Book excerpt: In a clear exposition this inspirational book shows the in situ three-dimensional morphology of cerebellar neurons, intracortical circuits and synaptic connections that underpin the functioning of cerebellar neurons in networks. It brings together in one volume a new view of the three-dimensional cytoarchitectonic arrangement of the cerebellar cortex. The book shows the cerebellar extrinsic and intrinsic intracortical circuits formed by mossy and climbing fibers as exposed by the cryofracture methods. The high degree of lateral collateralization of these fibers is also displayed providing new insights on the information processing in the cerebellar cortex. Besides, field emission high resolution electron microscopy shows its potential contribution to the study of synaptic morphology. The concluding chapter deals with the contribution of scanning electron microscopy to cerebellar neurobiology. This monograph is an authoritative survey and a must for anyone who is interested in the structure of the central nervous system. It will also appeal to an interdisciplinary audience who wants to learn more about electron microscopy and neurocytology.

Book excerpt: Unbiased Stereology, Second Edition is a practical guide to making unbiased 3-D measurements via the microscope. Only those stereological techniques which have been tried and tested by real application are included. Although this technology is essentially mathematical and statistical, the authors do not immerse the reader in complex analysis, but rather provide simple heuristic explanations and references to the original proof, and illustrate the theory by analogies drawn from everyday experience. To give practical experience in application of the techniques, exercises are provided at the end of each chapter, complete with detailed worked answers.

Book excerpt: Experimental methods for the mapping of nervous pathways are based partlyon the study of retrograde processes in the perikaryon, partlyon the demonstration of degenerative processes along the peripheral part of a transected axon. For this purpose, the Marchi method by which a selective staining of degenerating myelin is obtained has been extensively used. However, when this method is used the non-myelinated terminals of the transected axons are not stained. The introduction, about two decades ago, of silver impregnation as a means of tracing degenerating axons (especially the Glees and Nauta methods) by which also terminal boutons can be demonstrated, led therefore to revolutionary progress in the investigation of interneuronal connections. Notwithstanding, there are weH known difficulties involved in this kind of research. The capriciousness of the silver methods not seldom results in failure of impregnation with loss of valuable experimental animals. But even when well impregnated sections are used, other fundamental difficulties exist. One of the major problems is to prove beyond doubt that the impregnated structures are degenerating boutons and not merely fragments of non-terminal fibres passing the area under examination. Furthermore, only on occasion will silver impregnation permit one to accurately define the specific part of the receiving neuron on which the impregnated fibres end, i. e. , whether the bouton makes contact with soma, dendrite or spine.

Book excerpt: In the vast field of neuroscience, the introduction over the last 30 years of new investigative techniques (such as transmission and scanning electron microscopy, freeze-fracturing technique, cell organelle isolation by differential centrifugation, autoradiography, tracing techniques and immunocytochemistry) has greatly expanded our knowledge of neurocytology. This new information, however, is generally dispersed in the specialist journals or collected in reviews on specific topics. As a result, those whose interests lie in neurocytology have difficulty not only in finding data relative to their particular research, but also and above all, in gaining an overall and systematic vision of their discipline. It was this situation which prompted Ennio Pannese to systematize the major acquisitions on the minute structure of nerve and neuroglial cells and their interrelationships, correlate them with the classical concepts of light microscopy and integrate them, where possible, with elements of biochemistry and cell physiology.

Beginning with background material on the neuron, the book then examines specific portions of the nerve cell, and of the various supporting cells. Structure is first described in a general fashion, followed by detailed coverage of the fine structure of each component, with full discussion of how the structural features relate to their functions. Extensively revised and rewritten, this book will bring readers up to date with the many important developments that have taken place since publication of the previous edition. It includes over 130 electron micrographs and line drawings, many of which are new to this edition.

Available in PDF, EPUB and Kindle. Book excerpt: Our understanding of the molecular mechanisms involved in mammalian brain development remains limited. However, the last few years have wit nessed a quantum leap in our knowledge, due to technological improve ments, particularly in molecular genetics. Despite this progress, the available body of data remains mostly phenomenological and reveals very little about the grammar that organizes the molecular dictionary to articulate a pheno type. Nevertheless, the recent progress in genetics will allow us to contem plate, for the first time, the integration of observation into a coherent view of brain development. Clearly, this may be a major challenge for the next century, and arguably is the most important task of contemporary develop mental biology. The purpose of the present book is to provide an overview that syn thesizes up-to-date information on selected aspects of mouse brain devel opment. Given the format, it was not possible to cover all aspects of brain development, and many important subjects are missing. The selected themes are, to a certain extent, subjective and reflect the interests of the contributing authors. Examples of major themes that are not covered are peripheral nervous system development, including myelination, the development of the hippocampus and several other CNS structures, as well as the developmental function of some important morphoregulatory molecules.

All aspects of the subject are covered, from its most elementary, at the molecular and cellular levels, to its systemic and holistic implications, including its role in cognition and involvement in human diseases and therapeutics. The twenty-two chapters are grouped under four main headings : I. Functional Morphology of the Acetylcholine Innervation in Cerebral Cortex (Including Hippocampus); II. Modes of Action of Acetylcholine in the Cerebral Cortex; III. Cortical Properties and Functions Modulated by Acetylcholine, and IV. Clinical, Pathological and Therapeutic Implications. This broad perspective updates the reader on recent advances and future trends in the study of a major neuromodulatory system in brain.
