Book excerpt: The endothelium, a monolayer of endothelial cells, constitutes the inner cellular lining of the blood vessels (arteries, veins and capillaries) and the lymphatic system, and therefore is in direct contact with the blood/lymph and the circulating cells. The endothelium is a major player in the control of blood fluidity, platelet aggregation and vascular tone, a major actor in the regulation of immunology, inflammation and angiogenesis, and an important metabolizing and an endocrine organ. Endothelial cells controls vascular tone, and thereby blood flow, by synthesizing and releasing relaxing and contracting factors such as nitric oxide, metabolites of arachidonic acid via the cyclooxygenases, lipoxygenases and cytochrome P450 pathways, various peptides (endothelin, urotensin, CNP, adrenomedullin, etc.), adenosine, purines, reactive oxygen species and so on. Additionally, endothelial ectoenzymes are required steps in the generation of vasoactive hormones such as angiotensin II. An endothelial dysfunction linked to an imbalance in the synthesis and/or the release of these various endothelial factors may explain the initiation of cardiovascular pathologies (from hypertension to atherosclerosis) or their development and perpetuation. Book excerpt: "Many birth defects arise as a result of improper patterning of neural crest cells (NCCs), the most rostral of which migrate ventrally from the neural folds into the pharyngeal arches, transient structures on the ventral embryo. This thesis examines the instructive signals and transcription factor networks that establish dorsoventral identity of NCCs and thus eventual formation of the craniofacial skeleton. One such signal is endothelin-1 (Edn1), which is produced by the surrounding arch ectoderm. Produced as an immature form, Edn1 undergoes two processing events, the second mediated by endothelin converting enzyme (Ece1). Mature Edn1 then binds to the endothelin-A receptor (Ednra) on NCCs. Loss of Ece1 leads to a lack of mature Edn1 and thus subsequent defects in dorsoventral patterning. However little is known about the factors that regulate Ece1. We show here that Nkx2.5 is required for proper craniofacial development and acts by regulating ece1 expression ..."--Abstract.
