Significant yield reductions can result if the plant does not have proper levels of iron to maintain certain cellular functions. Iron deficiency chlorosis (IDC) is a major abiotic stress that affects soybeans grown on high-pH calcareous soils, primarily in the North Central region of the USA (Hansen et al., 2003). The degree of chlorosis depends on many aspects such as soil pH, temperature, and free carbonate levels. This research project studied the genetic control of IDC using conventional breeding and biotechnology methods. Specifically, three studies were conducted to gain a comprehensive view of genetic control of IDC in soybean to enhance development of IDC tolerant soybean cultivars for soybean producers, by i) genomic analysis of a long-term recurrent selection program, ii) mapping of QTLs using a recombinant inbred line population, and iii) phenotypic characterization of the Arabidopsis FRO2 gene on calcareous soils in a soybean background.

Book excerpt: Iron deficiency chlorosis (IDC) is a frequent problem throughout many areas of the United States where soils are high in calcium carbonate (CaCO3), including the Blackland Prairie regions of Mississippi. The main objectives of this study were to 1) determine the effects of seven different cropping systems on IDC visual symptomology and grain yield in rainfed soybeans grown in calcareous soils, and 2) evaluate the effects of soil water tension (SWT) on IDC. Rotating soybeans with corn produced significantly higher yields for both tolerant and susceptible soybean varieties. IDC symptomology was worse, and yields were lower in cropping systems with lower average SWT, or wetter soils, throughout the growing season. Last, this study evaluated the use of multispectral imagery and apparent soil electrical conductivity (ECa) to identify IDC-prone areas of a field for the site-specific implementation of management strategies that produced higher yields in the plot-scale study.

Book excerpt: Iron-Deficiency Chlorosis (IDC) causes yield losses in soybean and other crop species as a result of the plant's inability to efficiently acquire iron from calcareous soils. Although plants differ in their response to iron deficiency and IDC resistance has been improved in soybean, the biology of IDC is not well understood. We undertook an interdisciplinary approach towards increasing our knowledge of IDC resistance and iron acquisition strategies in higher plants. Firstly, we examined the usefulness of molecular markers to improve breeding efficiency for IDC resistance in soybean. After examining Simple Sequence Repeats (SSR) markers genetically linked to previously identified quantitative trait loci for IDC resistance in a soybean breeding population, we determined that SSR marker Satt481 may improve selection efficiency 2.6-fold and reduce the evaluation costs by 70% relative to traditional breeding approaches. Secondly, we investigated the potential relationship of IDC with Soybean Cyst Nematode (SCN) and calcareous soil properties. Using two soybean breeding populations and fourteen soybean genotypes grown on calcareous soils infested with Heterodera glycines and concurrent greenhouse studies, we were not able to establish a consistent relationship between IDC-related chlorosis and SCN. However, an increase in foliar chlorosis in SCN-susceptible genotypes was associated with a decrease in SCN reproduction, and SCN-resistant genotypes were more susceptible to IDC during greenhouse evaluation relative to evaluation on SCN-infested calcareous soils. In addition, calcareous soil properties predicted foliar chlorosis in only one breeding population evaluated on SCN-infested soils during one year. Lastly, we examined the evolution of Strategy I and II mechanisms for iron acquisition in higher plants. The phylogenetic distribution in plants of nine iron acquisition-related genes for both strategies was determined using Expressed Sequence Tags (ESTs) from eleven plant species (one gymnosperm, five dicots, and five grasses). Our results suggested gene conservation across strategies and Strategy II was more recently developed than Strategy I in higher plants. In our studies, we have further demonstrated the complexity of IDC, while increasing knowledge on the the biology of IDC and iron acquisition using an interdisciplinary approach involving methodologies in classical and molecular genetics, plant pathology, and soil science.

There are several limitations that come with trying to grow soybeans in this region, including fertility constraints, moisture stress, and improper use of fertilizers. However, the largest constraint at this time seems to be the presence of micronutrient deficiencies, specifically iron. This thesis has an introduction, and three major chapters. The objective of the first study on agronomics was the evaluation of the effect of Fe fertilizer application using foliar and seed-applied methods in combination with variety selection for Fe deficiency management of soybean grown under irrigated conditions in Western Kansas. The second study uses multivariate analysis as an exploratory tool useful in determining simultaneous observation and analysis of more than one variable in a multidimensional space. Factor analysis is used to find underlying factors that one variable alone cannot measure. The objective of this study was to determine the underlying factors and the multi-linear models that are associated with soil parameters that can create Fe chlorosis in the Great Plains. The third study looked at different application rates of seed-applied Fe fertilizer to try and determine the optimum application rate for application of chelated Fe in Western Kansas.
