Book excerpt: Besides maintaining high cell viability and achieving dose control, an important feature of NEP is the delivery of large molecules such as plasmids and quantum dots directly into the cell cytosol. In contrast, delivery of large plasmids during bulk electroporation, wherein cells and genes/plasmids are mixed in a buffered medium and an external electric field is applied across the mixture which electroporates the cells, is via formation of cell membrane bound aggregates which get endocytosed post pulsation. Various mechanisms of DNA transport across the membrane have been proposed including passive diffusion of DNA through pores formed during electroporation, electrophoretic transfer of DNA into the cytosol through membrane pores, and electrically induced trapping of DNA in the endosome-like vesicles. In this study we visualized the transport of DNA molecules during NEP to understand the associated delivery mechanism.

In this dissertation, we study the DNA stretching dynamics in DNA combing, DNA electrophoretic dynamics in the microchannel and nanochannel, cell electroporation mechanism in NEP, and the detection of membrane poration.

Book excerpt: Abstract: Electroporation is a common technology for delivering genes and small or large molecules into cells. Local electroporation using a thin membrane with random micro- pores has shown significant enhancements over bulk electroporation on transfection efficiency and cell viability. In this study, we demonstrate a much less invasive and more efficient gene delivery method - membrane sandwich electroporation (MSE). We use a poly (e-caprolacton) (PCL) membrane with well defined micro-pore arrays for cell-trapping and gene delivery. Cells are immobilized on the micro-pore arrays by vacuum for localized electroporation. The localized electroporation is realized by applying voltage pulse or DC signal to achieve dielectric breakdown of the cell membrane. The conically circular pore arrays provide a great potential gradient when an electrical field is applied. This provides an efficient way to accelerate the rigid carriers to a high momentum for delivering species inside cells without damage to cell membrane. For device fabrication, the PCL membrane films are replicated from patterned SU-8 structures. The hollow microarrays in SU-8 with 10l̃5 um in height are transferred to a PDMS (Polydimethylsiloxane) film by soft lithography. The PDMS film with transferred circular pillar patterns is used as the mold for another replication of uniform PCL thin polymer films with a thickness of 5 to 8 um. NIH 3T3 cells are trapped and immobilized on micro-pore arrays by vacuum. Opti-MEM ITM reduced-serum medium is filled into the channels and the center reservoir before the DNA (pEGFP) sample is loaded into the inlet reservoir. Electroporation is carried out followed by a cell culture process for transfection efficiency characterization. The fluorescence images of NIH 3T3 cells after DNA delivery by electroporation in a MSE device with 5 [mu]m pores array indicate a uniform gene delivery. Evidently, the cells are uniformly trapped on the micro-pores array and each cell experiences the same condition during electroporation. Impedance measurements and device modeling are performed to study the behaviors at different biases and frequencies. DNA transfection with fluorescence expression by using probe station electroporation is able to be observed from 1 Volt to 40 Volt. Such devices for local electroporation with well defined micro-pore array structures have demonstrated great improvement in comparison with devices with random pores.
