C'est une technique largement utilisée pour densifier un mélange pulvérulent et le rendre comprimable ; il s'agit d'un procédé complexe mettant en jeu deux étapes principales, la granulation (en présence d'un liquide de mouillage) puis le séchage, dont la maîtrise est essentielle pour garantir la qualité et la reproductibilité du grain obtenu. La partie bibliographique s'attachera à présenter les différentes méthodes et appareillages permettant l'obtention du grain. La technique de séchage par micro-ondes sera plus détaillée, un paragraphe lui sera consacré. Parmi ces équipements, il existe des mélangeurs-granulateurs-sécheurs (dits MGS), assurant les 2 principales étapes au sein d'une même enceinte. Ce sont ces types d'appareils, les Turbosphères® que nous retrouvons sur le site de Gidy des Laboratoires SERVIER pour la fabrication du Daflon®500mg. La partie pratique de ce travail s'intéressera à l'étude préalable de mise en place du transfert de production d'un MGS avec séchage micro-ondes vers un MGS non équipé, où le séchage se fera de façon traditionnelle (double enveloppe). L'impact de ce changement de méthodologie sur le grain sera plus particulièrement étudié.

Ainsi, l’application d’une approche PAT faisant intervenir du proche infrarouge en cours de mélange a permis l’amélioration des procédés de fabrication par optimisation des opérations de pesées, de chargement et de mélange. Pour ce faire, un processus de validation a été déroulé, de l’initiation d’une demande de changement à la clôture de celle-ci, en passant par la réalisation d’essais sur un procédé validé, d’une étude de faisabilité, d’une analyse de risque et d’une validation à proprement parler aboutissant à la rédaction d’un rapport de validation. Ainsi, le suivi en temps réel des opérations de mélange par proche infrarouge a permis de valider les modifications envisagées en apportant une meilleure compréhension, et par conséquent une maîtrise complète des procédés de fabrication considérés.

L'objectif central de ce projet est d'améliorer les délais de production tout en réduisant les coûts, en ayant recours à des méthodes agiles, notamment la standardisation, la simplification, la digitalisation et une démarche d'amélioration continue qui promeut la culture du Lean Management. Ce mémoire a pour finalité de présenter l'une des nombreuses initiatives d'amélioration continue mises en place.
