Book excerpt: olymerases are the nucleotidyl transferases that are responsible for synthesizing DNA and RNA. They are crucial for essential cellular processes including cellular and viral genome replication, DNA repair and damage tolerance, and transcription. Consistent with their vital roles, polymerases are found in all domains of life. The overall chemistry employed by these enzymes is conserved but there are variations among the different groups of polymerases that confer different substrate specificities and nucleotide incorporation fidelities that allow them to be involved in a wide array of cellular activities. Since polymerases were first isolated more than six decades ago, we have made great progress in understanding how different polymerases have adapted to their specific roles. In this Research Topic we will focus on the enzymatic mechanisms of these enzymes and the relationships between polymerase structure and mechanism, to highlight common themes and unique adaptations.

Available in PDF, EPUB and Kindle. Book excerpt: In biological systems, the normal processes of oxidation (plus a minor contribution from ionising radiation) produce highly reactive free radicals. These can readily react with and damage other molecules. In some cases the body uses free radicals to destroy foreign or unwanted objects, such as in an infection. However, in the wrong place, the body's own cells may become damaged. Should the damage occur to DNA, the result could be cancer. Antioxidants decrease the damage done to cells by reducing oxidants before they can damage the cell.Virtually all studies of mammals have concluded that a restricted calorie diet extends the lifespan of mammals by as much as 100 percent. This remarkable finding suggests that food is actually more damaging than smoking. As food produces free radicals (oxidants) when metabolised, antioxidant-rich diets are thought to stave off the effects of ageing significantly better than diets lacking in antioxidants. The reduced levels of free radicals, resulting from a reduction in their production by metabolism, is thought to be a major cause of the success of caloric restriction in increasing life span. Antioxidants consist of a group of vitamins including vit

Book excerpt: Considering the current interest in cellular regulation and intracellular signalling systems, it is surprising that the contribution of ADP-ribosylation reactions to the modulation of a variety of specific cell processes, in parallel with other post-translational modifications such as phosphorylation, has not been generally recognized. While it is not feasible to cover all aspects of ADP-ribosylation, the thirty-one articles contained in this volume provide a valuable overview of recent progress in the field within the context of cell control mechanisms. For the convenience of the reader, the various topics have been grouped into several sections: (a) poly(ADP-ribosyl)ation; (b) mono-ADP-ribosylation; (c) toxin mono-ADP-ribosylation; (d) inhibitors and activators; (e) protein modification with ADP-ribose and its analogues; and (f) non-modification forms of ADP-ribose. The contents of the individual chapters reflect the ideas of the contributors, many of whom have spent their careers attempting to resolve the biological functions of ADP-ribosylation. We hope that this publication will serve as a useful reference for those investigators that are new to the area as well as those who are actively studying ADP-ribosylation.

Available in PDF, EPUB and Kindle. Book excerpt: Cancer therapeutics include an ever-increasing array of tools at the disposal of clinicians in their treatment of this disease. However, cancer is a tough opponent in this battle, and current treatments, which typically include radiotherapy, chemotherapy and surgery, are not often enough to rid the patient of his or her cancer. Cancer cells can become resistant to the treatments directed at them, and overcoming this drug resistance is an important research focus. Additionally, increasing discussion and research is centering on targeted and individualized therapy. While a number of approaches have undergone intensive and close scrutiny as potential approaches to treat and kill cancer (signaling pathways, multidrug resistance, cell cycle checkpoints, anti-angiogenesis, etc.), other approaches have focused on blocking the ability of a cancer cell to recognize and repair the damaged DNA that primarily results from the front-line cancer treatments; chemotherapy and radiation. This comprehensive and timely reference focuses on the translational and clinical use of DNA repair as a target area for the development of diagnostic biomarkers and the enhancement of cancer treatment. - Saves academic, medical, and pharmaceutical researchers time in quickly accessing the very latest details on DNA repair and cancer therapy, as opposed to searching through thousands of journal articles - Provides a common language for cancer researchers, oncologists, and radiation oncologists to discuss their understanding of new molecular pathways, clinical targets, and anti-cancer drug development - Provides content for researchers and research clinicians to understand the importance of the breakthroughs that are contributing to advances in disease-specific research

This has led to the development of new anticancer agents that have the ability to block the activity of proteins involved in neoplastic cell development and proliferation. Targeted Therapies in Oncology, Second Edition provides a concise timely panorama of existing targeted therapies and progress into future anticancer treatments. These therapies notably include: Targeted agents of immune checkpoints Signal-transduction inhibitors Antiangiogenic agents Vascular-disrupting agents Apoptosis modulators Stem cell inhibitors Tumor profiling for drug development The book emphasizes the biology behind this new class of drugs as well as the clinical achievements obtained. The contributors to this volume stand at the cutting edge of cancer research and treatment around the world.
