Divers facteurs sont proposés pour la régulation locale de la sécrétion du lait chez la chèvre. Parmi eux, le Feedback Inhibitor of Lactation (FIL) qui serait une glycoprotéine du lactosérum de 7,6 kDa et un caséinophosphopeptide (CPP), le fragment 1-28 de la caséine β. Le mécanisme d'action du FIL passerait par une déstabilisation de réseau transgolgien des cellules épithéliales mammaires, voire l'inhibition de certains gènes. Le fragment de la caséine serait capable de rompre les jonctions serrées en modifiant la concentration extracellulaire en calcium. La première partie de notre travail a consister à tenter de purifier le FIL à partir de lait de chèvre d'après le brevet déposer par Addey et al. en 1998. La purification de CPP caprins natifs et enrichis en calcium puis la réalisation d'une expérimentation sur des chèvres par injections intramammaires de ces CCP caprins contenant le β 1-28, de CPP du commerce et d'EGTA a constitué notre seconde partie du travail. La mise au point de la mesure de l'activité de la plasmine, une protéinase du lactosérum, a clôturé notre travail. Nous n'avons pas retrouvé le pic en FPLC censé contenir le FIL dans la fraction 10-30 kDa du lactosérum, et une analyse par LC/MS n'a pas révélé de pic d'une masse de 7,6 kDa. Un effet sur l'inhibition de la production lait est apparu sur les chèvres traitées avec des CPP natifs de chèvre. Laissant envisager que le fragment β 1-28 pourrait bien avoir un rôle de régulation dans la sécrétion du lait. La mesure de l'activité de la plasmine dans le lait par la méthode de la gamme d'étalonnage semble être prometteuse mais nécessite des essais dans des matrices laitières.
