hormonally-associated cancers. We are indebted to the Scientific Advisory Board members who worked with us reviewing and offering suggestions to finalize the scientific program. We offer special thanks for the guidance and support of Dr. Gerald Mueller. His wisdom played an indispensable role in maintaining the excellence of these Symposia. We also acknowledge the numerous external reviewers that worked diligently to revise and improve the quality of the manuscripts. We are very grateful to Ms. Tandria Price.

Book excerpt: There is little doubt that quality of life can be substantially improved by hormone replacement therapy (HRT). Evidence has also accumulated concerning the effects of estrogen in the primary prevention of cardiovascular diseases and osteoporosis. However, recent studies have cast some doubts about the effectiveness of HRT in the secondary prevention of cardiovascular diseases as well as highlighting the possibility of undesirable effects in long-term use. A comprehensive clinical reference, Menopause: Hormones and Cancer examines the issues involved in the use of HRT. It presents new information on steroid receptors, estrogens and estrogen therapy, oral contraception, hormone therapies for ovarian cancer, progesterone therapy, endometrial cancer, phytoestrogens, hormone replacement therapy, estrogen receptors, neurosteroids, prophylactic oophorectomy and estrogen or testosterone implants, and the relationship between sex steroid therapy and colon cancer. The use of HRT is still relatively new and must be studied for many years before the alleged beneficial preventive effects can be seen and understood. In addition, long-term treatment, other than having the desired positive effects, also has some possible undesirable side effects that require further study. In Menopause: Hormones and Cancer top experts in the field focus on understanding the cardiovascular benefits of HRT and the long-term risks.

Available in PDF, EPUB and Kindle. Book excerpt: Sex steroids, including androgens, estrogens, and progestogens, are known to have widespread physiological actions beyond the reproductive system via binding to the sex hormone receptors. Meanwhile, emerging evidence has indicated that sex hormone receptor signals are involved in the outgrowth of some malignancies, such as prostate and breast carcinomas, as well as others that have not traditionally been considered as endocrine-related neoplasms. This Special Issue “Sex Hormone Receptor Signals in Human Malignancies” covers various aspects of the potential role of sex hormone receptors and related signals in prostate cancer, breast cancer, and other neoplastic conditions by depicting promising findings derived from in vitro and in vivo experiments as well as the analyses of surgical specimens. The current observations described may thus provide a unique insight into novel or known functions of sex hormone receptors and related molecules.
