The genus Brassica belongs to the mustard family whose members are known as cruciferous vegetables, cabbages or mustard plants. Rapeseed-mustard is world’s third most important source of edible oil after soybean and oil palm. It has worldwide acceptance owing to its rare combination of health promoting factors. It has very low levels of saturated fatty acids which make it the healthiest edible oil that is commonly available. Apart from this, it is rich in antioxidants by virtue of tocopherols and phytosterols presence in the oil. The high omega 3 content reduces the risk of atherosclerosis/heart attack. Conventional breeding methods have met with limited success in Brassica because yield and stress resilience are polygenic traits and are greatly influenced by environment. Therefore, it is imperative to accelerate the efforts to unravel the biochemical, physiological and molecular mechanisms underlying yield, quality and tolerance towards biotic and abiotic stresses in Brassica. To exploit its fullest potential, systematic efforts are needed to unlock the genetic information for new germplasms that tolerate initial and terminal state heat coupled with moisture stress. For instance, wild relatives may be exploited in developing introgressed and resynthesized lines with desirable attributes. Exploitation of heterosis is another important area which can be achieved by introducing transgenics to raise stable CMS lines. Doubled haploid breeding and marker assisted selection should be employed along with conventional breeding. Breeding programmes aim at enhancing resource use efficiency, especially nutrient and water as well as adoption to aberrant environmental changes should also be considered. Biotechnological interventions are essential for altering the biosynthetic pathways for developing high oleic and low linolenic lines. Accordingly, tools such as microspore and ovule culture, embryo rescue, isolation of trait specific genes especially for aphid, Sclerotinia and alternaria blight resistance, etc. along with identification of potential lines based on genetic diversity can assist ongoing breeding programmes. In this book, we highlight the recent molecular, genetic and genomic interventions made to achieve crop improvement in terms of yield increase, quality and stress tolerance in Brassica, with a special emphasis in Rapeseed-mustard.

Book excerpt: Brassica napus L. from Brassicaceae consisting AACC genome is one of the most important oil crops that contributes to the Canadian economy. The oil produced by B. napus can be made into high profit oil-related products such as cooking oil and machinery lubricant, and it can be classified as either rapeseed or canola depending on the amount of erucic acid and glucosinolates in the seed. Numerous studies on mapping quantitative trait loci (QTL) associated with the seed oil content (SOC) in B. napus have been conducted. Studies have identifed the location of oil-related genes such as BnFAE genes located on chromosomes A8 and C3 which are responsible for the production of erucic acid in the seed. One doubled haploid (DH) population ZT (Topas x Zhongyou821) consisting of 94 DH genotypes was used to collect SOC data in field experiments and under controlled environmental conditions in the greenhouse. A new molecular marker detection method that includes PCR-based sequencing library preparation and Illumina sequencing detection (PISD) was used to identify single nucleotide polymorphisms (SNPs). Two genetic maps, named PISD and combined (standard genotyping-by-sequencing, GBS), were created and 1,092 and 1,821 SNPs molecular markers were used to identify QTL for SOC in the B. napus DH ZT genotypes. In total, six and 8 major QTL were identified using the two genetic maps respectively. In addition, QTL for SOC on chromosomes A8 and C3 were identified as being closely linked to BnFAE1.1 and BnFAE1.2, which are responsible for the production of erucic acid. These results show a better understanding of gene control and can be useful for B. napus breeding industries for erucic acid content in the future.

Book excerpt: Clubroot (Plasmodiophora brassicae) is an important disease of canola/rapeseed (Brassica napus) and other crucifers. In this study, resistance loci/genes effective against P. brassicae pathotypes 2B, 3A, 3D, 3H, 5C, 5X and 8J were identified and mapped via genotyping-by-sequencing, QTL analysis, conventional linkage mapping, and by exploring the effect of individual genes on quantitative traits in combination with marker and trait data. Four of 34 B. napus accessions, CGN06902, CGN17369, AAFC695, and CN46235, were resistant to pathotypes 3A, 2B, and 3D, while 16 B. rapa families did not show any resistance. The resistant accession AAFC695, B. napus 'Mendel' (resistant to 13 of 17 pathotypes identified on the Canadian Clubroot Differential set), and B. rapa line ECD02 (resistant to all 17 pathotypes) were used to map resistance genes by developing a BC1S1 population based on ECD02 and doubled haploid (DH) populations based on 'AAFC695' and 'Mendel'. A single QTL Rcr9ECD02 was identified on chromosome A08 of ECD02 using 93 BC1S1 lines; this QTL conferred resistance to pathotypes 3A, 3D, 3H, and 5X, accounting for 68.9-74.4% phenotypic variation explained (PVE). Bulked segregant and KASP analysis confirmed the QTL. In 'AAFC695', a QTL Rcr9AAFC695 was identified on chromosome A08 using 102 DH lines; Rcr9AAFC695 explained 92.6-95.0% PVE in response to pathotypes 3A, 3D, 3H, and 5X, with LOD values of 60.3 to 70.8. In 'Mendel', the QTL Rcr3Mendel was mapped on chromosome A08 using 137 DH lines; this was a major locus explaining 88-90% PVE for resistance to pathotypes 3D, 5C, and 8J, with an additive effect of 47.2-47.3%. Testing of 60 lines from this population against pathotype 3H indicated that Rcr3Mendel is the Rcr3 locus, but that it also bestows resistance to pathotypes 3D, 5C, and 8J. The identification of TIRNBSLRR (TNL) genes (via analysis of gene annotation with Blast2GO and gene ontology information) in the QTL regions of the three populations confirmed that the identified QTL Rcr9ECD02, Rcr9AAFC695 and Rcr3Mendel are different from each other, and distinct from Rcr9. The CR loci/genes identified in this study will enrich the repository of resistance genes available to protect canola/rapeseed from clubroot disease.
