La recherche des Enterovirus dans le milieu hydrique pose encore de nombreux problèmes techniques notamment en ce qui concerne la concentration des virus dans des échantillons de nature différente. S'il est admis que dans les eaux usées urbaines, la concentration en virus peut être suffisamment importante pour permettre leur détection directe, en revanche, dans tous les autres types d'eaux, elle est la plupart du temps faible et il devient très difficile de réaliser une détection directe. Il est alors indispensable de procéder systématiquement à une concentration sous un faible volume des particules virales présentes dans un grand volume d'eau à analyser. En tenant compte de ces données, il est important de disposer de méthodologies appropriées permettant la détection des virus entériques dans le milieu hydrique. Notre travail comporte deux objectifs essentiels. Le premier objectif de notre travail est essentiellement technique. Il est fondé sur le fait que les virus entériques sont présents, en général, en faible quantité dans le milieu hydrique. Il est donc nécessaire, la plupart du temps, de les concentrer sous un faible volume avant de pouvoir ensuite les détecter. Dans ces conditions, la valeur d'une analyse virologique d'une eau dépend d'abord de la qualité virologique des techniques de concentration utilisées. Notre objectif est d'évaluer des techniques de concentration primaire et secondaire des Enterovirus. Trois méthodologies de concentration primaire fondées sur l'adsorption-élution sur verre (poudre de verre et laine de verre) et deux techniques de concentration secondaire fondées sur la précipitation (précipitation organique et précipitation au polyéthylène glycol 6000) seront testées comparativement sur différents types d'eau artificiellement ou naturellement contaminés, de façon à tenter de déterminer les protocoles présentant le meilleur rendement et la plus grande efficacité pour l'isolement des virus infectieux et de leur génome. Le deuxième objectif est de déterminer le niveau de contamination microbienne et en particulier virologique du milieu hydrique au Maroc. Pour ce faire, nous rechercherons des Enterovirus et des bactéries pathogènes (Salmonelles, vibrions, ... ) dans différents types d'eaux (eaux usées , eaux d'estuaire, eaux de mer, eaux de puits). Mais, nous isolerons et quantifierons aussi les indicateurs bactériens et viraux (coliphages somatiques). Celte recherche simultanée des pathogènes et des indicateurs devrait permettre de tenter d'établir des corrélations et d'identifier les indicateurs les plus représentatifs de chaque catégorie de pathogène.
