Book excerpt: This volume contains the proceedings of the symposium on "Ganglioside Structure, Function and Biomedical Potential" which was held at Parksville, Vancouver Island, B. C. , Canada on July 6-10, 1983. The symposium was organized ao a satellite to the ninth meeting of the International Society for Neurochemistry, held immediately afterward in Vancouver City, B. C. Close to 50 speakers from 9 countries presented papers on a wide range of topics on the ganglioside theme. These encompassed the many aspects of basic research that have evolved over the past half-century, as well as some newer topics relating to the biomedical potential of gangliosides as therapeutic agents. One of the purposes of the meeting was to encourage dialogue between investigators in these seemingly diverse areas in the hope that each would come away with fresh perspective toward his own work. Judging from the many spirited, informed discussions that took place throughout the four day meeting, this goal was achieved. The charm and beauty of Vancouver Island undoubtedly contributed to the congenial atmosphere which quickly developed among the 120 or so participants. Drawn from 13 countries of Europe (East and West), America (North and South), Asia and the Middle East, this gathering reflected the broad geographical scale on which ganglioside research is now conducted. That this field is still in its "logarithmic" stage of growth is indicated by the increasing number of citations appearing each year in the various abstracts (e. g. fig. next page).

About 70 participants from 10 countries presented papers on a wide range of topics all concerning the ganglioside theme. The content of the workshop consisted of the following chap ters: (1) Fundamentals for research on function of gangliosides; (2) Brain ganglioside metabolism; (3) Cell specific distribution and differentiation-related ex pression of brain gangliosides; (4) Gangliosides and neuronal plasticity; (5) Bio-medical potential of exogenous ganglioside application on neuronal function; and (6) Critical assessments on aspects of ganglioside therapy. The aim of the workshop was to sum up and discuss the present knowledge in the respective fields. Despite intensive research on gangliosides ever since Ernst Klenk discovered them in 1936, the biological function of. these amphiphilic, "sphinx-like" glycosphingo lipids remains elusive. Over the past few years, several thousands of articles on gangliosides have been published demonstrating very special properties of these glycolipids and their possible in volvement in diverse biological phenomena. As far as we know up to the present time, these properties may be due to particular physico-chemical attributes of these compounds, especially in com bination with calcium. The results obtained so far have shed light not only on pos sible functional roles of gangliosides with regard to cellular induction and organization, but have also unexpectedly opened up VI a whole new field of therapeutic applications for these compounds.

It is a sequel to the first conference bearing this title which took place, also in the vicinity of Strasbourg, in March, 1977. In either case the meetings were coorganized by L. Freysz of Strasbourg, France and S. Gatt of Jerusalem, Israel. The present meeting was set up as a joint NATO Advanced Research Workshop and CNRS-INSERM International Symposium. The conference was guided by two principles, namely, that science has no bounderies, neither has the study of lipid metabolism. Participants came from Europe, the USA, Israel and Japan and represented areas of research in lipid metabolism involving fatty acid s, cholesterylesters, glycero-and sphingolipids. The experimental approaches utilized purified enzymes, artificial and biological membranes, as well as a variety of cells, primary or cultured lines. A session was also devoted to modification of lipid enzymes and metabolism resulting from inherited, inborn defects such as the lipid storage diseases which are caused by genetic modification of degradative enzymes of lipid metabolism. A second type of disease stemming from a defect in a cell organnel (ie, the peroxisome) was also discussed. The eight and one and a half years which elapsed since the previous meeting, highlighted the changing emphasis of research in. lipid metabolism.

The complexity of organization, the heteroge neity of cell types and their interactions, and the difficulty of controlling experimental variables in intact organisms make this a formidable task. Because of the ability that it affords to analyze smaller components of the nervous system (even single cells in some cases) and to better control experimental variables, cell culture has become an increasingly valuable tool for neuroscientists. Many aspects of neural development, such as proliferation, differentiation, synaptogenesis, and myelination, occur in culture with time courses remarkably similar to those in vivo. Thus, in vitro methods often provide excellent model systems for investigating neurobiological questions. Ross Harrison described the first culture of neural tissue in 1907 and used morphological methods to analyze the cultures. Since that time the technique has been progressively modified and used to address an ever widening range of developmental questions. In recent years a con vergence of new or improved cell culture, biochemical, electrophysiol ogical, and immunological methods has occurred and been brought to bear on neurobiological questions. This volume is intended not to be comprehensive but rather to highlight some of the latest findings, with a review of previous important work as well, in which combinations of these methods are used.

Book excerpt: Although there are over 400,000 people each year in the United States alone who suffer from traumatic injury to the central nervous system (CNS), no phar macological treatment is currently available. Considering the enormity of the problem in terms of human tragedy as well as the economic burden to families and societies alike, it is surprising that so little effort is being made to develop treatments for these disorders. Although no one can become inured to the victims of brain or spinal cord injuries, one reason that insufficient time and effort have been devoted to research on recovery is that it is a generally held medical belief that nervous system injuries are simply not amenable to treatment. At best, current therapies are aimed at providing symptomatic relief or focus on re habilitative measures and the teaching of alternative behavioral strategies to help patients cope with their impairments, with only marginal results in many cases. Only within the last decade have neuroscientists begun to make serious inroads into understanding and examining the inherent "plasticity" found in the adult CNS. Ten years or so ago, very few researchers or clinicians would have thought that damaged central neurons could sprout new terminals or that intact nerve fibers in a damaged pathway could proliferate to replace inputs from neurons that died as a result of injury.
