Book excerpt: We present a dynamical effective medium theory (EMT) of the dielectric properties of nanoparticle aggregates formed from DNA-linked gold nanoparticles. Experimental measurements show that such aggregates have reduced UV extinction and plasmon bands that are considerably red-shifted and broadened relative to the plasmon absorption feature observed in spectra of dispersed colloid. The EMT, which can be used to reproduce the observed spectral changes, is tested by comparing aggregate spectra calculated using the EMT dielectric function with spectra from explicit coupled particle calculations, and good agreement is found. The EMT dielectric function is used as well in discrete dipole calculations to calculate extinction spectra for a variety of aggregate shapes not amenable to analytic solution, and the sensitivity of the spectra to aggregate shape is examined. We find that the spectra are only weakly sensitive to aggregate shape, and conclude that, when calculating extinction of the DNA-linked aggregates for comparison with experiment, spherical shapes can be assumed.

Book excerpt: Engineered nanomaterials (ENMs) are used in numerous applications due to their unique advantages. Gold nanomaterials (AuNMs) are one such functional ENM, which are utilized as model nanoparticles due to their controlled properties. AuNMs can be synthesized with highly uniform size and shape, where increase in the anisotropy gives rise to novel properties; e.g., unique Plasmon resonance, shape dependent physico-chemical properties, and quantum confinement, etc. These advantages have popularized the use of AuNMs in a wide range of applications in sensing, drug delivery, and photodynamic therapies. Increased use of these ENMs calls for systematic evaluation of the AuNMs to assess the effect of their size and shape on fate and transport in aquatic environment. The natural environment, due to the presence of natural colloids and potential presence of secondary ENMs, presents additional system complexities in studying ENM fate and transport. This dissertation was designed to address two major data gaps: i.e., roles of i) anisotropy and ii) presence of secondary particles on aggregation and deposition of AuNMs. Poly-acrylic acid (PAA) coated uniform-sized gold nanospheres (AuNS) and nanorods (AuNR) are utilized as model nanoparticles. The first part of this research investigated the effect of shape of AuNMs on their aggregation and deposition in singular particulate system under relevant ionic strengths. The second part of this dissertation focused on investigation of the effects of a secondary nanoparticle, i.e., pluronic acid modified single-walled carbon nanotubes (PA-SWNTs), on aggregation and filtration of AuNSs under a wide range of ionic strength. Effects of shape of AuNMs on their aggregation kinetics were investigated by employing time resolved dynamic light scattering (TRDLS) in a wide range of mono- and di-valent electrolyte conditions. Aggregation histories were obtained for the AuNSs and AuNRs in presence of a wide range of NaCl and CaCl2 conditions. The critical coagulation concentrations (CCCs), computed from the stability plots of the AuNMs, were used to compare the aggregation propensity of the PAA coated AuNSs with the AuNRs. The deposition kinetics was monitored using the quartz crystal microbalance with dissipation (QCM-D) technique for a range of NaCl concentrations. Deposition rates determined were used to understand the effects of shape of the AuNMs on their deposition kinetics. Experimental findings suggest that the shape of nanomaterials can influence interfacial properties and result in unique aggregation and deposition behavior under typical aquatic conditions Effects of AuNS size on their aggregation behavior was investigated using citrate stabilized 30 nm and 60 nm sized National Institute of Standards and Technology (NIST) standard particles in biological media. Continuous size evolution of AuNS aggregates was monitored over a 24 h period employing DLS and static light scattering (SLS). Electrokinetic measurements, UV-vis spectroscopy, and electron microscopy were performed for material characterization. The findings from this study indicate that initial differences in AuNS sizes diminish over time as the particles aggregate under the influence of elevated ionic strength in the media. The results from this study will likely influence characterization and experimental design considerations for in vitro nanotoxicity studies. The effects of the presence of a secondary particle, PA-SWNTs, on hetero-aggregation behavior of PAA coated AuNS were systematically studied over a wide range of mono- and divalent electrolyte conditions. PA modification of SWNTs made them stable in the entire range of electrolyte concentration where SWNT-SWNT aggregation has been eliminated via steric interaction. Hetero-aggregation mechanisms of AuNS were deciphered utilizing electron microscopy and electrokinetics. Experimental results suggest that the AuNS aggregates faster in hetero-dispersion in presence of PA-SWNT than in homo-disperstion at elevated ionic strength. However, hetero-rates are slower in case of low ionic strength cases. Techniques developed in this study can be can be adopted elsewhere to assess hetero-aggregation of a wider set of ENMs, hence achieving reliability in nanoparticle environmental exposure and risk. Co-transport of AuNS in presence of PA-SWNTs through saturated porous media was also assessed using bench-scale column experiments under a wide range of aquatic conditions (1-100 mM NaCl). Homogenous AuNS suspensions were utilized as control to compare their breakthrough properties with those of the AuNS hetero-dispersions (in presence of PA-SWNTs). This study also assessed the role of pre-coating of the collectors (with PA-SWNTs) on AuNS' mobility to understand the order of introduction of the secondary particles. The study results demonstrate that the presence of secondary particles and the order in which these are introduced to the experimental system strongly influence AuNS mobility. Thus ENM can be highly mobile or can get strongly filtered out, depending on the secondary PA-SWNT and background environmental conditions. This research not only evaluates the role of material attributes in their fate and transport but also extends to assess the role of environmental complexities on the same. The research findings enhance our current understanding of environmental interaction of the ENMs and call for further studies, incorporating additional complexities (i.e., material and system), to assess the fate, transport, and effects of the ENMs in the natural environment.

These aggregates are an interesting material, posing potential applications in the fields of chemistry, biology and materials science. This study examined the dependence of aggregate size and morphology on temperature of formation, using AFM and TEM imaging. The aggregates formed at 70°C averaged 105nm in width, as compared to 70nm for the room temperature aggregates. The TEM images showed increased density for the 70°C aggregates. In a further study, the room temperature aggregates were functionalized through a place exchange reaction with 1 -mercapto-undecane- l-sodiumsulfonate (MUS), a thiolated ligand with a polar head group. A two-phase test of the water-solubility indicated that the aggregates were fully soluble. TEM images showed a slight increase in size, though similar morphology to the insoluble aggregates. The ability to induce water solubility in the aggregates opens up many potential applications in the field of bionanomaterials.

My two main objectives were to develop (synthesize, characterize) DNA sequences that have a novel quadruplex structural motif that offers non-electrostatic binding site(s) for metal ions, and to study the interaction of metal nanoparticle(s) (MNP) with such DNA quadruplex structures and possibly develop methods for the predictable assembly of MNPs on DNA quadruplex sequences. My focus was the synthesis of gold and silver nanoparticles. My results included characterization of the DNA quadruplexes and how such sequences interact with metal ions. I have used UV-visible spectrophotometry, agarose gel electrophoresis, and circular dichroism to visualize the folding of the sequences into quadruplex structures in the presence of metal ions. I have also used dynamic laser light scattering to investigate the formation of nonspecific DNA aggregates. I synthesized gold nanoparticles in the presence of G-rich DNA sequences and the results obtained showed that the presence of G-rich sequence does modulate the size of the gold nanoparticles formed. Larger gold nanoparticles were observed in the presence of some of the sequences used.

To achieve this goal, we investigated the feasibility of using colloidal gold nanoparticle as an anticancer agent delivery vehicle for treatment of cancer. Then, long-term clearance, toxicity, and biodistribution of colloidal gold nanoparticle were studied to further aid in understanding of using colloidal gold nanoparticle as a drug delivery platform. In particular, two representative sizes of gold nanoparticles, 5nm and 60nm, were investigated for the size effect on the therapeutic efficacy, toxicity, biodistribution, and clearance in cancer nanotherapy. We believe that nanoparticle size plays a critical role in not only delivering the drug to the target site but also determining the in vivo behavior such as biodistribution and clearance. By choosing an appropriate size scale for the system, we successfully used the small-sized gold nanoparticles for drug delivery applications, which also displayed no apparent toxicity with desirable clearance from the biological system. This work is significant by providing an insight on a potential ideal candidate for drug delivery carrier for cancer nanotherapy.

Book excerpt: Spherical nucleic acids (SNAs) comprise a nanoparticle core, and a densely packed and highly oriented nucleic acid shell. They have novel structure-dependent properties that differ from those of linear nucleic acids and that makes them useful in chemistry, biology, the life sciences, medicine, materials science, and engineering. This book is a reprint volume that compiles 101 key papers that have been published by the Mirkin Group at Northwestern University, USA, and their collaborators over the past more than two decades. Volume 1 provides an overview and a historical framework of SNAs and discusses their enabling features, which set them apart from all other forms of matter. Volume 2 covers the general design rules for colloidal crystal engineering with DNA, spanning the building blocks and DNA- and RNA-based "programmable bonds" that can be utilized in preparing such structures. Volume 3 continues the discussion of colloidal crystallization processes and routes to hierarchical assembly, featuring dynamic nanoparticle superlattices and lattices prepared on surfaces or via templating strategies, and explores what one can uniquely learn from and do with colloidal crystals prepared from nucleic acid–functionalized nanomaterials in optics, plasmonics, and catalysis. Volume 4 covers the role of SNAs in biomedicine, especially as diagnostic probes both inside and outside of cells, and treatments based on gene regulation and immunotherapy.
