Book excerpt: Soybean (Glycine max L.) seed yield, composition, and quality can be negatively affected by environmental stresses during the growing season and harvest. The central hypothesis of this thesis is that soybean seed testing techniques, i.e. testing media, need to be adjusted for seed moisture content, production environment, and seed composition, as these factors alter the measured germination in laboratory tests. Water and nutrient deficit during pod filling usually lead to poor yields by reducing seed set, increasing seed abortion, and reducing seed size. While maturation drying and the acquisition of desiccation tolerance are critical physiological processes that help orthodox seeds (seeds from species that can be stored at low moisture content) survive periods of unfavorable conditions for germination, the warm and dry conditions during harvest can often over-dry seeds. Extremely dry seeds experience stresses during imbibition which are likely to result in imbibitional damage and seedling abnormalities. The production environment can also lead to composition differences within seeds and may play important roles in imbibitional injury. Being able to accurately evaluate seed germination will be crucial regardless of seed composition, production environment, and, inherent genetic differences. Seed analysts must consider soybean seed composition and seed moisture, and, their interaction with testing media to avoid inconsistent test results within and among seed testing laboratories.

The nutritional quality of these seed components depends upon the relative abundance of specific proteins and fatty acids. Additionally, secondary metabolites such as isoflavones, which are present in soybeans, have been shown to impact human health. Genetics, environmental conditions, and agronomic practices have a bearing on accumulation of each of these seed components. Work presented here reveals that the constituents of the protein and oil components vary with the nodal position of seed development. Sodium dodecyl sulfate polyacrylamide gel electrophoresis provided evidence that proteins rich in the sulfur amino acids accumulate preferentially in seed from the basal nodes while proteins poor in these amino acids are found in the apical nodes. Fatty acid content determined by gas chromatography showed a nodal dependent difference in accumulation of monounsaturated and polyunsaturated fatty acids but no difference in that of the saturated fatty acids. A long-term crop rotation study revealed that environmental factors and putative changes in soil ecology could affect seed protein and oil content. Protein content notably increased with a concomitant decrease in oil over the 11-year span of the study. Using one- and two- dimensional gel electrophoresis and gas chromatography, we determined that the protein and fatty acid profiles respectively, of soybeans cultivated in an early planting system were comparable to that of a traditionally cultivated crop. Although irrigation did not improve the protein and oil accumulation, it did elicit a dramatic increase in the isoflavone content of the seed. Continued research devoted to the elucidation of soybean genetics, physiology, and biochemistry is crucial for breeding and development of this vital food crop.

In this study we tested the germination of TM-Miniature, a soybean mutant with dwarf phenotype, and TM-Normal and two other genotypes in response to high temperatures of 38 °C, 40 °C, and a control temperature of 28 °C. The results showed that a mutation might cause TM-Miniature seedlings to be more thermotolerant than TM-Normal and the two other genotypes at emergence. We also examined the morphology and anatomy of TM-Miniature and TM-Normal in response to a range of temperature and photoperiods conditions. The data indicated clearly that TM-Miniature phenotype was influenced by night temperature and light interruption of the dark period. Leaves of TM-Miniature plants were very small at low night temperature while the leaf thickness remained unchanged. The upper and lower epidermal cell layers decreased in thickness as nighttime temperature increased. In addition, stomatal density and stomatal index decreased in TM-Miniature. TM-Miniature is controlled by a single mutant gene. Glyma14g36890 is an interesting candidate gene and was found near the Satt560 marker that co-segregated with the miniature trait. This gene was amplified and sequenced to study nucleotide differences between TM-Normal and William 82. The sequenced armadillo gene in our study did not show nucleotide differences in the predicted promoter region and exon gene region. However, there appear to be sequence differences in a predicted intron.
